Уникальные каталитические свойства ферментов обеспечили им применение в самых разных областях биотехнологии: от научных исследований до промышленных процессов. К настоящему времени охарактеризовано большое число ферментов, для многих из которых получены трехмерные структуры с высоким разрешением, достаточно подробно описаны их биохимические и каталитические свойства и предложен вероятный механизм проявления биологической активности. Кроме того, на основе полученных данных, используя метод направленного мутагенеза, создаются ферменты с повышенной стабильностью и каталитической эффективностью. Все вышеперечисленное расширяет сферу практического использования ферментативного катализа.

Семейство лакказ, первый представитель которых впервые был обнаружен более столетия назад в соке японского лакового дерева Rhus vernicifera, попрежнему остается предметом интенсивных фундаментальных исследований.

Ферменты этого семейства найдены не только в растениях, но также в грибах, бактериях и насекомых. Лакказа [КФ 1.10.3.2] обладает способностью непосредственно катализировать окисление различных соединений молекулярным кислородом, включая о- и w-дифенолы, аминофенолы, полифенолы, полиамины, арилдиамины, фенольные подструктуры лигнина, а также некоторые неорганические ионы. Кроме того, фермент способен осуществлять прямой биоэлектрокатализ реакции восстановления молекулярного кислорода по механизму прямого безмедиаторного переноса электрона с электрода на активный центр лакказы с последующим восстановлением кислорода до воды.

Физиологические функции лакказ разнообразны: участие в процессах формирования пигментов и образования плодовых тел грибов, биодеградация лигнина и детоксификация фенолов. В связи с вышеперечисленными особенностями данного семейства ферментов они интенсивно используются в различных отраслях биотехнологии.

Большое число работ посвящено поиску и изучению новых штаммов базидиальных грибов - продуцентов лигнинолитических ферментов, и нахождению ферментов с новыми физико-химическими свойствами, что связано с возможными широкими прикладными аспектами их использования. Примерами применения лакказ являются получение биосвязующих для экологических древесно-стружечных плит [Болобова с соавт., 2002], создание биотопливных элементов [Heller et al., 2003; Heller, 2006; Coman et al., 2008], биодеградация ксенобиотиков [Johannes et al., 1996; Shintaro et al., 2001], производство антимикробных [Johansen et al., 2003], синтетических моющих [Boutique et al., 2004; Gardner et al., 2006] и косметических средств [Onuki et al., 2000; Aaslyng et al., 2002; Lang & Cotteret, 2006], использование ферментов в органическом синтезе [Echido et al., 2001; Karamyshev et al., 2003], аналитических системах [Kruus, 2000; Duran et al., 2002; Ярополов с соавт., 2005; Shleev et al., 2006; ], а также текстильной [Barfoed et al., 2004; Damkhus et al., 2002], пищевой [Huang et al., 2001; Si, 2001] и целлюлозно-бумажной промышленностях [Cheng et al., 2003; Pedersen et al., 2001; Lund & Felby, 2003; Jetten et al., 2004].

Интерес к изучению этих ферментов обусловлен, прежде всего, их каталитическими характеристиками и субстратной специфичностью. В последнее время все большие масштабы приобретает использование биокаталитических технологий в промышленности, что, возможно, связано с экологической безопасностью производства на их основе [Call and Мьске, 1997; Kruus, 2000;

Mayer and Staples, 2002; Couto and Herrera, 2006]. Промышленное получение биокатализаторов с использованием технических ферментных препаратов является экономически выгодным. Об этом свидетельствует возрастающий объем продаж технических препаратов ферментов на мировом рынке, который в 2007 году составил около 2.3 миллиарда долларов.

Помимо использования лакказ в качестве биокатализаторов различных биотехнологических процессов, они представляют большой интерес с точки зрения фундаментальных исследований их структуры и механизма катализа. Интерес связан с особенностями строения активного центра лакказ, в который входят четыре иона меди трех различных типов, координированное взаимодействие которых позволяет осуществлять восстановление молекулярного кислорода непосредственно до воды, минуя стадию образования пероксида водорода. Реакции окисления органических соединений с участием лакказ протекают по свободно-радикальному механизму, который в настоящее время изучен слабо. Кроме того, лакказа является одним из важнейших компонентов лигнолитического комплекса дереворазрушающих грибов белой гнили, осуществляющих разложение (а в некоторых случаях и синтез) одного из наиболее распространенных природных полимеров - лигнина, и роль лакказы в этих процессах до конца неизвестна.

В связи с вышесказанным, актуальным является проведение сравнительного исследования лакказ из различных источников с целью выяснения сходства и различий в механизмах их действия, что, в свою очередь, является основой для их дальнейшего практического использования в таких областях, как ферментативный синтез, биоэлектрокатализ и биодеградация ксенобиотиков. В зависимости от конкретного случая практического использования, к ферментам предъявляются весьма различные требования. Поэтому в настоящей работе было проведено сравнение физико-химических свойств лакказ из различных источников с целью определения и прогнозирования возможности их использования в тех или иных областях.

1. Грибная и древесная лакказы катализируют реакцию электровосстановления молекулярного кислорода на элеюродах из углеродных материалов по механизму прямого переноса электрона. Древесная лакказа способна катализировать эту реакцию при нейтральных значениях рН раствора со слабой степенью ингибирования ионами хлора. Грибная лакказа, напротив, обладает высокой биоэлектрокаталитической активностью только в кислой среде, при этом ферментативная активность значительно ингибируется ионами хлора.

2. Установлено, что бактериальная лакказо-подобная медьсодержащая оксидаза Bacillus halodurans является низко редокс-потенциальным ферментом, проявляющим электрокаталитическую активность по механизму прямого электронного переноса при нейтральных и слабощелочных значениях рН раствора в присутствии ионов хлора и двухвалентной меди.

3. Показано, что эффективность катализа (^ка-У-^м) высоко- и низкопотенциальных лакказ по отношению к субстратам-доноров электронов близкого строения линейно зависит от «движущей силы реакции», т.е. от разности редокс-потенциалов Т1 центра фермента и субстрата.

4. Экспериментально доказано, что хелатированные ионы двухвалентного марганца являются природными субстратами высоко редокс-потенциальных грибных лакказ и, образующиеся в результате ферментативной реакции хелатированные ионы трехвалентного марганца способны к неферментативному окислению модельного соединения лигнина - вератрового спирта, до вератровой кислоты. Низко редокс-потенциальная древесная лакказа не катализирует эту реакцию.

5. Показана способность высоко редокс-потенциальных грибных лакказ катализировать свободно-радикальную реакцию полимеризации анилина, а также отсутствие такой возможности при использовании низко редокс-потенциальных ферментов.
