Исследование ангиотензин-превращающего фермента (АПФ, пептидил-дипептидаза А, КФ 3.4.15.1) важно не только с практической, но и с фундаментальной точек зрения. Участие фермента в регуляции кровяного давления, водно-солевого обмена, различных сердечнососудистых патологиях определяет интерес к этому ферменту со стороны медиков [1, 2], а необычное, резко индивидуальное сочетание свойств, таких как ион металла в активном центре, высокое содержание углеводов, двудоменная структура с двумя активными центрами, активация анионами и ряд других свойств [3, 4], делает АПФ сложным, однако очень интересным и перспективным с точки зрения общей энзимологии объектом исследования для биохимиков.

Одной из наиболее важных особенностей ангиотензин-превращающего фермента является наличие в его составе двух каталитически активных центров [5], которые, несмотря на высокую степень гомологии, различаются по субстратной специфичности, сродству к ингибиторам, способности активироваться анионами [3]. В литературе имеется обширный материал, посвященный исследованию каталитических свойств соматического двудоменного фермента [6], однако вопрос о взаимодействии активных центров, расположенных на разных доменах (Ы-и С-) полноразмерного АПФ, остается практически неизученным.

Целью данной работы явилось сравнительное исследование каталитических характеристик двудоменного ангиотензин-превращающего фермента и его однодоменных форм, и, на основании полученных данных, определение механизма взаимодействия активных центров в составе полноразмерного фермента. В качестве объекта исследования был выбран фермент быка.

выводы

1. Разработан метод получения каталитически активного фрагмента ангиотензин-превращающего фермента, соответствующего М-домену белка, путем ограниченного протеолиза полноразмерной соматической формы трипсином.

2. Определена ингибиторная и субстратная специфичность различных форм АПФ быка - двудоменной соматической формы, однодоменной (С-домен) тестикулярной формы и отдельного >1-домена фермента. Выявлены различия в каталитических характеристиках активных центров, расположенных на разных доменах соматической формы АПФ быка. Продемонстрирована меньшая требовательность активного центра, расположенного на С-домене фермента, к структуре гидролизуемого субстрата.

3. Показано, что наблюдаемый формально-кинетический механизм функционирования активных центров в составе полноразмерной двудоменной формы фермента зависит от длины гидролизуемого субстрата и предельно упрощается до независимого в случае дека- и нонапептидов и взаимозависимого в случае трипептидных субстратов механизмов. На примере гидролиза пентапептидов Ь ей5 -энке ф ал и н а и Ме^-энкефалина показано, что не только длина, но и структура гидролизуемого субстрата оказывает влияние на механизм совместного функционирования активных центров АПФ.

4. Продемонстрирована отрицательная кооперативность при связывании второй молекулы «короткого» ингибитора лизиноприла с молекулой полноразмерного фермента. Показано, что природа субстрата играет принципиальную роль в эффективности ингибирования активности фермента. При связывании ингибитора с одним из активных центров двудоменного фермента второй центр теряет способность к связыванию и гидролизу «короткого» тринептидного субстрата, но сохраняет полную каталитическую активность по отношению к «длинному» нонапептиду.
