Актуальность проблемы. Развитие животноводства во всем мире связано с внедрением интенсивных технологий выращивания сельскохозяйственных животных, позволяющих получать целевой продукт (мясо, молоко, яйцо, шерсть и др.) зачастую в ущерб основным физиологическим функциям организма. Основными методами достижения наибольшей продуктивности до недавнего времени было использование стимуляторов роста, кормовых антибиотиков, гормонов, введение в рацион только rex кормов, которые способствовали бы наибольшему выходу требуемой продукции без учета их влияния на популяцию кишечных бактерий.

Постепенно становилось очевидным, что такая тактика животноводства может приводить к увеличению стрессовых нагрузок на организм животных и стабильной циркуляции в популяциях сельскохозяйственных животных и птицы инфекционных кишечных заболеваний. Применение антибиотиков для борьбы с кишечными инфекциями не только не сократило развития инфекций, но и напротив, обнаружено даже увеличение падежа (94). Кроме того, широкое использование антибиотиков в животноводстве привело к переносу антибиотикорезистентности от штаммов микроорганизмов животного происхождения к микробным штаммам человеческой популяции. Обнаружено также, что антибиотики, применяемые в животноводстве, накапливаются в мясе и молоке и могут отрицательно действовать на организм человека, особенно детей. Таким образом, для получения максимального выхода чистой продукции возникла необходимость изыскания альтернативных путей интенсификации животноводства с учетом экологических принципов влияния на рост и развитие животных. Такая постановка задачи связана прежде всего с тем, что сосредоточение большого числа животных на ограниченных площадях и изоляция их от окружающей среды приводит к формированию замкнутых популяционных отношений - животное - внешний микробиоценоз, животное - внутренний микробиоценоз организма. На внутренний микробиоценоз организма при интенсивных методах животноводства оказывается постоянное однонаправленное воздействие, что приводит к ослаблению здоровья и конституции животных, глубоким нарушениям обмена веществ, снижению продуктивности, появлению новых заболеваний, «стадных» патологий или болезней «индустриализации животноводства» (93). Связано это с тем, что не учитывался основной экологический закон сохранения равновесия кишечной микрофлоры и ее влияние на общий и местный иммунный гомеостаз макроорганизма.

Эта задача получила возможность разрешения после важнейших открытий последних лет - цитокнн-обусловлениой сигнальной системы, обеспечивающей взаимообмен информацией между клетками макроорганизма, что позволило расшифровать и объяснить многие феномены, Показано, что существуют различные группы цитокипов, ответственных как за провоспалительный, так и за противовоспалительный процесс. Так, такие цитокимы, как фактор некроза опухолей, интерлейкин I, интерлейкин б (TNF, IL-1, IL-6) играют ведущую роль в индукций ответа острой фазы, что является интегральной частью механизма защиты внутренней среды организма против чужеродных субстанций и инфицирующих микроорганизмов, Ряд других цитокинов - интерлейкин 2 и интерлейкин 10 (IL-2, IL-10) несут противовоспалительные функции и вовлечены в процесс блокирования воспалительного ответа на резидентную микрофлору (164). Выработка цитокинов в организме животного происходит в ответ на воздействие различных факторов окружающей среды, причем основную роль играют компоненты клеточной стенки микроорганизмов. В связи с этим предложен термин «бактериальные модулины», означающий класс бактериальных структур, которые обладают цитокин-индуцирующей активностью (309)

Макроорганизм существует в состоянии симбиоза с микроорганизмами, населяющими его кишечник, кожу, слизистые оболочки зева, дыхательных путей, мочеполового тракта и др. На поверхности соприкосновения (или разделения) внешнего мира с внутренней средой организма концентрация микроорганизмов наиболее высока и достигает на коже 105'6 КОЕ/г, а на слизистых кишечника свыше 101Ы2 КОЕ/г (369). Площадь соприкосновения стерильной внутренней среды макроорганизма с внешним микромиром огромна, например, для тонкого отдела кишечника при длине последнего в среднем 2,8-3,2 м площадь слизистых оболочек составляет 180-200 м2(371). Вероятно, важность происходящих на разделе стерильной и не стерильной фаз организма событий, отражается и в филогенетическом развитии. Так, иммунная система слизистых развивается из энтодермального зародышевого листка, остальная часть иммунной системы - мезодермального происхождения. Общая иммунная система локализована в стерильной зоне организма и имеет основной функцией формирование иммунной памяти, местная обслуживает органы дыхания, мочеполовую систему, молочные железы, кишечник и расположена на разделе двух зон - стерильной и не стерильной. Она ответственна за фагоцитоз и обучение остальной части иммунной системы, определяет уровень реактивности локальной и общей иммунной системы, формирует общий уровень гомеостаза (457).

Фагоцитоз и естественный лизис микробных клеток является основным источником микробных фрагментов - продуктов распада микробного клеточного материала - липидов, аминокислот, пептидов, углеводов, а также мелких молекул - мономеров разрушаемых белков, углеводов и липидов. Теоретически легко предсказать наличие бактериальных молекул-маркеров в крови животных и в качестве цитокин-индуцирующих факторов вирулентности, и в качестве молекул, уравновешивающих провоспалительную и противовоспалительную активность клеток, то есть обеспечивающих гомеостаз. Одна часть этих молекул способна активировать функции клеток хозяина (прежде всего, моноциты/макрофаги), индуцируя выброс эйкозаноидов (про-стагландинов, лейкотриенов, тромбоксанов), интерлейкинов и других цитокинов, другая часть обладает способностью ингибировать активность клеток. При этом допускается, что какая-то часть молекул может быть индифферентна (306). В подтверждение этого показано (44), что в норме маркеры превалирующих в кишечнике животных бактероидов, эубактерий и бифидобактерий либо отсутствуют, либо выявляются в крови лишь в небольших количествах, что косвенно свидетельствует в пользу существующей точки зрения об относительной иммунологической толерантности к индигенным анаэробам, и, следовательно, низкой вероятности фагоцитоза этих микроорганизмов.

Обучение организма происходит в новорожденном возрасте, когда кишечник не имеет ферментативной системы, что способствует нормофлоре формировать локальную и сетевую иммунную системы при помощи тех структур бактериальных клеток (мурамилдипептиды), которые формируют равновесную систему про- и противовоспалительных цитокинов. Наиболее ответственным моментом является гомеостатический баланс, так как микроорганизмы, продукты распада которых формируют только противовоспалительный профиль цитокинов, могут привести организм к состоянию имму-нодепрессии, а преобладание у микроорганизмов провоспалительных модулинов может вызвать предрасположенность к возникновению неадекватно острых иммунных реакций. Требует тщательного изучения состав компонентов клеточной стенки бактерий, относящихся к резидентной (пробиотической) микрофлоре на предмет наличия веществ, стимулирующих развитие толерантности к самому этом микроорганизму, а так же на наличие компонентов, являющихся адъювантами по отношению к патогенным и условно-патогенным микроорганизмам.

Изучение этого вопроса на данный момент крайне затруднено в силу того, что кишечник всех животных является пристанищем огромной микробной популяции, которая состоит из представителей большого количества видов и адаптирована к росту в симбиозе с хозяином определенного вида. Каждый отдел кишечника заселен типичной для него микрофлорой, состав которой зависит от окружающих условий, возраста, питания или иммунного статуса хозяина. Это так называемая пищеварительная экосистема, основным критерием существования которой является следующее: в кишечнике присутствует огромное число микроорганизмов, примерно 1014 бактерий; здесь присутствует большое разнообразие видов бактерий, эти виды полностью приспособлены к виду хозяина, к обитанию в кишечнике, составу пищи и возрасту хозяина; все эти микроорганизмы находятся в хорошо сбалансированном состоянии, если животное находится в оптимальных условиях содержания. Благодаря этому балансу вредные бактерии не могут преобладать в росте; существует большое количество факторов, нарушающих этот исключительный баланс. Это отъем молодняка от матери, смена кормового рациона или места содержания, антибиотикотерапия, плохие санитарные условия и др. Действие этих факторов характеризуется ростом популяции вредных бактерий в ущерб росту представителей полезной микрофлоры с последующим развитием заболеваний желудочно-кишечного тракта, нарушений пищеварения, диареей, ослабление роста и утилизации кормов.

Для восстановления нормальной экосистемы желудочно-кишечного тракта было предложено использовать микробную популяцию представителей нормофлоры для системного вытеснения патогенных микроорганизмов и возврата к естественному балансу между нормальной и вредной микрофлорой. Для обозначения этой субстанции Ричардом Паркером в 1977 году был введен термин «пробиотик» в противовес термину «антибиотик», который подразумевает нарушение одного или более элементов в кишечной экосистеме, что может привести к утрате контроля над заболеваниями. Таким образом, в основу концепции пробиотиков положен принцип восстановления нормальной микрофлоры после различных неблагоприятных воздействий на пищеварительную экосистему здорового животного (61).

В настоящее время пробиотики рассматривают как препараты, содержащие, как правило, живые бактерии-представители эндогенной кишечной микрофлоры животных. Штаммы микроорганизмов для приготовления пробиотиков отбираются по определенным критериям, основанным на научных знаниях физиологии и микробиологии кишечника, а также питания животных. Применение пробиотических препаратов позволяет балансировать кишечную экосистему хозяина путем поддержания роста микрофлоры, полезной для кишечника, что предотвращает экспансию вредных, патогенных микроорганизмов.

В естественных условиях животные рождаются практически стерильными, но быстро получают полезные бактерии от матери и из окружающей среды. Приобретение полезной микрофлоры коррелирует с приобретением естественной резистентности к кишечным заболеваниям, вызываемым Е. coli и другими микроорганизмами. Наглядным примером важности кишечной микрофлоры в формировании резистентности к инфекционным заболеваниям является то, что для гнотобионтных мышей смертельной дозой Salmonella enteritidis являются 10 клеток на мышь, в то время как для нормальных мышей смертельная доза равна 1000 миллионам клеток (60).

В настоящее время работами многих ученых показано, что общим и доминирующим компонентом микрофлоры верхних отделов желудочно-кишечного тракта различных взрослых здоровых животных являются лактобациллы, бифидобактерии и молочнокислые стрептококки (54). Механизмы их действия в качестве антагонистов вредной микрофлоре известны только в первом приближении и могут включать в себя следующее: выработку антибактериальных субстанций (органические кислоты, специфические бактериоцины), конкуренцию за питательные субстраты и исключение других бактерий с поверхности кишечного эпителия путем адгезии к слизистым и конкуренции с патогенными бактериями за сайты колонизации, показано их влияние на иммунную систему. Однако их влияние на цитокиновый профиль на уровне местной и общей иммунной системы организма в достаточной степени не изучено (52). Кроме того, применяемые пробиотические микроорганизмы нельзя рассматривать отдельно от уже сложившейся микрофлоры кишечника животного и его влияния на иммунную систему. Трудно предположить, что введение одних пробиотических штаммов при клинической картине кишечного заболевания может изменить соотношение про- и противовоспалительных цитокинов в благоприятную сторону.

Мы предположили, что совместное применение пробиотических микроорганизмов и цитокинов, обладающих противовоспалительными свойствами, сможет быть более успешным для борьбы с кишечными инфекциями с учетом функционального гомеопатического равновесия пищеварительной и иммунной систем организма.

Цель и задачи исследований. Целью наших исследований явилась разработка нового подхода к решению проблемы регуляции микробиоценоза кишечника и изучение нового класса препаратов - иммунопробиотиков. Основные задачи исследований:

1. Подобрать и изучить цитокины, обладающие противовоспалительными свойствами, для создания препаратов нового класса.

2. Разработать средства иммунокоррекции на основе цитокинов и изучить их влияние на основные звенья иммунной системы организма животных.

3. Разработать пробиотический компонент препарата нового класса и изучить его влияние на основные звенья иммунной системы.

4. Создать на основе данных о влиянии цитокинов и пробиотиков на основные звенья иммунной системы препараты нового класса - иммунопробиотики.

5. Изучить влияние иммунопробиотических препаратов на основные звенья иммунной системы.

Научная новизна работы

- сконструирован методом генной инженерии гибридный полипептид, состоящий из фактора некроза опухоли альфа и тимозина альфа 1 (Т-ФНО-Т), обладающий противовоспалительными свойствами, и изучено его влияние на основные звенья иммунитета;

- разработан иммуномодулирующий препарат неотим с противовоспалительными свойствами на основе гибридного белка Т-ФНО-Т;

- разработан новый пробиотический препарат лактобифид с использованием штаммов молочнокислых микроорганизмов и бифидобактерий, изучено их действие на иммунную систему хозяина, определена область применения;

- изучено влияние совместного введения противовоспалительных цитокинов и пробиотических микроорганизмов на систему общего и местного иммунитета;

- разработаны новые иммунопробиотические препараты лактоферон, бактонеотим и иммунобак.;

- изучено действие иммунопробиотиков на основные звенья иммунной системы и микробиоценоз кишечника;

Научная новизна подтверждена 3 патентами на изобретение: № 2102988; № 2146935; №2129878.

Практическая значимость работы. Разработаны и утверждены установленным порядком нормативные документы, регламентирующие промышленное производство и применение препаратов неотим, лактобифид, лактоферон, иммунобак, бактонеотим: технических условий -5; наставлений и инструкций по применению -5; технологических инструкций по производству и контролю -5. Разработаны Методические рекомендации по проведению лабораторных и производственных испытаний цитокинов животных (проходят процедуру утверждения Департаментом ветеринарии).

Апробация работы. Основные положения диссертации доложены на научных семинарах лаборатории генно-инженерных препаратов ВНИИПМ (1993-1995 г), заседаниях Ученого совета и проблемной научно-методической комиссии по химиотера-певтическим препаратам ВГНКИ встпрепаратов (1995-2001) и НИИЭМ им. Н.Ф. Гамалеи (1999-2001 г)

По теме диссертации опубликовано 38 работ, в том числе монография «Имму-нобиотические препараты в ветеринарной практике» (2000 г), получено 3 патента на изобретение.

Структура и объем диссертации.

Диссертация изложена на 318 страницах машинописного текста, включает введение, обзор литературы, материалы и методы, результаты собственных исследований, обсуждение, выводы, практические предложения, указатель литературы. Диссертация иллюстрирована 109 таблицами и 17 рисунками и диаграммами. Список использованной литературы включает 563 работ отечественных и зарубежных авторов. В приложении представлены копии 30 документов, подтверждающих результаты отдельных этапов работы, их научную новизну и практическую значимость.

ВЫВОДЫ

1. Гибридный белок Т-ФНО-Т обладает противовоспалительными и иммуно-модулирующими свойствами в концентрации 5x10 12 (М). На его основе разработан иммуномодулирующий препарат неотим.

2. При совместной иммунизации мышей вирусом бешенства неотим не стимулирует антителообразование, не увеличивает СПЖ и Кр50. При совместной иммунизации мышей вакциной против гепатита А неотим способствует увеличению среднего геометрического титра специфических антител в 4,5 раза. При совместном применении с вакциной против ньюкаслской болезни проявляет адъювантные свойства. Способствует пятикратному повышению титра антител к вакцине против парвовирусного энтерита собак при однократной иммунизации. Установлена оптимальная доза неотима (10 мкг/10 кг массы) и схема его введения животным (однократная в момент вакцинации, либо дважды с интервалом в 2 суток).

3. Разработан пробиотический препарат лактобифид, содержащий Bifidobacterium globosum БФ-4, Bifidobacterium adolescentis MS42, и Streptococcus faecium, Lactobacillus acidophilus K/AR-08. Лактобифид не оказывает иммуномодули-рующего действия, что подтверждается отсутствием усиления антителообразования в культуре спленоцитов, обработанных эритроцитами барана.

4. Эффективность профилактики колибактериоза поросят препаратом составила 92%. Лечебная эффективность препарата составляет 5%, однако совместном применении с иммуномодулирующим препаратом неоферон эффективность лечения составила 65%.

5. Разработан сухой иммунопробиотический препарат иммунобак для птицеводства, содержащий в своем составе бактерии пробиотики и лейкоцитарный и иммунный интерфероны в физиологических количествах. При применении иммунобака усиливается клеточное и гуморальное звено иммунной системы цыплят (Е-РОК% - 74,1, селезеночный индекс - 2). После обработки цыплят препаратом иммунобак показатели активности лизосомально-катионных белков возрастают до 2,9 ед., что является максимальным значением для взрослой птицы.

6. Применение препарата иммунобак при лечении колибактериоза цыплят способность Е. coli серовара 078 колонизировать слизистую оболочку кишечника птицы достоверно снижается в 3 раза. Профилактическое применение препарата иммунобак

270 способствует увеличению привесов птицы на 10,2% при одинаковом расходе корма в опытной и контрольной группе и увеличению сохранности поголовья до 99,3%.

7. Разработан сухой иммунопробиотический препарат для животных лактоферон, содержащий в своем составе бактерии эубиотики и лейкоцитарный и иммунный интерфероны. Лактоферон усиливает хемотаксическую активность, интенсивность синтеза белка, фагоцитарную, бактерицидную и переваривающую активность макрофагов, антителопродукцию и активность Т-хелперов и киллерных клеток. Установлено, что при применении препарата лактоферон реакция тимусных структур у телят сохранятся на физиологическом уровне, однако значительно усиливается развитие местного ответа лимфоидной ткани кишечника (увеличение уровня секреторного иммуноглобулина А).

В. Эффективность иммунопробиотического препарата лактоферон при профилактике колибактериоза телят составляет 98%, при лечении - 90%.

9. Разработан иммунопробиотический препарат бактонеотим, содержащий в своем составе бактерии пробиотики и гибридный белок Т-ФНО-Т.

10. Лечение кожных заболеваний собак и кошек с применением препарата бактонеотим проводит к нормализации уровня активности уропепсина, плазмопепсиногена, амилазы мочи и энтерокиназы кала, которое носит стойкий необратимый характер.

Рекомендация в производство

Иммуномодуляторы, пробиотики и иммунопробиогики рекомендуется использовать в ветеринарной практике для повышения естественной резистентности организма животных, регуляции микробиоценоза и иммунобиотического статуса кишечника при профилактике и лечении иммунодефицитов и кишечных болезней молодняка сельскохозяйственных животных и мелких домашних животных.
