Известно, что ФДТ с красителями порфиринового и хлоринового ряда инициируют супрессию Т-клсточного звена иммунитета in vivo. Известно таклсе, что супрессорное действие ФДТ с порфиринами, хлоринами и псораленами реализуются через стадию образования фотопродуктов [Kyagova, А.А., et. al., 2005; Kyagova, A.A., et. al., 2002; Potapenko A.Ya., 1998]. Однако в литературе нет данных о том, могут ли фотопродукты МЦ540 влиять на иммунную систему. В этой связи исследование влияния фотопродуктов МЦ540 на Т-клеточный иммунный ответ in vivo является актуальным.Цель и задачи исследования. Целью настоящей работы было исследовать фотолиз и фотовыцветание мероцианина 540 в различных агрегатных состояниях и оценить иммуносупрессорное действие фотопродуктов данного красителя.Для достижения этой цели были поставлены следующие задачи; 1. Разработать методику получения неискаженных спектров резонансного светорассеяния красрггелей.2. Изучить методами резонансного светорассеяния, спектрофотомерии и спектрофлуориметрии влияние растворителя на агрегацию и фотолиз МЦ540.3. Изучить действие предоблученного МЦ540 (пМЦ540) на Т-клеточный иммунный ответ in vivo в модели реакции контактной чувствительности у мышей.

ВЫВОДЫ:

1. Разработана методика исправления измеренных спектров резонансного светорассеяния (PCP) агрегатов красителей: на 1) спектральную чувствительность прибора, 2) эффекты внутреннего светофильтра (экранировка возбуждающего света и реабсорбция рассеянного света в исследуемом образце). Введено понятие резонансного фактора (РФ), который зависит только от размеров и структуры агрегатов и не зависит неспецифического рэлеевского светорассеяния. Предложено строить спектры PCP как зависимость РФ от длины волны.

2. Обнаружено, что добавление солей к водным растворам мероцианина 540 (МЦ540) приводит к возникновению интенсивного резонансного светорассеяния, что доказывает возникновение индуцированных солями крупных надмолекулярных агрегатов красителя. Для солей одновалентных катионов показано существование критической концентрации соли (ККС), только выше которой формируются крупные агрегаты МЦ540 с максимумами поглощения и PCP около 517 нм. Выявлено, что величина ККС обратно пропорциональна концентрации МЦ540. Ниже ККС обнаружено формирование агрегатов МЦ540 с максимумом PCP около 420-460нм, не выявляемых спектрофотометрически.

3. Выявлено, что в водно-солевых растворах, в которых присутствуют несколько агрегатных форм МЦ540, фотолабильность убывает к ряду: крупные надмолекулярные агрегаты с максимумами спектров PCP при 517 и 420 нм » димеры (максимум поглощения около 500 нм) > мономеры (максимум поглощения около 535 нм).

4. Обнаружено, что предоблученный МЦ540 (пМЦ540) способен влиять на Т-клеточный иммунный ответ in vivo в модели реакции контактной чувствительности (КЧ) у мышей к гаптенам 2,4-динитрофторбензолу (ДНФБ) и оксазолону. При этом пМЦ540 вызывал зависимую от дозы предоблучения супрессию реакции КЧ к гаптенам.

5. Показано, что супрессия КЧ, вызванная предоблученный МЦ540, адоптивно переносится другим животным. В основе иммунного механизма супрессорного действия пМЦ540 лежит угнетение функций клеток-эффекторов и активация клеток с неспецифическим супрессорным действием на КЧ
