Актуальность проблемы. Эволюционные преобразования макромолекулярных структур клеток, таких как нуклеиновые кислоты и белки, составляют одну из актуальных проблем современной молекулярной генетики. Последние достижения в области геномики и биоинформатики привели к резкому увеличению числа фундаментальных исследований, направленных на выяснение механизмов молекулярной эволюции и реконструкцию эволюционной истории генов у организмов различного таксономического ранга. Ядерные и митохондриальные ДНК широко используются для выявления родственных взаимосвязей между отдельными особями или группами особей, для изучения динамики генетических процессов в популяциях, описания структуры вида и истории его возникновения [Avise, 2003].

С этой точки зрения особый интерес представляют сложные биологические системы, состоящие из двух или нескольких коэволюционирующих организмов, например паразитов и их хозяев. Среди паразитических организмов одну из наиболее многочисленных групп составляют плоские черви - трематоды, или сосальщики (Platyhelminthes: Trematoda), объединяющие более 18000 видов паразитов беспозвоночных и позвоночных животных. Трематоды отличаются от остальных эукариот наличием сложного жизненного цикла, связанного со сменой животных-хозяев и чередованием нескольких последовательных партеногенетических и одного гермафродитного поколения. Все трематоды - эндопаразиты, обитающие во взрослом состоянии во внутренних органах позвоночных животных и человека, которые являются их дефинитивными (окончательными) хозяевами, а роль первого промежуточного хозяина всегда выполняют различные виды моллюсков [Скрябин, 1948; Гинецинская 1968; Gibson et al., 2002]. Общебиологический интерес представляют проблемы систематики и филогении трематод, процессы взаимодействия паразита с окружающей средой, в качестве которой выступает не только комплекс природных абиотических факторов, но, в первую очередь, сам организм животного-хозяина. Молекулярно-генетические исследования являются весьма перспективными для выяснения механизмов адаптаций, возникающих при разных способах размножения и смены животных-хозяев.

Различные факторы влияют на эволюционную динамику нуклеотидных изменений ядерного и митохондриального генома, и это отражается на уровне отдельных особей, популяций и видов в процессе филогенеза [Ballard, 2000]. К их числу можно отнести 5 различия в скоростях геномной репликации и репарации, смещения в соотношении нуклеотидных замен, различную скорость возникновения внутривидовой гетерогенности за счет неполной сортировки геномов и адаптивную эволюцию.

Помимо фундаментального значения исследование молекулярной филогении трематод может иметь практическое приложение в медицине и сельском хозяйстве при разработке методов генотипирования возбудителей заболеваний человека и животных, а также для экологического мониторинга, создания нового поколения лекарств и усовершенствования вакцин.

Среди трематод одну из древних и высокоспециализированных групп представляют кровяные сосальщики птиц и млекопитающих из семейства Schistosomatidae. Структурная организация и эволюция генома этой группы до сих пор слабо изучена. К 2007 г. получены полные последовательности белок-кодирующих генов мт генома шистосом млекопитающих из рода Schistosoma [Zarowiecki et al., 2007]. Полные последовательности ядерных геномов расшифрованы лишь в 2009 г. для двух представителей этого рода - S. mansoni и S. japonicum, вызывающих тяжелые поражения мочеполовой системы и органов пищеварения человека [Berriman et al., 2009; Liu et al., 2009]. Различные виды и штаммы шистосом подробно изучены с помощью маркеров ядерного и митохондриального генома, что позволило подтвердить выделение внутри рода четырех основных морфо-экологических групп, а также выявить ряд новых закономерностей в распределении и происхождении латино-американских и афро-азиатских видов и популяций [Rollinson, 1996; Lockeretal., 2003].

Менее изучены повсеместно распространенные кровяные сосальщики птиц, а именно сосальщики из рода Trichobilharzia, личинки которых являются возбудителями церкариального дерматита человека (бильгарциоза или "лихорадки купальщика")- Их объединяют в большой род, содержащий около 40 видов, для которых основным дефинитивным хозяином являются птицы из отряда Anseriformes. В Европе известны три вида висцеральных (т. е. паразитирующих в кровеносных сосудах внутренних органов птицы) вида - Т. szidati, Т. franki, Т. salmaticensis, и один назальный вид Т. regenti, паразитирующий в носовой полости и центральной нервной системе птиц. При экспериментальном заражении показан более высокий патогенный эффект для млекопитающих церкарий назальной формы [Horak et al., 1999; 2002]. В настоящее время известен полный мт геном лишь для Т. regenti, имеющий около 60% сходства с азиатским видом S. japonicum в структуре белок-кодирующих генов и их аранжировке [Webster et al., 2007].

Ввиду высокого морфологического сходства шистосом из рода Trichobilharzia только применение молекулярных маркеров позволяет идентифицировать видовую принадлежность церкариальных изолятов. Одними из наиболее эффективных маркеров оказались последовательности двух внутренних транскрибируемых спенсеров рДНК (ITS1 и ITS2) [Dvorak et al., 2002]. С их помощью удалось провести видовую диагностику европейских шистосом и показать различия в структуре консервативных и вариабельных участков рДНК. Изучено распространение трех видов шистосом (T. szidati, T. franki, Т. regenti) в водоемах Чехии, Польши, Франции, Исландии [Rudolfovâ et al., 2005, 2007; Aldhoun et al., 2009; Jouet et al., 2009, 2010]. В некоторых сообщениях использованы последовательности мт гена coxl для демонстрации межвидовых отличий птичьих шистосом с территории Франции и Исландии [Jouet et al., 2009, 2010]. Практически неизученным остается представитель другого широко распространенного в Евразии монотипного рода птичьих шистосом — Bilharziella polonica. Считается, что в отдельных случаях церкарии этого вида, наряду с трихобильгарциями, могут вызывать "лихорадку купальщика" [Szidat, 1938; Horak and Kolarova 2000; Zbikowska, 2003].

Цель и задачи исследования. Цель работы - с помощью молекулярно-генетических маркеров (методов) изучить особенности структурной организации и дивергенции ядерных и митохондриальных генов птичьих шистосом разных видов из родов Trichobilharzia и Bilharziella, обитающих на территории России и Беларуси.

Основные задачи:

1. Оценить особенности структурного полиморфизма последовательностей рДНК (28S, ITS1, ITS2) и генов мтДНК (coxl, сохЗ) птичьих шистосом из двух родов.

2. На основании сравнения с известными последовательностями ядерных и митохондриальных генов провести видовую дифференциацию изученных изолятов шистосом.

3. Разработать молекулярно-генетическую систему видовой идентификации шистосом рода Trichobilharzia.

4. На основании полиморфизма ITS2 рДНК провести видовую молекулярно-генетическую идентификацию промежуточных хозяев-моллюсков из семейства

Ьутпае1с1ае и оценить уровни гостальной специфичности в группе видов птичьих шистосом из рода ТпсИоЬИИата.

5. Реконструировать возможную историю происхождения и филогенетические связи птичьих шистосом, распространенных на территории Евразии.

V. выводы

1. На изученной части ареала наряду с тремя ранее известными в Европе видами птичьих шистосом из рода Trichobilharzia (T. szidati, T.regenti, T. frankï) показано существование нового вида, названного нами Trichobilharzia sp. var. narochanica. Он характеризуется уникальными последовательностями рДНК (28S, ITS1, ITS2) и мт ДНК (coxl, сохЗ) , а также жесткой приуроченностью к определенному виду хозяина-моллюска (Radix ampia).

2. Впервые у птичьих шистосом обнаружена индивидуальная изменчивость рДНК, которая заключается в появлении копий ITS1 рДНК разного размера. Они обнаружены в составе генома у четырех изолятов T. franki из Беларуси и связаны с появлением одного дополнительного или редукцией одного из трех основных повторяющихся звеньев, характерных для большинства изолятов этого вида.

3. Для исследованных популяцияй птичьих шистосом трех видов, Т. szidati, T.regenti, В. polonica, характерно отсутствие четко выраженной внутривидовой структуры по ядерным и митохондриальным генам. У видов Т. franki и Т. sp. var. narochanica выявлены несколько генеалогических линий, конкордантных по мт гену coxl, гену 28S и ITS2 рДНК.

4. Особенности дивергенции митохондриальных и ядерных генов в отдельных популяциях T. franki позволяют рассматривать этот вид как сборный, состоящий из нескольких критических видов с выраженной приуроченностью к разным видам промежуточных хозяев-моллюсков рода Radix.

5. Для видов рода Trichobilharzia обнаружены несоответствия в топологии дендрограмм, построенных на основании вариабельности участков ядерного и митохондриального геномов, вызванные неполной сортировкой предкового полиморфизма и/ или возможной интрогрессией геномов в процессе адаптации к моллюскам из родов Radix и Lymnaea.

6. На основании полиморфизма 28S рДНК разработана молекулярно-генетическая система видовой идентификации четырех видов шистосом рода Trichobilharzia.
