Перемещение мобильных элементов - транспозонов играет важную роль в обеспечении генотипического разнообразия у эукариот. В настоящее время подробно изучены транспозоны, обнаруженные в геноме плодовой мушки Drosophila melanogaster. Более половины спонтанных мутаций, возникающих в геноме дрозофилы, вызваны встраиванием транспозона в тот или иной ген. Наиболее исследованным мобильным элементом у Drosophila melanogaster является Р элемент, который имеет на концах короткие инвертированные повторы. Ранее в нашей лаборатории были открыты супернестабильные мутации, которые возникали с высокой частотой в результате мобилизации активности Р элемента. Было показано, что такие мутации индуцированы химерным элементом, состоящим из уникальных участков генома, окруженных делетированными копиями Р элемента. Мобилизация активности Р элемента приводила к инверсиям последовательности между Р элементами, увеличению или уменьшению фрагментов генома, входящих в состав химерных элементов. При этом внутренние части химерного элемента, содержащие регуляторные последовательности, могли влиять на экспрессию рядом расположенного гена, что приводило к проявлению мутантного фенотипа. Таким образом, химерные элементы способны играть значительную роль в эволюции регуляторных последовательностей, участвуя в создании наиболее эффективно действующей регуляторной системы для конкретного гена. В представленной работе было продолжено изучение механизмов возникновения и распространения химерных элементов, кроме того, химерные элементы были использованы для выявления новых регуляторных последовательностей и изучения взаимодействия между ними.

Также в работе было продолжено исследование наиболее изученного инсулятора дрозофилы, состоящего из 12 сайтов связывания для белка Su(Hw). Известно, что одна 3 копия Su(Hw) инсулятора в составе трансгенной конструкции полностью блокирует взаимодействие между энхансерами тела и крыльев и промотором гена yellow, если находится между ними. Ранее в нашей лаборатории было показано, что две копии Su(Hw) инсулятора, расположенные между энхансером и промотором, не могут блокировать их взаимодействие. В представленной работе с помощью модельной системы гена yellow Drosophila melanogaster было доказано, что Su(Hw) инсуляторы не формируют независимых транскрипционных доменов, а степень инсуляции зависит от расстояния между ними.

Цели и задачи исследования

Основными целями данного исследования являлись:

1) изучение регуляторных элементов, входящих в состав химерных элементов;

2) изучение зависимости взаимодействия между регуляторными элементами от их взаимного расположения и расстояния между ними.

В работе были поставлены следующие задачи:

1. Изучить механизмы возникновения химерных мобильных элементов, состоящих из Р элементов и уникальных фрагментов генома и механизмы их транспозиций.

2. Исследовать влияние перемещения химерных элементов из одного локуса в другой на регуляцию генной экспрессии.

3. Исследовать свойства регуляторных элементов, входящих в состав химерных элементов.

4. Выяснить, как расстояние между двумя Su(Hw) инсуляторами, окружающими энхансеры, влияет на их способность блокировать взаимодействие между энхансерами и изолированным промотором гена yellow Drosophila melanogaster.

1 Обзор литературы

У высших эукариот энхансер может активировать промотор на расстоянии, достигающем нескольких сотен тысяч пар нуклеотидов (West and Fraser 2005; West et al. 2002; de Laat and Grosveld 2003; Bondarenko et al. 2003; Sipos and Gyurkovics, 2005; Dorsett 1999). Недавно для некоторых энхансеров млекопитающих было продемонстрировано, что белки, связанные с энхансером, непосредственно взаимодействуют с белками основного транскрипционного комплекса или вспомогательными белками, собранными на промоторе, при этом ДНК между ними образует петлю (Tolhuis et al., 2002; de Laat and Grosveld 2003). Возникает вопрос, каким образом энхансер может взаимодействовать с промотором на больших дистанциях и при этом правильно узнавать свой промотор. Несмотря на то, что к настоящему моменту описано огромное количество энхансеров и промоторов, изучено множество факторов транскрипции, вопрос о механизме специфичности взаимодействия между энхансером и промотором остается открытым.

В понимании механизмов дальних взаимодействий между регуляторными элементами большую роль может сыграть изучение инсуляторов. Инсуляторами были названы регуляторные элементы, которые блокируют взаимодействие между энхансером и промотором, если находятся между ними (Capelson and Corces 2004; Geyer and Clark 2002; West et al. 2002; Kuhn and Geyer 2003). При этом инсуляторы не влияют непосредственно на активность энхансера и промотора, т.е. промотор может быть активирован любым другим энхансером, а энхансер может активировать любой другой промотор. Наиболее хорошо инсуляторы исследованы у дрозофилы и позвоночных, именно они представлены в данном обзоре.

5 Выводы

1. Продемонстрирован один из механизмов образования химерных элементов и их перемещения. Такие события происходят в результате последовательных рекомбинаций между Р элементами химерного элемента из регуляторной области гена yellow и Р элементами, которые находятся в регуляторной области AS-C комплекса.

2. В химерном элементе обнаружен энхансер EnlA, который был перенесен в химерный элемент из дистального 1А района X хромосомы. Энхансер EnlA имеет неоднородную структуру: состоит из активаторной и коммуникаторной частей, которые вместе способны компенсировать отсутствие собственных энхансеров гена yellow и активировать экспрессию reHajye/W во всех кутикулярных структурах.

3. При изучении мутационных изменений химерного элемента был обнаружен регуляторный элемент 1A-RE, который активирует экспрессию yellow на большом расстоянии и является инсулятором, специфичным для гена yellow. Исследование 1А-RE при помощи трансгенных конструкций позволило выдвинуть гипотезу, что 1A-RE в исходном генетическом окружении в 1А районе X хромосомы выполняет функцию промотора гена CG3777 и взаимодействует с EnlA, расположенным в интроне этого гена.

4. Показано, что одна копия Su(Hw) инсулятора может эффективно блокировать взаимодействие между энхансером и промотором, которые находятся на расстоянии 7-9 т.п.н. друг от друга. Однако добавление второй копии инсулятора, в результате чего энхансеры оказываются между двумя Su(Hw) инсуляторами, расположенными на расстоянии 10 т.п.н. друг от друга, приводит к восстановлению взаимодействия между энхансером и промотором (нейтрализации активности инсулятора). Таким образом, взаимодействующие Su(Hw) инсуляторы не формируют независимых доменов транскрипции, как предполагалось ранее.
