VI. выводы

1. Адаптированная методика геномной in situ гибридизации позволяет идентифицировать геномный состав межвидовых гибридов лилий, получаемых с использованием 2п-гамет. Установлен механизм формирования 2п-гамет основанный на редукции первого деления мейоза (FDR). С использованием разработанного метода многоцветной геномной in situ гибридизации для лилий впервые установлена возможность создания трехгеномных гибридов (2п=36). Показана возможность преодоления стерильности межвидовых гибридов лилий и получения форм с различным геномным составом при использовании образцов формирующих 2п-гаметы.

2. Оптимизированная методика совместного применения дифференциального окрашивания хромосом, геномной гибридизации in situ и ДНК-маркирования, позволяет проводить эффективное выявление межгеномных транслокаций у тритикале.

3. Высокоповторяющиеся геномспецифичные последовательности ДНК на примере GenBR-1 (специфичного для генома В) могут быть использованы в качестве молекулярно-цитогенетических маркеров геномов капустных.

4. Высокоповторяющиеся последовательности ДНК Ty-l-copia-подобшлх ретротранспозонов являются эффективными молекулярно-цитогенетическими маркерами для изучения организации геномов растений. Показано, что у томата ретротранспозоны локализованы в прицентромерных гетерохроматиновых регионах хромосом. Подтверждена эффективность использования пахитенных хромосом томата для такого анализа. Прицентромерная локализация ретротранспозонов указывает на невысокую эффективность ДНК маркеров создаваемых на их основе для маркирования хозяйственно-ценных признаков томата. В тоже время .для растений с крупными геномами (лилии, крокусы, тюльпаны и лук) показано равномерное распределение Ty-1-copia-подобных ретротранспозонов, за исключением С-бэндинг положительных регионов хромосом, где они не выявляются.

5. Клонирована высокоповторяющаяся последовательность ДНК хмеля — размером 378 п.о., которая является эффективным цитогенетическим маркером для идентификации половых хромосом хмеля. С помощью FISH анализа показана субтеломерная локализация этой последовательности на аутосомах. Для X и Y хромосом выявлено характерное распределение сайтов гибридизации. Поиск в геномных базах данных не выявил гомологий с ранее клонированными последовательностями ДНК.

6. Установлено, что последовательности генов рибосомной РНК (45 S и 5S) являются эффективными цитогенетическими маркерами для идентификации трех из 10 хромосом хмеля с использованием флуоресцентной гибридизации in situ.

7. Выявлен высокий полиморфизм межмикросателлитных последовательностей ДНК (ISSR) на видовом и межвидовом уровнях у видов Solanum, Humulus lupulus и Rubus. Рекомендован набор ISSR праймеров для анализа исходных родительских и гибридных форм при внутри- и межвидовой гибридизации, а также форм растений с различным уровнем интрогрессии генетического материала. На основе выявленного полиморфизма ISSR-маркеров создана интегрированная генетическая карта межмикросателлитных последовательностей ДНК томата и высокоэффективные ДНК-маркеры для выявления пола у растений хмеля и признака ремонтантности малины.

8. Созданы эффективные SCAR маркеры для выявления пола растений хмеля.

9. Идентифицированые молекулярные маркеры WA-типа ЦМС риса и молекулярные маркеры, тесно сцепленные с геном восстановления фертильности Rf4, могут быть использованы в селекционных программах по созданию гибридного риса.

10. Клонированы фрагменты последовательностей генов устойчивости подсолнечника. Установлена их высокая степень гомологии: с ранее клонированными генами устойчивости растений.

11. Продемонстрирована высокая эффективность сравнительного анализа локусов генов устойчивости устойчивых и неустойчивых генотипов растений для создания надежных ДНК-маркеров. Сравнительный анализ ДНК последовательностей показал высокую степень гомологии между нуклеотидными последовательностями локусов Ve и 12 у устойчивых и неустойчивых генотипов томата. У аллеля i2 неустойчивого генотипа. S.lycopersicum выявлены последовательность, имеющая высокую степень гомологии.к ,гену I2G-5, и ранее неизвестная последовательность ДНК, гомологичная гену 12 S. pimpinellifolium. Создан ДНК маркер гена устойчивости к фузариозному увяданию томата. Выявлен полиморфизм по сайту, рестрикции рестриктазы Xba I между аллелями* гена Ve устойчивых и, неустойчивых генотипов и на основании данного полиморфизма создан кодоминантныи CAPS-маркер.

12. Установлено наличие Ogura-типа ЦМС, возникшей в,растениях капусты, пекинской Brassica pekinensis. Jusl, в результате межвидовой гибридизации с турнепсом и капустой абиссинской.

13. Созданные ДНК диагностикумы на гены устойчивости томата - к фузариозу и вертициллезу, хмеля — на определение пола растений, риса — на ЦМС и ген восстановления фертильности, и оптимизированые условия применения ДНК-маркеров на гены устойчивости томата к нематоде, ВТМ, кладоспориозу позволяют в значительно мере (в два раза) ускорять селекционный процесс по вышеуказанным генам.

14. Предложенью оптимальные высокопроизводительные методики выделения ДНК позволяют проводить массовый скрининг селекционного материала.

15. Разработаные методы и схемы высокопроизводительной оценки на наличие хозяйственно-ценных признаков в исходном селекционном материале с помощью ДНК-технологий дают возможность создавать перспективные формы томата, сочетающие несколько генов устойчивости к болезням и вредителям в одном генотипе, в частности в созданном коммерческом гибриде Б] Албаши.

V. ЗАКЛЮЧЕНИЕ.

Нами продемонстрированы возможности молекулярно-генетических и молекулярно-цитогенетических подходов для создания селекционного' материала растений с заданными свойствами. Эффективность применения таких методов зависит от каждого конкретного случая и напрямую связана с возможностью ускорения селекционного процесса. Стратегия применения должна основываться на надежности, эффективности, информативности и взаимодополняемости вышеуказанных подходов, а также базироваться на достижениях в геномике и биоинформатике.

В программах с использованием методов отдаленной гибридизации на первых этапах главную роль могут играть современные молекулярно-цитогенетические методы исследований, такие как геномная гибридизация in situ, ПЦР анализ родительских геномов и их гибридов с использованием диспергированных повторяющихся последовательностей ДНК и др. Несмотря на трудоемкость и невысокую производительность таких методов, они позволяют выработать оптимальную стратегию для быстрого и контролируемого переноса генетического материала от диких сородичей в культурные растения. В дальнейшем, при создании сортов и коммерческих гибридов.Fj на первый, план должны выходить молекулярно-генетические подходы, позволяющие быстро, надежно и качественно охарактеризовать селекционный материал на наличие тех или иных хозяйственно-ценных генов. И здесь очень важную роль играет наличие ДНК маркеров тесно-сцепленных с генами, контролирующими эти признаки. Такие маркеры (SCAR, CAPS, SSR) в купе с высокопроизводительными технологиями ДНК анализа (выделение ДНК, ПЦР, RT-PCR) могут значительно ускорить селекционный процесс при создании сортов и коммерческих гибридов Fj с заданными свойствами.
