Выводы

1. Установлено, что в конструкции e-h с обеих сторон от энхансера мобильного элемента copia происходит изменение структуры хроматина. В районах, прилегающих к энхансеру, возрастает уровень таких компонентов активного хроматина, как РНК-полимераза II и ТВР (ТАТА-связывающий белок), а также - меток активного хроматина НЗК9Ас и НЗК14Ас. Инсулятор gypsy в конструкции e-g-h приводит к ингибированию связывания РНК-полимеразы II и ТВР и уменьшению уровня ацетилирования гистона НЗ в этих же областях.

2. Энхансер мобильного элемента copia в конструкции e-h вызывает увеличение доступности хроматина для ДНКазы I во всех исследованных районах данной конструкции. Наибольшее влияние он оказывает на область, содержащую репортёрный ген люциферазы, а наименьшее — на область промотора. Район самого энхансера также имеет открытую структуру хроматина, доступную для ДНКазы I. Инсулятор gypsy в конструкции e-g-h приводит к «закрыванию» хроматина. Наибольшее влияние он оказывает на области репортёрного гена, энхансера и промотора.

3. Пограничные районы форум-доменов 70С и 84D содержат сайленсеры различных типов: 70С проявляет полярные свойства, a 84D — неполярные.

4. Пограничные районы форум-доменов 70С и 84D, изученные в. данной работе, обогащены модифицированными гистонами, характерными для репрессированного состояния хроматина: район 84D содержит НЗК9шеЗ и H3K27me3, а район 70С содержит НЗК27теЗ и НЗК4теЗ, одновременное присутствие которых встречается в областях бивалентного хроматина.
