Благодаря фотосинтезу, растения и фотосинтезирующие бактерии улавливают солнечную энергию. В результате их жизнедеятельности ежегодно в атмосферу Земли выделяются миллиарды тонн молекулярного кислорода и образуются сложные органические соединения. Аэробные организмы получают необходимую для жизнедеятельности энергию из внешней среды в процессе дыхания, то есть в ходе окисления органических соединений кислородом воздуха. Важнейшую роль в дыхательных и фотосинтетических процессах преобразования энергии играют электронтранспортные цепи митохондрий, бактерий и хлоропластов. Ключевым звеном цепей переноса электронов являются энерго-преобразующие мембранные ферменты.

В митохондриях и аэробных бактериях трансформация энергии происходит в дыхательной цепи - совокупности сложно организованных ферментных комплексов, локализованных во внутренней мембране митохондрий эукариотических клеток или в цитоплазматической мембране прокариот. В процессе клеточного дыхания энергия окислительно-восстановительных реакций трансформируется электронтранспортной дыхательной цепью в разность электрохимических потенциалов ионов водорода на сопрягающей мембране (АцН+). Возникающая таким образом протон-движущая сила служит в клетке источником энергии в реакциях биохимического синтеза, мембранного транспорта, механического движения бактериальных клеток и тд.

В концевом участке дыхательных цепей аэробных организмов образование протон-движущей силы обеспечивается в результате работы терминальных оксидаз, катализирующих восстановление кислорода цитохромом с или хинолом. Основная подгруппа этих ферментов, включающая цитохромоксидазу (ЦО) из митохондрий, образует суперсемейство структурно-родственных гем-медных оксидаз. Отличительной особенностью представителей суперсемейства является каталитический центр, который образован близко расположенными ионом железа гемовой группы и ионом меди (т.н. биядерный или бинуклеарный центр, ВЫС). В биядерном центре, непосредственно, происходит восстановление молекулы кислорода с образованием двух молекул воды.

В ходе каталитического цикла, цитохромоксидаза образует ДцН+ в результате двух процессов. Во-первых, электроны поступают в ВИС от цитохрома с с периплазматической стороны мембраны (Р-сторона), в то время как участвующие в восстановлении кислорода и образовании молекул воды протоны (т.н. "субстратные" или "химические" протоны) переносятся электрогенно в ВИС из матриксной водной фазы (И-сторон а). Во-вторых, цитохромоксидаза является редокс-зависимым протонным насосом и дополнительно переносит с Ы-стороны на Р-сторону мембраны в среднем четыре протона (т.н. "помпируемые" протоны) на каждую восстановленную молекулу кислорода.

Таким образом, цитохромоксидаза представляет собой сложную и уникальную в своем роде молекулярную машину, осуществляющую сопряжение одноэлектронного окисления цитохрома с, 4-х электронного восстановления и протонирования молекулы кислорода с образованием воды и редокс-зависимого направленного трансмембранного переноса протонов. Сочетание этих процессов требует сложной организации и точной взаимной артикуляции перемещения зарядов (электронов и протонов) и превращения молекулы кислорода в ходе каталитического цикла. Выяснение молекулярного механизма редокс-сопряженного разделения зарядов в цитохромоксидазе, описание динамики и траекторий внутрибелкового переноса протонов относятся к ряду наиболее важных и сложных задач биэнергетики, решение которых является необходимой предпосылкой для установления общих закономерностей механизмов образования трансмембранного потенциала ферментами энерго-сопрягающих мембран.

В каталитическом цикле цитохромоксидазы можно выделить две части: восстановительную и окислительную. Восстановительная часть включает в себя перенос от цитохрома с в окисленный биядерный центр 1-го и 2-го электронов. В результате оба редокс-центра ВИС (гемовое железо и атом меди) переходят в восстановленное состояние. В окислительной фазе молекула кислорода связывается с восстановленным атомом железа гема, входящего в состав В1ЧС. Далее происходит разрыв 0-0 связи, одновременно с которым 4 электрона переносятся из молекулы белка на атомы кислорода с образованием состояния Р фермента, в котором атом железа гема образует оксоферрильный комплекс. Каталитический цикл завершается переносом в ВЫС от цитохрома с последовательно 3-го и 4-го электронов, в результате которых происходит, соответственно, переход цитохромоксидазы из состояния Р в состояние Р и восстановление Р с образованием исходного, полностью окисленного состояния (О).

Каждая из отдельных одноэлектронных стадий каталитического цикла цитохромоксидазы включает в себя набор частных реакций переноса электронов и протонов, без детального исследования которых невозможно установить молекулярный механизм сопряженного трансмембранного переноса зарядов. Все эти реакции происходят во временном диапазоне, недоступном для методов быстрого смешивания. Первоначальные сведения о механизме сопряженного переноса протонов в цитохромоксидазе базировались на результатах различного рода стационарных измерений. В связи с этим, в наши задачи входило изучение механизма электрогенного переноса протонов в цитохромоксидазе с помощью предстационарных подходов.

Цель заключалась в том, чтобы максимально полно описать механику векторного переноса зарядов в цитохромоксидазе, разрешив во времени частные стадии электрогенного переноса электронов и протонов, составляющие суть отдельных одноэлектронных переходов в каталитическом цикле фермента. В задачи исследования входило выяснить точную стехиометрию переноса помпируемых протонов в отдельных одноэлектронных переходах каталитического цикла цитохромоксидазы. Установить сопряженные электрон-транспортные стадии и конформационные изменения, контролирующие частные стадии транслокации протонов. Выяснить точную структуру каталитических интермедиатов ЦО, включая промежуточную локализацию протонов в кислород-редуктазном центре и в его окрестности.

Возможности изучения промежуточных стадий в каталитическом цикле цитохромоксидазы в предстационарном режиме с адекватным временным разрешением появились сравнительно недавно, благодаря развитию импульсной лазерной техники и фотохимии. Фотоинициированная синхронизация электронтранспортных реакций в ЦО позволяет производить одновременный запуск реакции переноса электрона внутри всей популяции фермента, используя наносекундный импульс лазерного света, и исследовать фотоиндуцированные частные стадии транслокации зарядов и превращений каталитического центра с микросекундным временным разрешением.

Первый метод, использующийся в предстационарных измерениях ЦО, был разработан в 60-х годах Гиббсоном и Гринвудом и представляет собой фотолиз окиси углерода от восстановленного биядерного центра ЦО в присутствии молекулярного кислорода. Этот метод позволяет исследовать окисление изначально восстановленного фермента в режиме одного оборота каталитического цикла. Другой вариант представляет собой восстановительные одноэлектронные пульсы в цитохромоксидазу с помощью фотоактивных комплексов рутения. Этот подход был разработан первоначально Греем и соавторами в 80-х годах и адаптирован применительно к ЦО в начале 90-х Нильсоном.

Наиболее продуктивными подходами к мониторингу и изучению быстрых стадий переноса протонов и электронов в цитохромоксидазе оказались метод "прямой" электрометрической регистрации трансмембранной разности электрических потенциалов (емкостная потенциометрия) и разрешенная во времени спектрофотометрия. "Прямой" электрометрический метод был разработан изначально в лаборатории Л.А.Драчева в НИИ им. А.Н. Белозерского МГУ для исследования бактериородопсина и фотосинтетических реакционных центров и позволяет отслеживать в реальном времени электрогенную составляющую переноса зарядов в мембранных белках.

В последние годы для цитохромоксидазы из митохондрий и ряда бактерий была получена трехмерная кристаллическая структура с атомным разрешением. Это позволило локализовать гипотетические внутрибелковые структуры, содержащие консервативные аминокислотные остатки и потенциально способные к проведению протонов из внутренней водной фазы в гидрофобную область цитохромоксидазы к каталитическому двухядерному центру. В сочетании с возможностями направленного мутагенеза и кинетических методов с необходимым временным разрешением, это предоставляет уникальную возможность выяснения деталей механизма сопряжения энергии и внутрибелкового переноса протонов в данном ферментном комплексе. В связи с этим, в наши задачи входило сравнительное изучение механизма электрогенного переноса протонов в гомологичных бактериальных цитохромоксидазах с использованием возможностей направленных генетических манипуляций по замене и тестированию функциональной роли консервативных аминокислотных остатков в проведении протонов.

Цель заключалась в том, чтобы локализовать траектории проведения протонов в цитохромоксидазе и выявить взаимную артикуляции частных электрон и протон-транспортных реакций внутри отдельного одноэлектронного перехода фермента. Выяснить роль протон-проводящих путей в отдельных одноэлектронных переходах в каталитическом цикле ЦО. Выяснить структурно-функциональные особенности неканонических цитохромоксидаз, обусловливающие сниженную эффективность трансмембранной перекачки протонов.

1. Использование фотоактивируемых комплексов рутения в качестве донора электрона позволяет в предстационарном режиме исследовать отдельные стадии сопряженной транслокации электрона и протонов в каталитическом цикле цитохромоксидазы с микросекундным временным разрешением, недоступном при использовании методов быстрого смешивания.

2. Общая организация стадий внутрибелкового электрогенного переноса зарядов в терминальных гем-медных оксидазах семейства А (митохондриальной цитохромоксидазе и цитохромоксидазе ааз из К sphaeroid.es) сходна. Восстановление цитохромом с входного редокс-центра, Сид, является неэлектрогенной реакцией. Перенос электрона от Сид к гему а (т~ 10-40 мкс) сопровождается генерацией мембранного потенциала, соответствующей транслокации отрицательного заряда с внешней стороны мембраны на -1/3 толщины мембранного диэлектрика.

3. Перенос от гема а в биядерный кислород-редуктазный центр 3-го и 4-го электронов в каталитическом цикле цитохромоксидаз семейства А (переходы Р—»Б и Р—>0, соответственно) сопряжен с трансмембранной транслокацией одного помпируемого протона и переносом из внутренней водной фазы к биядерному центру одного субстратного протона

4. Сопряженный электрогенный перенос протонов в переходах Р—>Р и Р—Ю каталитического цикла цитохромоксидазы митохондрий включает две кинетически разрешаемые стадии. В первой стадии с т~0.3 мс (Р—>Р) и г~\2 мс (Б—Ю), соответственно, происходит перемещение помпируемого протона из внутренней водной фазы к промежуточному акцептору протона, располагающемуся над биядерным центром. Вторая стадия (т~1.3 мс и т~4.5 мс, соответственно) включает высвобождение помпируемого протона на внешнюю сторону мембраны, сопряженное с электрогенным перемещением субстратного протона с внутренней стороны мембраны к биядерному центру.

5. В каталитическом цикле цитохромоксидазы ааз из R.sphaeroides, протон-проводящий Б-канал служит для переноса всех помпируемых протонов, а также субстратных протонов на стадиях Р—»Е и Р—>0 переходов (перенос на кислород 3-го и 4-го электронов, соответственно). Протон-проводящий К-канал необходим для переноса субстратных протонов из внутренней водной фазы в ходе восстановления биядерного центра первыми двумя электронами в каталитическом цикле.

6. Снижение вдвое эффективности протон-транслоцирующей функции цитохромоксидазы баз из ТлкегторЫ'1ш семейства В гем-медных оксидаз обусловлено отсутствием сопряженного трансмембранного переноса протона в части одноэлектронных стадий каталитического цикла, тогда как другие одноэлектронные стадии сопряжены с трансмембранным переносом ~1 протона.

7. Цитохромоксидаза баз из ТлкегторкИш семейства В гем-медных оксидаз отличается от оксидаз семейства А локализацией внутреннего акцептора протона в каталитическом интермедиате, соответствующем во времени образованию состояния Б в реакции с кислородом в режиме одного молекулярного оборота. В цитохромоксидазах семейства А, перенос субстратного протона на данной каталитической стадии происходит в биядерный центр (к связанному с Сив гидроксил-аниону), тогда как в цитохромоксидазе баз акцептор протона локализован вне биядерного центра.

8. При восстановлении низкоспинового гема б каталитический биядерный центр окисленной цитохромоксидазы баз из ТлИегторкИш переходит в открытое состояние и приобретает способность связывать внешние лиганды.
