Введение

Актуальность исследования. Настоящая работа представляет собой подробное описание пространственно-временного паттерна распределения двух нейромедиаторных веществ (серотонина и RF-амидов) в онтогенезе четырех представителей типа Стрекающие (Cnidaria). В качестве объектов исследования выбрано четыре вида, два из которых относятся к классу гидроидных (Hydrozoa) и два - к классу сцифоидных (Scyphozoa). До последнего времени строение нервной системы было наиболее изучено у представителей классов гидроидных и кораллов, в то время как классу сцифоидных в этом аспекте было уделено значительно меньше внимания. Эмбриональное развитие стрекающих, включая проблему дифференцировки клеток в ходе эмбриогенеза, также долгое время оставалось в тени. Актуальность данной работы заключается также в выборе объектов, поскольку тип Стрекающие (Cnidaria) является одной из наиболее древних групп многоклеточных, характеризующейся наличием нервной системы. Следовательно, можно ожидать, что эти животные сохранили в себе предковые черты организации нервной системы. С этой точки зрения, предметом обсуждения может являться значительное разнообразие, а также широкий спектр функций нервной системы, зарегистрированные у изученных представителей типа Стрекающие (Galliot et al., 2009; Watanabe et al., 2009). Особый интерес представляет динамика нервных структур и их изменение в течение онтогенеза каждого представителя, поскольку большинство стрекающих характеризуются сложным метагенетическим жизненным циклом, включающим не только личиночную стадию, но также и чередование поколений с бесполым и половым размножением (полипоидную и медузоидную стадии, соответственно).

Цель работы: провести сравнительный анализ развития нервной системы и роли нейромедиаторных веществ (серотонин и RF-амиды) в онтогенезе стрекающих на примере Gonothyraea loveni (Hydrozoa), Cladonema radiatum (Hydrozoa), Aurelia aurita (Scyphozoa) и Cassiopea andromeda (Scyphozoa).

Для этого необходимо было решить следующие задачи:

• Изучить распределение иммунореактивности к нейромедиаторным веществам на определенных стадиях онтогенеза представителей класса Hydrozoa (Cladonema radiatum и Gonothyraea loveni) и Scyphozoa {Aurelia aurita и Cassiopea andromeda).

• Проследить динамику развития нервной системы на протяжении жизненного цикла исследуемых видов стрекающих.

• Определить принадлежность серотонин- и (FM)RF-aMHfl-положительных клеток к тому или иному гистологическому типу.

• Выявить функции серотонинэргической системы, её возможную связь с развитием и метаморфозом личинок.

Новизна исследования. В работе впервые прослежено пространственно-временное распределение серотонина в онтогенезе сцифоидных и гидроидных. Высказано предположение, что серотонин играет роль в ходе метаморфоза личинки-планулы, а также планулоподобной почки (при бесполом размножении). Серотонинэргическая нервная система, выявленная у гидрантов Cladonema radiatum, является первым доказательством наличия нейрональных сигналов серотонина у полипов гидроидных.

В работе впервые показано наличие нервных клеток в ценосарке побега колониальных гидроидных на примере Gonothyraea loveni. Впервые описан процесс дифференцировки и усложнения нервной системы в ходе закладки новых элементов колонии (междоузлий) у G. loveni.

Теоретическое и практическое значение. Данное исследование создаёт предпосылки для последующего детального изучения определённых аспектов биологии стрекающих. С теоретической точки зрения наиболее важным моментом в работе является сравнение организации нервной системы на разных стадиях развития как у одного вида, так и у разных видов, относящихся к разным отрядам, или даже классам. Результаты настоящего исследования могут быть использованы для изучения особенностей поведения и жизнедеятельности изученных видов и углублённого исследования сравнительной нейробиологии, что позволит судить о родстве и происхождении исследованных видов стрекающих. С практической точки зрения представляют интерес методы, отработанные в ходе изучения выбранных объектов и представляющие собой тонкие модификации стандартных протоколов. Основные результаты, полученные в работе, будут использованы в спецкурсах по зоологии и биологии развития беспозвоночных, читаемых на биологическом факультете МГУ им. М.В. Ломоносова.

Объекты исследования. В качестве объектов изучения были выбраны два представителя класса Hydrozoa (Gonothyraea loveni и Cladonema radiatum) и два представителя класса Scyphozoa {Aurelia aurita и Cassiopea andromedd). Внутри классов, выбранные представители относятся к разным отрядам, что предполагает определённые различие во внутренней организации животных, включая нервную систему. В то же время, особенности морфологии, поведения и жизненного цикла всех исследованных видов были в той или иной степени описаны ранее. Это позволило в ходе работы наиболее продуктивно подходить к вопросам изучения нервной системы этих объектов, учитывая их биологию.

Исследование нервной системы стрекающих минимально охватывает

представителей класса сцифоидных, так же как и представителей отряда

ТЬесарЬога класса гидроидных. Этот факт стал основным при выборе объектов исследования, вследствие чего в работу было включено изучение двух представителей класса БсурЬогоа и гидроида О. Ьует, относящегося к отряду ТЪесарИога.

Сопогкугаеа 1оует и А. а иг Па являются видами, вынашивающими потомство, что делает их методически удобными для изучения ранних стадий развития. Также для планул этих видов известны способы инициации метаморфоза. Эти особенности создают методические предпосылки детального изучения пространственно-временной динамики организации нервной системы на протяжении всего жизненного цикла указанных видов.

Сцифоидное Сазяюреа апйготесЬ было выбрано благодаря особенности бесполого размножения, в процессе которого сцифистома С. апс1готес1а продуцирует свободноплавающие планулоподобные почки. В связи с этим, особое внимание в работе уделено сравнению планулоподобных почек С. апс1готес1а и планул С. 1оует и А. аигНа, являющихся результатом полового процесса.

Гидроид С. гасИшит стал объектом настоящего исследования вследствие особой стратегии захвата добычи, которая заключается в активном движении гидранта навстречу жертве с последующим удерживанием и поднесением добычи к гипостому фактически одним щупальцем, что требует хорошо развитых сенсорной, регулирующей и сократительной систем.
