1.1 Актуальность проблемы.

Стремительный прогресс в изучении молекулярных механизмов канцерогенеза в последние десятилетия связан с открытием большой группы генов, которые были объединены под общим названием протоонкогенов. Было показано, что в нормальных клетках они участвуют в регуляции клеточной пролиферации на всех уровнях передачи митогенных сигналов: от рецепторов и сигнальных белков сигнальных каскадов (протеинкиназы sarc, raf, mos, erb-B) до ядерных транскрипционных факторов (reí, fos, jun). Мутации в протонкогенах превращают их в онкогены, что, как правило, нарушает строгую регуляцию передачи митогенных сигналов. Именно это свойство лежит в основе автономной пролиферации опухолевых клеток.

Открытие метода переноса генов в клетки эукариот позволило получить ряд новых данных о молекулярных механизмах канцерогенеза и идентифицировать клеточные мишени, поражение которых необходимо для трансформации клеток. В частности, было показано, что введение ядерного онкогена El А аденовируса 5 типа нарушает строгий контроль за прохождением клеток по циклу путем инактивации продуктов генов - опухолевых супрессоров (Weinberg, 1991) и изменения активности ряда транскрипционных факторов (АР-1, NF-kB, ATF-2, ), регулирующих продвижение клетки по клеточному циклу (Hagmeyer, 1993). Для полной клеточной трансформации необходимо введение комплементирующего онкогена, например Ras (Ruley, 1983). Один из наиболее часто мутирующих в человеческих опухолях генов - протоонкоген cHa-ras активирует МАР-киназные каскады, имитируя действие ростовых факторов (Campbell, 1998). Единичные аминокислотные замены в 12, 13 и 61 положениях приводят к появлению постоянно активированных форм Ras белков. Экспрессия такой формы Ras ведет к конститутивной активации митогенных каскадов независимо от экзогенных ростовых факторов, что, по-видимому, лежит в основе неконтролируемой пролиферации опухолевых клеток. Конечной целью сигнальных путей являются транскрипционные факторы, участвующие в регуляции клеточной пролиферации, такие как: c-Jun, Elk-1, р53, ATF-2 и др. (Garrington and Johnson, 1999). Поэтому изучение активности различных МАР-киназных каскадов в связи с поведением транскрипционных факторов может пролить свет на молекулярные механизмы канцерогенеза.

Недавно появились данные, демонстрирующие участие протоонкогенов семейства Reí, кодирующих транскрипционный фактор NF-кВ, в регуляции клеточной пролиферации, апоптоза, а также в неопластической трансформации клеток in vitro и in vivo (Sonenshein, 1997; Gilmore et al, 1996). Например, транслокация и амплификация генов, кодирующих Rel/NF-кВ и их ингибиторов 1кВ, наблюдается в 20-25% случаев лимфоидных опухолей человека. Также было обнаружено, что активация NF-кВ является необходимым условием клеточной трансформации мышиных эмбриональных фибробластов NIH3T3 онкогеном cHa-Ras (Baltimore, 1997). В связи с этим, детальное исследование закономерностей регуляции транскрипционных факторов семейства Reí в нормальных и трансформированных клетках представляется весьма актуальным для понимания молекулярных механизмов, лежащих в основе превращения нормальных клеток в опухолевые.

ВЫВОДЫ.

1. Трансформация первичных клеток эмбриональных фибробластов крысы комплементирующими онкогенами Е1А+сНа-газ вызывает значительные изменения в регуляции транскрипционного комплекса МмсВ:

2. Трансактивирующая субъединица транскрипционного фактора кВ р65/Яе1А постоянно локализована в ядре трансформированных клеток, в отличие от нормальных клеток КЕБ, где внутриклеточная локализация регулируется ростовыми факторами сыворотки.

3. Ядерная локализация р65/Яе1А является следствием нарушения системы негативного контроля активности М^-кВ ингибиторными белками 1кВ-а.

4. ДНК-связывающая активность ЫР-кВ в трансформантах конститутивно высока и не регулируется ростовыми факторами и форболовым эфиром. Тем не менее, комплекс №"-кВ сохраняет способность к регуляции транс-активирующей активности при стимуляции сывороткой.

5. Установлено, что ядерные онкобелки Е1А находятся в физической ассоциации с субъединицей КБ-кВ- комплекса р65/Яе1А, что, возможно, определяет его постоянную ядерную локализацию и высокий трансактивирующий потенциал.
