Полиамидные миметики нуклеиновых кислот (ПАНКМ), первоначально описанные как пептидно-нуклеиновые кислоты (ае^-ПНК) [1], нашли широкое применение в различных областях физико-химической биологии. В частности на основе ПАНКМ {aeg-ПHK) предложены и реализованы технологии биосенсеров и микрочипов, системы для селективного расщепления нуклеиновых кислот, определения генетических мутаций и др [2, 3].

По своей функции классические ПАНКМ {aeg-YШK) являются миметиками нуклеиновых кислот, а по структуре представляют собой олигомеры на основе 14-(2-аминоэтил)-глицина с присоединенными через карбоксиметилированный спейсер нуклеиновыми основаниями [4].

Большой интерес представляет исследование свойств хиральных ПАНКМ, содержащих различные заместители в а- и у-положениях псевдопептидного фрагмента. Как известно, введение заместителей различного строения оказывает влияние на свойства ПАНКМ: растворимость, прохождение через клеточную и ядерную мембраны, направление закручивания спирали олигомера и гибридизационные свойства при связывании с природными нуклеиновыми кислотами [45].

На кафедре биотехнологии и бионанотехнологии МИТХТ им. М.В. Ломоносова проводится систематическое изучение модификаций нуклеиновых кислот - хиральных отрицательно заряженных ПАНКМ и незаряженных а- и у-ПАНКМ. Ранее в нашей лаборатории были синтезированы в препаративных количествах и полностью охарактеризованы четыре тиминсодержащих мономера ОЗ-ПАНКМ, а также два новых тиминсодержащих декамера ОЗ-ПАНКМ на основе ¿-глутаминовой кислоты и глицина.

В руководстве работой принимала участие к.х.н., доц. Кириллова Ю.Г. кислот различного строения на примере а- и у-метилзамещенных мономеров ПАНКМ, а также подход к определению оптической чистоты данных мономеров.

ЛИТЕРАТУРНЫЙ ОБЗОР

Разработка модифицированных олигонуклеотидов занимает одно из центральных мест в биотехнологии и медицинской химии ввиду использования их в качестве терапевтических агентов и инструментов молекулярной биологии. «Классические» пептидно-нуклеиновые кислоты (ПНК) представляют собой аналоги нуклеиновых кислот и являются полностью синтетическими ДНК/РНК-распознающими лигандами с нейтральным пептидо-подобным остовом и природными гетероциклическими основаниями. Такие свойства ПНК, как способность образовывать высокопрочные комплексы с нуклеиновыми кислотами даже при низкой ионной силе, биологическая и химическая устойчивость, могут служить основой для усовершенствования обычных, достаточно длительных и трудоемких методов диагностики нуклеиновых кислот.

1. Разработана и оптимизирована универсальная схема синтеза незаряженных хиральных мономеров ПАНКМ.

2. Синтезированы в препаративных количествах и полностью охарактеризованы четыре мономера хиральных а-ПАНКМ на основе псевдопептида 01уу/Ь-А1а и четырех нуклеиновых гетероциклических оснований, три из них получены впервые.

3. Синтезированы в препаративных количествах и полностью охарактеризованы четыре мономера хиральных у-ПАНКМ на основе Ь-А1ау/С1у и четырех гетероциклических оснований, три из них получены впервые.

4. Проведен анализ структуры полученных по разработанной схеме тимин-, аденин-, цитозин-, гуанинсодержащих мономеров на основе псевдопептидов С1уц/Ь-А1а и Ь-А1а\]/01у с помощью методов двумерной ЯМР-спектроскопии, который показал однозначность структуры выделенных продуктов реакции алкилирования гетероциклических нуклеиновых оснований бромацильными производными защищенных псевдопептидов 01уц/Ь-А1а и Ь-А1ац/01у.

5. Показано, что синтез незаряженных мономеров ПАНКМ идет энантиоселективно; разработанные нами подходы к определению энантиомерной чистоты мономеров с использованием ВЭЖХ позволяют однозначно подтвердить высокое значение энантиомерных избытков, достаточных для проведения последующих стадий олигомеризации.
