1. Осуществлен подбор четырех ДНК-маркеров, характеризующихся широким спектром аллелей и высокой гетерози-готностью, пригодных для популяционно-генетического анализа из числа коротких тандемных повторов (КТП) гипервариабельных локусов ДНК человека: НитАРА (АБ1) , локус Xqll.2-12; Ниш уТЯБ , локус - 12р12; НитНРКТВ (НРВД, локус - Хд2 6.1-д2 6.2 и НитЗТКХ1 (ЭТКХ1); на основе полимеразной цепной реакции (ПЦР), оптимизирован метод анализа их полиморфизма и созданы "лэддеры" для точного определения размеров аллелей каждого КТП.

2. Впервые для трех этнических групп СНГ определены число и частоты аллелей четырех КТП; установлены распределения генотипических классов, проанализированны частоты встречаемости гетерозигот и вычислены потенциалы индивидуализации.

3. По критерию согласия Колмогорова-Смирнова показано, что русская популяция достоверно отличается (р<0.05 -0.005) по четырем изученным КТП от узбеков и по трем (БТКХ1, НРИТВ, АИ) от грузин. Грузины и узбеки достоверно отличаются между собой по локусам НиппЛлГРэ-ю, НитНРКТВ, НитЭТЮа.

4. Наиболее информативными для популяционно-генетических исследований на уровне этнических групп и народов, а также для генетической идентификации личности являются КТП генов АИ и уЭДРд-ю, обладающие высокими индексами гетерозиготности (в среднем 0.70 и 0.77, соответственно) и выраженными потенциалами индивидуализации (0.05 и 0.12) .

5. На 50 полных семьях (более 100 мейозов) показан устойчивый менделевский тип наследования изученных КТП; в 250-ти экспертизах при установлении отцовства и генетической идентификации личности, проведенных с использованием КТП AR, HPRTB, STRX1, vWF9i0 наряду с сертифицированными для геномной дактилоскопии ДНК-зондами (АроВ, YNZ22, D1S80 и др.), подтверждена высокая информативность и надежность КТП. Среднее значение силы исключения, при совместном использовании четырех КТП, составило 99,97 %.

6. ПЦР-анализ КТП с мотивами повторов в 3 - 4 п.о. и размером идентифицируемых аллелей до 4 00 п.о. является удобным методическим подходом в популяционных и, особенно, в судебно-медицинских исследованиях.

ГЛАВА 6. ЗАКЛЮЧЕНИЕ

Полученные в нашей работе экспериментальные данные нужно рассматривать как предварительные, поскольку они г характеризуют не всю этническую группу грузин или узбеков в целом, а лишь представителей отдельных изолятов. Однако, уже на данном этапе они позволяют получить определенное представление о генетическом разнообразии трех различных этнических групп, в частности о том, что русские, грузины и узбеки имеют разные группы-основатели.

Обнаруженные в данной работе тенденции, а именно, "выпадение" некоторых аллелей в локусах с КТП по направлению от европеоидных народностей к азиатским не являются исключением (Рис. 21 и 22) , однако, получение полной этнической характеристики требует более широких популяционных исследований.

Важным представляется тот факт, что можно говорить о принципиальной возможности использования КТП, как молекулярных маркеров для генетической идентификации личности .

Большое число аллелей, высокий уровень гетерозигот-ности, возможность вести исследования на различных биологических объектах, при минимальном количестве материала (менее 100 нг ДНК-матрицы) и даже на образцах частично деградированной ДНК открывает широкие перспективы использования данных КТП для целей генетической идентификации личности (геномной дактилоскопии) при установлении отцовства или истинного виновника преступления в области судебной медицины.
