В настоящее время основным средством борьбы с кровососущими насекомыми остаются химические инсектициды, к которым у насекомых достаточно быстро развивается устойчивость, что требует либо повышения дозировок, либо ротации инсектицидов. Не обладая избирательностью действия, химические инсектициды вызывают гибель не целевых, зачастую полезных организмов. Накопление инсектицидов в природных компонентах (воде, почве и др.) делает их экологически опасными. Указанные недостатки обуславливают необходимость поиска новых экологически безопасных методов борьбы с кровососущими насекомыми. Для борьбы с взрослыми комарами альтернативы химическим препаратам до сих пор нет, однако для уничтожения личинок все шире применяют биологические препараты.

Использование бактериальных препаратов против насекомых, имеющих медицинское значение (в первую очередь против комаров и мошек), началось в конце 70 годов XX столетия. Связано это было с открытием бактерий Bacillus thuringiensis ssp. israelensis (Bti). Позднее было показано, что бактерии Bacillus sphaericus (Bsph) также могут быть использованы в качестве средств борьбы с личинками комаров.

Основным преимуществом биологических препаратов при сравнении с химическими является избирательность действия. Препараты на основе Bti используют на протяжении более 20 лет, и за это время не было зафиксировано ни одного случая их отрицательного воздействия на другие организмы. Не отмечено также возникновения у насекомых устойчивости к этим препаратам. Учитывая способность Bti к синтезу 4 типов белковых токсинов, вряд ли возможно прогнозировать появление устойчивости и в дальнейшем. Несколько иная картина выявляется при использовании препаратов, изготовленных на основе Bsph. Сохраняя высокую избирательность действия, определяемую двумя белками, составляющими бинарный токсин, препараты на основе Bsph при длительном применении в некоторых случаях приводят к появлению резистентных популяций комаров, что было выявлено в Индии, Бразилии, Франции. Тем не менее, отказываться от использования препаратов на основе этой бактерии нецелесообразно, т.к. их перспективность связана с длительным остаточным действием как в водоемах с чистой, так и загрязненной водой. Устойчивость насекомых к препаратам на основе Bsph возможно преодолеть ротацией с ßi-содержащими препаратами и другими ларвицидами.

К недостаткам бактериальных инсектицидов следует отнести относительно непродолжительное остаточное действие. Отсутствие воспроизводства бактерий Bti в природных условиях диктует необходимость повторных обработок водоёмов. Bsph способна воспроизводиться в окружающей среде на погибших личинках комаров, что делает препараты на их основе в ряде случаев более предпочтительными. Однако, следует отметить более узкий спектр инсектицидного действия Bsph, что не позволяет примененять их для борьбы с комарами р. Aedes и вынуждает увеличивать дозировки для воздействия на личинок p. Anopheles.

По экспертным оценкам в дальнейшем доля биологических препаратов для борьбы с кровососущими комарами должна увеличиваться за счет совершенствования препаративных форм, снижения стоимости и разработки рациональной тактики их применения (84, 85, 169, 189, 288). Приведённые соображения подтверждают актуальность проведенных исследований.

Цель и задачи исследования. Поиск и отбор новых активных видов и штаммов энтомопатогенных бактерий и совершенствование биологического метода борьбы с личинками кровососущих комаров. Дня достижения поставленной цели потребовалось решить следующие задачи:

• провести отбор новых более эффективных штаммов бактерий Bti и Bsph;

• осуществить поиск бактерий, обладающих ларвицидной активностью, в других таксонономических группах;

• оценить эффективность инсектицидного действия рекомбинатнтного штамма с клонированным геном синтеза ларвицидного токсина;

• выявить факторы, влияющие на сохранение и продолжительность ларвицидного действия Bti и Bsph в водной среде;

• изучить особенности взаимодействия энтомопатогенных бактерий с простейшими в водных экосистемах;

• создать биологическую систему, обеспечивающую длительное сохранение ларвицидных свойств энтомопатогенных бактерий.

Научная новизна результатов исследования. В результате проведенных исследований нами впервые:

• Выявлены и охарактеризованы новые штаммы бактерий Bti и Bsph, эффективность которых превышает известные ранее штаммы.

• Создан рекомбинанатный штамм Ме1ку1оЪасИ1и$ flagellatum с клонированным геном синтеза Сгу4В токсического белка Вй, эффективный по отношению к личинкам малярийных комаров.

• Выделены 4 кристаллообразующих штамма ВгепЬасШш ШегоБрогш, обладающих высокой ларвицидной активностью.

• - Определены факторы, влияющие на сохранение и продолжительность ларвицидного действия ВЦ и ВБрк. Установлена значительная устойчивость споро-кристаллической смеси Вй в водной среде (24 недели).

• Охарактеризованы параметры пролонгирования действия энтомопатогенных бактерий в водной среде. Сроки действия Вй зависят от исходной концентрации споро-кристаллического комплекса и численности личинок комаров.

• Показано, что простейшие Т. руг1/огт1в и Е. .товккоуБкИ потенциируют основные ларвицидные свойства энтомопатогенных бактерий Вй, Вярк, В. ШегоБрогт и рекомбинантного штамма М. flagellatum (снижение эффективных концентраций и увеличение скорости действия).

• Выявлены различия в характере взаимоотношений простейших с разными видами энтомопатогенных бактерий при длительном контакте.

• Разработана методология создания симбиотической системы Т. руп/огт15+ Вй, позволяющая без дополнительного введения бактерий поддерживать их активность на протяжении длительного времени (72 недели срок наблюдений).

Практическая ценность работы. Результаты исследований вносят вклад в решение важной медико-биологической проблемы - контроль численности комаров (переносчиков возбудителей инфекционных болезней) эффективными и экологически безопасными средствами. Значимость работы для практического использования заключается в следующем:

• Получены новые эффективные штаммы бактерий Вй и ВБрН, пригодные для промышленного производства.

• Выделенные кристаллообразующие штаммы В. laterosporus могут быть использованы в качестве альтернативы биологическим ларвицидам, изготовленным на основе Bti и Bsph.

• Доказана возможность пролонгирования действия при инкапсуляции споро-кристаллического комплекса Bti в инфузории T.pyriformis. Однократное введение этого комплекса в условия изолированных природных водоемов обеспечивает гибель личинок комаров рAnopheles на протяжении сезона потенциальной передачи малярии (10 недель).

• Предложена симбиотическая система «инфузории Т. pyriformis + бактерии Bti», сохраняющая ларвицидные свойства бактерий в течение 72 недель (срок наблюдений).

Материалы диссертации были использованы в педагогической работе на курсах, организованных ВОЗ для стран Европейского региона «Малярия и борьба с ней» (Москва, 2002г.) и для специалистов центров Госсанэпиднадзора Москвы и Московской области (2001-2002гг.), на курсах по систематике, морфологии, биологии, экологии переносчиков малярии Средней Азии и методам борьбы с ними (республика Таджикистан, г. Душанбе, 2004), а также на курсах усовершенствования для эпидемиологов, энтомологов ЦГСЭН на медико-профилактическом факультете ППОТОУ ММА им. И.М.Сеченова (2000-2004гг.)

Апробация работы. Основные результаты исследования были доложены и обсуждены со специалистами на чтениях, посвященных 60-летию кафедры медицинской энтомологии в РМАПО (1996), на Международном генетическом конгрессе в Бирмингеме (Англия, 1993), на рабочей встрече ВОЗ по генетическим изменениям ларвицидных бактерий, используемых для борьбы с переносчиками в Кордобе (Испания, 1996), на 9ой Международной конференции по бациллам в Лозанне (Щвейцария, 1997), на VII Съезде Всероссийского общества эпидемиологов, микробиологов и паразитологов (Москва, 1997), заседании общества протозоологов (Москва, 1999), на 10он Международной конференции по бациллам в Бавено (Италия, 1999), на ежегодном координационном совещании (Annual Coordination

Workshop US AID CDR/CAR and INTAS European Union, Ben Gurion, Israel, april-may, 1999), Субрегиональном совещании экспертов по проблеме пестицидов из числа стойких органических загрязнителей, интегрированным системахм защиты растений и контролю сельскохозяйственных вредителей и переносчиков инфекционных заболеваний (Санкт-Петербург, 2000 г), на конференции «Теория и практика борьбы с паразитарными болезнями» (Москва, 2002), в отделе биохимии Кембриджеского Университета (Англия, 2003), на Бюро отделения профилактической медицины РАМН (Москва, 2003), на Ученом Совете ИМПиТМ им. Диссертация апробирована на заседании Специализированной комиссии по предварительной экспертизе диссертаций в ИМПиТМ им. Е.И.Марциновского ГОУ ММА им. И.М.Сеченова (протокол №77 от 18 ноября 2003 г.).

Публикации по теме работы. Результаты диссертации отражены в 26 печатных работах (из них 6 в зарубежных изданиях), 1 СанПиНе, 4 методических указаниях, 2 патентах.

Положения, выносимые на защиту.

1. Направленный поиск природных видов и штаммов энтомопатогенных бактерий, выделение генов токсинообразования, получение рекомбинантных штаммов - продуцентов биологических инсектицидов, перспективных для промышленного производства, позволяют расширить возможности, повысить эффективность биологического метода борьбы с переносчиками возбудителей болезней и кровососущими насекомыми. Изучение выделенных нами штаммов Brevibacillus laterosporus доказало способность некоторых штаммов продуцировать кристаллы, обладающие инсектицидной активностью, что делает этот вид потенциальным кандидатом для биологической борьбы с кровососущими комарами. В результате проведения комплексных исследований по выделению и изучению природных бактериальных изолятов, получению генов токсинообразования и создания рекомбинантного штамма Methylobacillus flagellatum с клонированным геном синтеза Сгу4В токсического белка Bti предложены новые, обладающие инсектицидной активностью штаммы Bacillus thuringiensis var.israelensis 1-5 и JI2 и штамм Bacillus sphaericus 16 - S25, включенные во Всероссийскую коллекцию промышленных микроорганизмов, перспективных продуцентов биологических инсектицидов для борьбы с двукрылыми насекомыми.

2. Продолжительность действия биологических препаратов зависит от влияния ряда биотических и абиотических факторов. При этом выраженность воздействия одних и тех же факторов на различные биологические агенты существенно отличается. Сроки действия Bti и Bsph значительно варьируют в присутствии погибших личинок комаров и при увеличении исходной концентрации споро-кристаллической смеси. Наличие первого условия более важно для Bsph , а второго - для Bti.

Воздействие даже слабого неионизирующего излучения (микроволн и магнитных полей) приводит к снижению эффективности биологических препаратов. Поэтому антропогенные источники таких излучений могут опосредованно влиять на эффективность биологической борьбы с насекомыми.

3. Контакт простейших и энтомопатогенных бактерий потенциирует ларвицидные свойства последних. Обнаруженный феномен имеет универсальный характер образования симбиотической системы. При кратковременном контакте простейшие (Tetrahymena pyriformis и Entamoeba moshkovskii) увеличивают основные ларвицидные свойства энтомопатогенных бактерий Bti, Bsph, В. laterosporus и рекомбинантного штамма М. flagellatum (снижение эффективных концентраций и увеличение скорости действия). При длительном контакте инфузории Т. pyriformis пролонгируют инсектицидное действие бактерий Bti. Смесь Т. pyriformis и

Вй в условиях изолированных природных водоемов на личинках малярийных комаров первой генерации обеспечила их отсутствие на протяжении сезона потенциальной передачи малярии (2 месяца). Взаимодействие Т. руп/огт15 и Вй в питательной среде позволило без дополнительного введения бактерий поддерживать активность симбиотической системы на протяжении длительного времени -72 недели (срок наблюдений).

Выводы

1. Выделены из природных условий и охарактеризованы оригинальные штаммы энтомопатогенных бактерий Bti (2 штамма), Bsph (1 штамм) и 4 кристалл ообразующих штамма B.laterosporus (новый научный факт), пригодные для промышленного производства бактериальных препаратов для борьбы с личинками кровососущих комаров. На штаммы Bti и Bsph получены патенты № 2122791, 1998 и № 2080066, 1997. Штаммы депонированы во Всероссийскую коллекцию промышленных микроорганизмов.

2. Получен рекомбинантный штамм M.flagellatum с клонированным, оригинальным геном Сгу4В, ответственным за синтез токсического белка Bti, обладающий выраженной эффективностью по отношению к личинкам малярийных комаров и высоким потенциалом для промышленного производства.

3. Изучена устойчивость споро-кристаллических комплексов Bti и Bsph в водной среде. В модельных условиях, приближенных к природным, споро-кристаллический комплекс Bsph сохраняет активность свыше 60 недель, Bti-12 недель. Сохранение эффективности споро-кристаллического комплекса Bsph в условиях, приближенных к природным, объясняется размножением бактерий на каркасах погибших личинок, Bti практически не способны развиваться на погибших личинках. При отсутствии личинок споро-кристаллический комплекс Bti сохраняет свою активность в течение 24 недель, Bsph - 4 недели.

4. Даже слабое неионизирующее излучение - облучение магнитным А полем и микроволнами (десятки мкТ и до сотен мкВ/см соответственно) -препаратов, изготовленных на основе Bsph, уменьшает их инсектицидную активность. Это подтверждает, что антропогенные источники магнитных полей и микроволновых излучений могут негативно влиять на эффективность биологических препаратов.

5. Инкапсуляция энтомопатогенных бактерий ВН, Вэрк, В.ШегоБрогт и рекомбинантного штамма М. fla.gela.tum в простейшие (Т. руп^гт\Б и Е. тоэккоУБкИ) приводит к кратковременному усилению основных ларвицидных свойств (позволяет снизить концентрации вводимых бактерий и повышает скорость гибели личинок комаров).

6. Возникновение симбиоза в биологической системе Т.руп^гт1з + ВН приводит к пролонгированию (до 22 недель) ларвицидного эффекта бактерий ВН. С другими энтомопатогенными бактериями такого эффекта не наблюдается.

7. Отработана методология создания симбиотической перевиваемой культуры "Т.ру^огтьу+ ВН", позволяющая без дополнительного введения бактерий поддерживать их ларвицидную активность на протяжении не менее 72 недель.

8. Однократное введение смеси споро-кристаллического комплекса ВН и Т.руггрогтгя в изолированные природные водоемы обеспечивает гибель личинок комаров рЛпорке1еБ на протяжении сезона потенциальной передачи малярии (срок наблюдений 10 недель).

Практические рекомендации.

Использование биологического метода борьбы с кровососущими комарами, несмотря на экологические преимущества (воздействие только на целевую фауну, отсутствие опасного загрязнения окружающей среды) пока сдерживается рядом проблем технико-экономического характера: относительно медленным проявлением ларвицидных свойств и непродолжительным остаточным действием, в результате чего требуются повторные обработки, которые приводят к увеличению стоимости противокомаринных мероприятий.

С целью увеличения эффективности бактериальных препаратов для борьбы с комарами нами проведен поиск новых штаммов бактерий из разных таксономических групп, выделены высокоактивные штаммы энтомопатогенных бактерий Bti, Bsph , а также впервые изолированы кристаллообразующие штаммы B.laterosporus. Созданный рекомбинантный штамм M.ßagellatum с клонированным, оригинальным геном, ответственным за синтез токсического белка Cry4B Bti, обладает выраженной эффективностью по отношению к личинкам малярийных комаров. Полученные штаммы могут быть использованы в качестве продуцентов биологических ларвицидных препаратов, что позволит значительно расширить их ассортимент.

Продолжительность действия бактериальных препаратов зависит от влияния ряда биотических и абиотических факторов. При этом выраженность воздействия одних и тех же факторов на различные биологические агенты существенно отличается. Сроки действия Bti и Bsph значительно варьируют в присутствии погибших личинок комаров и при увеличении исходной концентрации споро-кристаллической смеси. Наличие первого условия более важно для Bsph , а второго - для Bti. В модельных условиях, приближенных к природным (при наличии личинок), спорокристаллический комплекс ВБрк сохраняет активность свыше 60 недель, ВЦ -12 недель. При отсутствии личинок споро-кристаллический комплекс ВН сохраняет свою активность в течение 24 недель, Вьрк - 4 недели.

Таким образом, чем выше численность личинок в водоёмах, тем менее продолжительно действует ВН. Погибшие личинки незначительно пролонгируют действие ВН. Проблему увеличения продолжительности действия ВН может решить создание препаративной формы с дискретным выходом кристаллов из нее, т.к. в воде споры и кристаллы сохраняют свою активность не менее 6 месяцев.

При использовании препаратов, изготовленных на основе ВБрИ, в оптимальной концентрации высокая численность личинок увеличивает продолжительность их действия, т.к. в каркасах погибших личинок происходит активное размножение бактерии.

Для пролонгирования остаточного действия, ускорения гибели личинок комаров и снижения концентрации действующего вещества предложена методика инкапсуляции споро-кристаллических комплексов энтомопатогенных бактерий в простейшие (Т. руг1/огт1я и Е. тоБИкоУБки). Возникновение симбиоза в биологической системе Т.руп/огтгБ + ВЦ приводит к пролонгированию (до 22 недель) ларвицидного эффекта бактерий ВН. Лабораторные данные подтверждены в полевых условиях средней полосы России. Однократное введение с*меси споро-кристаллического комплекса ВН и Т.руп/огтгБ в изолированные природные водоемы обеспечивает гибель личинок комаров Ап. теББеае на протяжении сезона потенциальной передачи малярии (срок наблюдений 10 недель). Результаты наших исследований по созданию биологической системы ВН +Т.руН/огт1Б доказывают возможность устранения основного недостатка, который присущ современным бактериальным ларвицидам - непродолжительное остаточное действие в обработанных водоемах.

Практическое использование биологической системы ВЦ +Т.руп/огт1я в разных ландшафтно-климатических зонах необходимо предварять испытаниями по уточнению реальной эффективности и продолжительности ларвицидного действия в конкретных условиях в зависимости от специфики регионов и экологических требований разных видов комаров. Следует иметь в виду, что продолжительность может зависеть не только от конкретных природных условий, но и, как показано в диссертации, от антропогенного воздействия (неионизирующее излучение).
