Ионы кальция и процессы фосфорилирования/ дефосфорилирования белков используются клетками всех типов для передачи и регуляции самых разнообразных внешних и внутриклеточных сигналов, управляющих такими фундаментальными биологическими функциями, как клеточная пролиферация и дифференцировка, иммунитет, транспорт. Благодаря методическому удобству работы с фоторецепторными клетками (исключительно высокое содержание сигнальных белков, возможность "запуска" передачи сигнала светом), зрительный пигмент родопсин, принадлежащий к семейству сопряженных с С-белками мембранных рецепторов, и зрительный каскад родопсин - трансдуцин - сОМР-фосфодиэстераза послужили прототипами для большого числа работ по системам рецептор - С-белок -эффекторный белок, функционирующим в клетках других типов.

Многие эффекты Са2+ на системы клеточной сигнализации опосредованы Са2+-связывающими белками, мишенями для которых часто служат протеинкиназы и протеинфосфатазы. Са2+-связывающим белкам и механизмам регуляции ими процессов фосфорилирования/ дефосфорилирования посвящено большое число работ; фоторецепторная клетка в этом отношении - была исключение. К моменту начала настоящей работы была ясна общая картина и многие детали зрительной трансдукции у позвоночных животных, в частности, доказана важность фосфорилирования родопсина для атгенюации сигнала в зрительном каскаде и для световой адаптации фоторецепторной клетки. Однако систематическое и интенсивное исследование роли Са2+-связывающих белков в регуляции фототрансдукции у позвоночных животных, в том числе - в регуляции фосфорилирования зрительного рецептора (родопсина) и соответствующей протеинкиназы (родопсинкиназы), было начато благодаря обнаружению в 1991 году Са2+-связывающего белка рековерина, функция которого к моменту начала настоящей работы не была установлена.

Рековерин - родоначальник белков семейства нейрональных Са2+-сенсоров, как и остальные представители этого семейства, содержит на 1ч[-конце миристоильный остаток. В свободном от Са2+ состоянии миристоильный остаток рековерина погружен в гидрофобный "карман" белка, вследствие чего рековерин находится в растворимой фазе. В присутствии Са2+, благодаря экспонированию миристоильного остатка из гидрофобного "кармана", рековерин транслоцируется из раствора на

VI мембрану. Этот механизм, получивший название Са -миристоильного переключателя, впервые описан для рековерина и позднее - также для других нейрональных Са2+-сенсоров.

Актуальность данной работы обусловлена тем, что к моменту начала работы отсутствовала информация о (1) функции рековерина в фоторецепторных клетках и (и) механизме Са2+ -миристоильного переключателя рековерина и других представителей семейства нейрональных Са2+-сенсоров и о роли каждого Са2+-связывающего центра белка в работе этого механизма.

ЗАКЛЮЧЕНИЕ

С учетом результатов, полученных в настоящей работе, суммируем основные сведения по ключевым механизмам фототрансдукции по состоянию на сегодняшний день.

Темновое состояние фоторецепторной клетки: компоненты каскада (родопсин, трансдуцин, ФДЭ) находятся в неактивированном состоянии; [cGMP] высокая и катионные каналы в плазматической мембране

94открыты; [Са ]f высокая, поэтому: (i) кальмодулин а) способствует поддержанию cGMP-зависимых катионных каналов в открытом состоянии и б) активир-ует аденилатциклазу, что - через активацию РКА и ингибирова-ние фосдуцина - предотвращает ингибирование трансдуцина; (ii) АГЦ 1 и 2 не активируют гуанилатциклазу, синтез cGMP гуанилатциклазой идет с низкой скоростью; (iii) рековерин находится в комплексе с родопсинкиназой и предотвращает фосфорилирование родопсина. Поглощение кванта света приводит к активации компонентов каскада, падению цитоплазматического уровня cGMP, закрыванию cGMP-завиеимых катионных каналов, гиперполяризации плазматической мембраны НСП и электрофизиологическому ответу; [Ca2+]f и функциональное состояние Са2+ -связывающих белков (кальмодулина, АГЦ 1 и 2, рековерина) то же, что и в темновом состоянии.

Возврат фоторецепторной клетки к темновому состоянию (recovery): в дополнение к понижению цитоплазматической [cGMP] происходит падение [Ca2+]f, в результате: (i) кальмодулин а) диссоциирует из комплекса с cGMP-завиеимыми катионными каналами, что способствует связыванию cGMP с каналами и их открыванию и б) диссоциирует из комплекса с аденилатциклазой, что приводит к снижению ее активности и затем - через уменьшение уровня сАМР и активности РКА - к прекращению фосфорилирования фосдуцина, что создает условия для ингибирования трансдуцина фосдуцином; (ii) АГЦ 1 и 2 активируют гуанилатциклазу, синтез cGMP ускоряется, что сопровождается повышением концентрации cGMP и открыванием cGMP-зависимых катионных каналов; (iii) рековерин диссоциирует из комплекса с родопсинкиназой.

Особого рассмотрения требует вопрос о том, на какую из реакций -фосфорилирование "темнового" или фотовозбужденного родопсина -направлено ингибирующее действие рековерина.

Большинство результатов по Са2+ -зависимому ингибированию фосфорилирования родопсина рековерином было получено в условиях in vitro: на суспензии НСП или в реконструированной системе. Если, однако, принять, что эти результаты могут быть экстраполированы к физиологическим условиям и что функция рековерина (или по крайней мере одна из его функций) в фоторецепторной клетке состоит в зависимой регуляции родопсинкиназы, то, с учетом наших данных о способности рековерина ингибировать фосфорилирование как фотовозбужденного, так и "темнового" родопсина, следует рассмотреть следующие возможные механизмы его действия in vivo: (1) рековерин ингибирует родопсинкиназу только в реакции фосфорилирования фотовозбужденного родопсина, тем самым обеспечивая кальциевый контроль передачи сигнала в зрительном каскаде родопсин - трансдуцин -ФДЭ; (2) рековерин Са -зависимым образом ингибирует родопсинкиназу только в реакции фосфорилирования темнового родопсина, но не действует на фосфорилирование Rho* и, соответственно, на передачу сигнала в каскаде; (3) в фоторецепторной клетке реализуются обе указанные возможности (1) и (2). Если функция рековерина в фоторецепторной клетке действительно заключается в

Са2+

-зависимом ингибировании фосфорилирования "темнового" родопсина, или, иначе говоря, "фосфорилирования с высоким выходом", то возникает вопрос, каков может быть физиологический смысл описанного явления?

Ответ на этот вопрос указывают следующие весьма специфические характеристики палочек сетчатки: а) концентрация родопсина в фоторецепторной мембране составляет величину порядка ЗмМ, что совершенно беспрецедентно для мембранных рецепторов; б) палочка способна работать в режиме счетчика квантов, когда насыщение ее электрофизиологического ответа достигается при фотовозбуждении всего лишь нескольких десятков молекул родопсина на НСП. Вследствие этого концентрация Rho в НСП в физиологических условиях может на много порядков превышать концентрацию Rho*.

Огромное число данных по киназам, фосфорилирующим сопряженные с G-белками рецепторы (к ним относится также и родопсин), указывает на то, что их функция заключается в фосфорилировании и тем самым в "выключении" (в англ. литературе используется термин down-regulation) активированного рецептора (в нашем случае - фотовозбужденного родопсина). В большинстве клеток концентрация мембранных рецепторов ничтожно мала, и благодаря высокой специфичности киназ в отношении активированных рецепторов, по сравнению с неактивированными, вероятность побочной реакции фосфорилирования нестимулированного рецептора невелика. Иное дело -палочка сетчатки, где в норме концентрация активированного рецептора (Rho*) на порядки ниже, чем концентрация нестимулированного рецептора (Rho), а это создает условия для конкуренции "темнового" родопсина с фотовозбужденным пигментом за родопсинкиназу, концентрация которой в НСП более чем на 2 порядка ниже концентрации родопсина, и тем самым для ее участия в "паразитной" реакции фосфорилирования Rho.

Если это так, то схему функционирования рековерина в палочке сетчатки можно представить следующим образом:

- в темноте, когда [Са ^ в цитоплазме НСП высока, родопсинкиназа находится в комплексе с рековерином;

- свет вызывает появление Шю* и передачу сигнала в каскаде Шю* - трансдуцин - ФДЭ;

94- затем начинается снижение [Са в цитоплазме, как следствие, комплекс родопсинкиназа-рековерин диссоциирует, родопсинкиназа связывается с Шю*, активируется им и фосфорилирует его;

- фосфорилирование Шю* (в совокупности с некоторыми другими механизмами, функционирующими в НСП) выключает передачу сигнала в каскаде, при этом в цитоплазме НСП повышаются концентрации свМР и, что особенно важно в данном контексте, - [Са ;

- с увеличением числа фосфатных остатков, включенных в молекулу Шю*, сродство киназы к Шю*-Р снижается, она диссоциирует из комплекса, но фосфорилировать присутствующие в избытке (по отношению к Шю*) молекулы Шю не может, т.к. вновь связывается с рековерином и переходит в неактивную форму, - клетка вернулась к исходному "темновому" состоянию.

Таким образом, согласно нашей гипотезе, одной из функций рековерина в фоторецепторной клетке может быть предотвращение нежелательного фосфорилирования "темпового" родопсина родопсинкиназой, находящейся в активированном состоянии после ее диссоциации из комплекса с фотовозбужденным родопсином.
