ВВЕДЕНИЕ

Актуальность работы. Контаминация пищевых продуктов пестицидами является одной из основных угроз пищевой безопасности. Такое положение связано не только с тем фактом, что практически все пестициды являются канцерогенными и особо опасными веществами, но и с возрастающими объемами их использования (за последние несколько лет они увеличились во многих странах более чем на 30%). На территории Российской Федерации разрешено к использованию более 250 наименований действующих веществ пестицидов. Эта цифра возрастает более чем в 2 раза, если говорить о разрешенных пестицидах в других странах, а также неизрасходованных запасах устаревших, запрещенных и особо стойких пестицидов, существующих в объектах окружающей среды.

Сложившаяся обстановка усугубляется весьма слабым методическим обеспечением, не позволяющим выявить и малой доли всего разнообразия действующих веществ пестицидов. Методики определения пестицидов по ГОСТ и методическим указаниям в РФ оказываются весьма дорогостоящими, продолжительными, трудозатратными и предполагают использование больших объёмов высокотоксичных растворителей. Нормативные документы, как правило, предполагают проводить определение небольшого числа (2-5 наименований) наиболее широко применяемых и ожидаемых в данном объекте пестицидов. Лишь небольшое количество методик направлено на определение 6-15 пестицидов, однако, в большинстве случаев, это представители одного класса, или же проводится поэтапное (раздельное) определение соединений разных классов.

В настоящее время для извлечения пестицидов из овощей, фруктов и определения их методами ГЖХ(ВЭЖХ)-МС/МС используют

пробоподготовку по QuEChERS (Quick, Easy, Cheap, Effective, Rugged, Safe). Таким способом определяют более 100 пестицидов единовременно. Однако при такой пробоподготовке определение пестицидов более

доступными хроматографическими методами с диодно-матричным и электронно-захватным детекторами, а также методом ГЖХ(ВЭЖХ)-МС низкого разрешения затруднено в связи с недостаточной очисткой экстрактов для таких сложных матриц как мясо, молоко, зерно и корма. Кроме того, ГЖХ(ВЭЖХ)-МС/МС оборудование дорогостоящее и требует высокой квалификации оператора, поэтому его использование в рутинных анализах в настоящее время невыгодно.

Цель данной работы состояла в разработке экспрессного и простого способа одновременного определения пестицидов различных классов в продуктах питания и кормах из одной навески, путем совмещения пробоподготовки (ЗиЕСЬЕЫЗ и дисперсионной жидкостно-жидкостной микроэкстракции с последующим хроматографическим определением.

Для достижения поставленной цели необходимо было решить следующие задачи:

• установить возможность комбинации методов пробоподготовки РиЕСИЕЯЗ и дисперсионной жидкостно-жидкостной микроэкстракции (ДЖЖМЭ) для одновременного извлечения пестицидов различных классов из одной навески пробы;

• провести оптимизацию условий проведения подготовки проб для получения экстрактов необходимой для анализа хроматографическими методами чистоты при достаточной степени концентрирования;

• разработать способы определения широкого круга пестицидов различных классов в продуктах питания и кормах.

Научная новизна. Предложена методология извлечения 100 пестицидов 13 различных классов из одной навески образцов зерна, кормов, пищевых продуктов с использованием дисперсионной твердофазной микроэкстракции и ДЖЖМЭ, как метода дополнительной очистки и концентрирования экстракта.

Исследовано хроматографическое поведение наиболее актуальных пестицидов различных классов (ХОП, ФОП, пиретроиды, неоникотиноиды, карбаматы, производные мочевины, триазины, триазолы, производные феноксиуксусной кислоты, производные бензимидазола, фталимиды, дикарбоксимиды, производные имидазола) при их совместном присутствии в условиях газожидкостной и высокоэффективной жидкостной хроматографии.

Показана возможность хроматографического разделения смесей ХОП, ФОП, пиретроидов, триазинов, триазолов, производных бензимидазола, фталимидов, производных имидазола, дикарбоксимидов методом газовой хроматографии с электронно-захватным(ГХ-ЭЗД) или масс-спектрометрическим (ГХ-МС) детекторами, а также неоникотиноидов, карбаматов, триазинов, триазолов, пиретроидов, производных феноксиуксусной кислоты методом вывсокоэффективной жидкостной хроматографии с диодно-матричным детектором (ВЭЖХ-ДМД).

Предложено применение ДЖЖМЭ в качестве метода дополнительной очистки и концентрирования при извлечении пестицидов различной полярности.

Изучено влияние основных параметров проведения подготовки проб на эффективность экстракции пестицидов различной полярности при совмещении методов (ЗиЕСЬЕКБ и ДЖЖМЭ (полярность растворителей, объем растворителей, значение рН среды, ионная сила раствора, добавки высаливающих компонентов, сорбенты).

Практическая значимость работы. Предложены способы определения 100 пестицидов различных классов из одной навески овощей, фруктов, меда, молока, мяса, зерна и кормов. Пределы обнаружения и определения составили 0,05-2,5 и 0,14-3,6 мкг/кг соответственно в зависимости от методики анализа. Относительное стандартное отклонение результатов анализа не превышает 0,1. Разработанные методики превосходят действующие национальные стандарты: значительно снижены расходы и

продолжительность анализа, количество используемых токсичных растворителей, упрощена схема пробоподготовки.

Разработанный способ позволил решить актуальные на настоящий момент проблемы:

• значительно расширить число определяемых пестицидов из одной навески (44 наименования методом ГХ-ЭЗД, 69 наименований методом ГХ-МС, 30 наименований методом ВЭЖХ);

• сократить продолжительность анализа до 1-1,5 часов (в зависимости от метода) за счет разработки схем одновременного определения пестицидов из одной навески и с одновременным использованием ГХ-МС, ГХ-ЭЗД, ВЭЖХ-ДМД;

• значительно сокращены объемы используемых токсичных растворителей и реактивов;

• снижены затраты на проведение анализа.

Личный вклад автора заключался в проведении экспериментальных исследований по оптимизации условий хроматографического разделения пестицидов; оптимизации пробоподготовки QuEChERS и метода ДЖЖМЭ и их совмещении; разработке способов одновременного определения пестицидов из одной навески зерна, кормов и продуктов питания за один анализ; интерпретации результатов эксперимента, формулировании научных положений и выводов.

ВЫВОДЫ

1. Предложена методология одновременного определения 100 пестицидов из одной навески анализируемого материала пищевых продуктов и кормов.

2. Установлена возможность совмещения методов риЕСЬЕЯЗ и ДЖЖМЭ для одновременного извлечения и концентрирования различных классов пестицидов широкого диапазона полярности. Изучено влияние основных параметров подготовки проб на степень извлечения аналитов.

3. Проведено разделение смеси наиболее распространенных пестицидов классов ХОП, ФОП, пиретроиды, триазины, триазолы, производные бензимидазола, фталимиды, производные имидазола, дикарбоксимиды методом газовой хроматографии с электронно-захватным или масс-спектрометрическим детектором и пестицидов классов неоникотиноиды, карбаматы, триазины, триазолы, пиретроиды, производные феноксиуксусной кислоты) методом ВЭЖХ с диодно-матричным детектором. Изучено поведение пестицидов при их совместном присутствии в зависимости от параметров разделения.

4. Установлена возможность одновременного определения 30 полярных пестицидов методом ВЭЖХ с диодно-матричным детектором (продолжительность анализа 34 мин). Создана библиотека спектральных данных определяемых пестицидов для дополнительной идентификации и определения «чистоты» хроматографического пика.

5. Установлена возможность одновременного определения 70 неполярных пестицидов методом газовой хроматографии с электронно-захватным и масс-спектрометрическим детекторами (продолжительность анализа 60 и 25 мин соответственно).

6. Разработаны новые методики определения 100 пестицидов различных классов хроматографическими методами, основанными на совместном использовании дисперсионной твердофазной (СНгЕСЬЕКБ) и жидкостно-жидкостной микроэкстракции (ДЖЖМЭ) в пищевых продуктах и кормах из одной навески. Пределы обнаружения и определения снижены до 0,05-

2,5 мкг/кг и 0,14-3,6 мкг/кг соответственно в зависимости от методики анализа. Относительное стандартное отклонение результатов анализа не превышает 0,1. Разработанные методики превосходят действующие национальные стандарты: значительно снижены себестоимость, продолжительность анализа, количество используемых токсичных растворителей и реактивов, упрощена схема пробоподготовки.
