В настоящее время активно развивается область исследований, посвященная изучению взаимодействия с двухцепочечной ДНК низкомолекулярных органических соединений: антибиотиков, красителей, олигопептидов и других биологически активных соединений, взаимодействующих преимущественно с узкой бороздкой ДНК. Эти соединения являются не только объектами самостоятельного изучения, но и находят широкое применение в противоопухолевой и противовирусной терапии, а также в качестве ДНК-специфичных молекулярных зондов в медицинских и научных исследованиях.Перспективность развития исследований в области ДНК-специфичной фармакологии связана с возможностью создания низкомолекулярных соединений, способных проникать внутрь клетки и выполнять сложные биологические функции регуляторных белков: избирательно связываться с определенными последовательностями пар оснований ДНК и оказывать направленное воздействие на функционирование генома и клетки в целом. При этом важно отметить, что низкомолекулярные соединения обычно легко выводятся из организма и не вызывают нежелательных реакций иммунного ответа.В последнее время в химиотерапии рака и противовирусной терапии наблюдается тенденция замены традиционных лекарств на основе интеркаляторов и алкилирующих агентов, повреждающих ДНК неселективным способом, новыми агентами, нековалентно взаимодействующими с ДНК и влияющими на процессы ее репликации и транскрипции.Поэтому большой интерес представляет конструирование и синтез низкомолекулярных органических соединений, которые могли бы нековалентно и сайт-специфично связываться с узкой бороздкой ДНК.

Синтезированы семь ковалентно связанных димеров Хёхста 33258 (бис-ХТ и ДББИ), соединённые гибкими линкерами различной длины и состава.На примере 6nc-XT(NMe) установлено, что при взаимодействии с ДНК синтезирован ные димерные лиганды образуют три типа комплексов: первый тип имеет максимум КД при 370 нм, интенсивность его флуоресценции в присутствии избытка ДНК возрастает в 350 раз (максимумум эмиссии при 475 нм, максимумум возбуждения при 365 нм), он образуется при низких заполнениях ДНК лигандом Глиганд/ДНК < 0.05); второй тип имеет максимумом КД при 335 нм, не обладает заметной флуоресценцией, образуется при промежуточных заполнениях ДНК лигандом (0.05 < лиганд/ДНК < 0.1); третий тип имеет максимумом КД при 360 нм, не обладает заметной флуоресцен цией, образуется при высоких заполнениях ДНК лигандом (лиганд/ДНК > 0.1).Показано, что в комплексе с ДНК бис-ХТ и ДББИ локализуются в узкой бороздке Синтезированные соединения (бис-ХТ) являются новыми перспективными флуоресцентными красителями, способными проникать через клеточную и ядерную мембраны живых клеток и окрашивать ядра. Соединения бис-ХТ перспективны в цитогенетических исследованиях для дифференциальной окраски хромосом человека и животных.Установлено, что in vitro бис-ХТи ДББИ ингибируют в микро/наномолярных концентрациях ДНК-интегразу ВИЧ-1, один из ключевых ферментов репликации вируса иммунодефицита человека.
