выводы

1. Исследованные химические соединения обладают различным цитотоксическим и антипролиферативным действием на клетки эритромиелолейкозной линии человека К562, что объясняется их структурными особенностями и механизмом внутриклеточного действия. Жизнеспособность клеток в зависимости от условий обработки этими соединениями варьирует от 25% до 95%, а их пролифера-тивная активность частично или полностью подавляется.

2. Эритроидная дифференцировка опухолевых клеток К562 индуцируется нуклео-зидами, солями жирных кислот с укороченной углеводородной цепью, ДМСО, 2-Л/5,0<3, 4-Ш()0, ИРуО, циклофосфаном и 2-третбутиламино-4-тиоксо-4-хлорметил-1,3,4-тиазофосфол-2-ином. В клетках увеличивается активность 8-аминолевулинатсинтетазы, каспазы-3, повышается синтез гемоглобина. Соче-танное действие двух различных индукторов эритроидной дифференцировки вызывает либо синергический, либо антагонистический эффекты.

3. Миелоцитарная дифференцировка опухолевых клеток сопровождается экспрессией на их поверхности моноцито-макрофагальных - СОНЬ, СП 14, и мегака-риоцитарного - СИ41, маркеров, а также активацией гемоксигеназы и индуцируется преднизолоном, дексаметазоном, ФМА, 00, 2-Ме-4-N20, 4-И20, 2-анилино-4-тиоксо-4-хлорметил-1,3,4-тиазофосфол-2-ином.

4. Переключение процессов клеточной дифференцировки с эритроидного на мо-ноцито-макрофагальный или мегакариоцитарный влечет за собой переключение функциональной активности каспаз и модуляцию процессов апоптоза. Апоптоз, индуцированный в опухолевых клетках эритроидными индукторами, реализуется с участием каспазы-9. ФМА подавляет эритроидную, индуцирует моноцито-макрофагальную дифференцировку, блокирует активность каспазы-9 и апоптоз клеток К562, при этом активируется каспаза-6. Каспаза-3 в зависимости от условий участвует как в процессах дифференцировки, так и апоптоза опухолевых клеток.

5. Апоптоз опухолевых клеток К562, индуцированный стирильными производными тУ-оксидов хинолина и пиридина, а также производными тиазофосфола, осуществляется при участии митохондриального цитохрома С, каспаз-3, -8 и -9. Реагенты оказывают влияние на процессы окисления-восстановления цитохрома С, модулируют набухание-сжатие митохондрий и способствуют выходу данного белка в цитоплазму. Преобладание в цитоплазме клеток окисленной или восстановленной форм цитохрома С отражается на реализации программы апоптоза.

6. Изменение чувствительности опухолевых клеток К562 к лизису естественными клетками-киллерами лейкоцитов периферической крови человека зависит от типа химического соединения, его концентрации, времени воздействия, исходного состояния клеток, способных подвергаться дифференцировке и/или апоп-тозу. Обработка клеток индукторами эритроидной дифференцировки повышает их чувствительность к лизису. Влияние индукторов миелоцитарной дифференцировки и/или апоптоза зависит от процесса, первично индуцируемого в опухолевых клетках. Чувствительность клеток к лизису снижается, если первично индуцируется дифференцировка, и повышается, если индуцируются апоптоз и дифференцировка.

7. Эритроидная дифференцировка в клетках резистентных сублиний К56214-Ы(20, и особенно К562/2-0()0, протекает более интенсивно по сравнению с родительскими К562, при этом регистрируется резкое повышение экспрессии генов Р-глобина и каспазы-3. Экспрессия гена каспазы-9 не изменяется. Сублинии являются устойчивыми к апоптоз-индуцирующему действию исследуемых ксенобиотиков.

8. Процессы эритроидной дифференцировки и апоптоза в клетках К562Ая-р53, обработанных индукторами при 32°С, протекают более интенсивно по сравнению с родительской линией К562. В индукции эритроидной дифференцировки клеток участвует каспаза-3.

9. Стирильные производные /У-оксидов хинолина и пиридина, а также производные тиазофосфола являются наиболее перспективными реагентами для создания на их основе препаратов нового типа, использование которых может повысить эффективность лечения онкологических заболеваний.

ЗАКЛЮЧЕНИЕ

В заключение кратко остановимся на основных результатах экспериментальной части монографии, а также обозначим проблемы, которые требуют дальнейшей первостепенной разработки.

Цель представленных исследований заключалась в проведении сравнительного анализа биологического действия нескольких групп химических реагентов на индукцию или ингибирование процессов дифференцировки и апоптоза опухолевых клеток на примере эритролейкемической линии человека К562, а также модуляцию чувствительности опухолевых клеток указанной линии к НЦТ-лизису ЕКК. В работе предполагалось подобрать условия и оптимальные схемы обработки опухолевых клеток химическими реагентами, в результате которых наблюдалась бы одновременная индукция процессов дифференцировки и апоптоза клеток, либо модуляция только одного из обозначенных процессов, показать влияние индуцированных дифференцировки и/или апоптоза на чувствительность клеток к цито-токсическому лизису гетерологичными (спленоциты крыс) и гомологичными (ЛПК человека) клетками-эффекторами. Кроме того, предстояло установить взаимосвязь (либо ее отсутствие) между сходством (различием) в модуляции апоптоза и чувствительности клеток к лизису ЕКК реагентами, обладающими сходным типом дифференцирующего действия на опухолевые клетки. Поэтому в качестве специфических групп химических реагентов нами были использованы соединения с различным внутриклеточным механизмом действия: нуклеозиды (тимидин, ци-тидин, гуанозин), соли жирных кислот с укороченной углеводородной цепью (бу-тират, изобутират, изовалерат натрия), ДМСО, гормоны кортикостероидной природы и их синтетические производные (гидрокортизон, преднизолон, дексамета-зон), константные активаторы протеинкиназы С (ФМА), хинолин (0 и его производные (2-Ме0, (Ю, 2-Ме<20, 4-М()0, 2-Ме-4-Ы()0), в том числе новосинтезиро-ванные стирильные (2-0()0, 4-0()0, 4-Ы()0, 2-Ы8()0, 4-Ш()0), стирильное производное пиридина (йРуО), а также циклофосфан и новосинтезированные производные тиазофосфола.

В ходе проведенных исследований нами было установлено, что исследуемые химические реагенты, являющиеся модуляторами процессов эритроидной (нуклео-зиды, соли жирных кислот, ДМСО, 2-NSQO, 4-NSQO, DPyO, циклофосфан, трет-бутиламиновое производное тиазофосфола) и миелоидной (ФМА, дексаметазон, QO, 2-MeQO, 4-NQO, 2-Me-4-NQO, анилиновое производное тиазофосфола) диф-ференцировки клеток К562, также способны в условиях единичной или сочетанной обработки модулировать апоптоз опухолевых клеток. Так, большая часть используемых нами соединений является для клеток К562 сильными апоптогенами (ти-мидин, гуанозин, бутират натрия, TV-оксиды хинолина и пиридина), другие, напротив, либо не индуцируют апоптоз опухолевых клеток в выбранных нами условиях (цитидин, ДМСО, преднизолон, гидрокортизон), либо подавляют его (например, ФМА). Причем, тот или иной биологический эффект реагента на клетки К562 во многом определяется специфической химической структурой соединения.

В настоящее время известно, что апоптоз в опухолевых клетках может индуцироваться не только цитостатическими агентами, но также возникать в клетках-мишенях при взаимодействии с ЕКК и цитотоксическими Г-лимфоцитами (Podack, Kupfer, 1991: Barao, Ascensao, 1998). Нами показано, что дифференцировочный статус, а также степень индукции апоптоза эритролейкемических клеток К562 могут влиять на их чувствительность к литическому действию ЕКК. Обработка клеток высокими дозами индукторов приводит к быстрой гибели значительной части опухолевых клеток по типу апоптоза и/или некроза. Возникновение в организме подобного генерализованного процесса нежелательно, так как наряду с опухолевыми клетками в индукцию указанного процесса могут быть вовлечены нормальные клетки, в частности клетки костного мозга. При этом в диапазон разрушающего действия цитостатических средств попадает клеточный субстрат других жизненно важных органов, в совокупности создавая предпосылки для развития цито-статической болезни. Снижение дозы используемых соединений, как правило, сопровождается снижением их цитотоксического эффекта. Кроме того, низкие дозы реагентов могут стимулировать дифференцировку опухолевых клеток, которая в зависимости от условий может быть как обратимой, так и необратимой (терминальной). Финальным этапом терминальной дифференцировки является апоптоз клеток (Епифанова, 1997), тогда как обратимая дифференцировка может привести к сохранению части клеточной популяции с последующим появлением резистентного к индуктору клона. И в том, и в другом случае может иметь место модуляция чувствительности опухолевых клеток к лизису ЕКК. В связи с этим, при сочетан-ной обработке опухолевых клеток индукторами различных путей дифференциров-ки может наблюдаться как снижение, так и увеличение чувствительности клеток к лизису ЕКК. Поэтому при клиническом использовании комбинаций цитостатиков необходимо учитывать эффекты модуляции клеточных функций, в частности чувствительности к цитотоксическому лизису ЕКК, возникающие под действием или на фоне каждого составляющего комбинации. Результаты наших исследований свидетельствуют о том, что обработка клеток К562 индукторами эритроидной дифференцировки в большинстве случаев увеличивает (либо не изменяет) их чувствительность к литическому действию ЕКК. Напротив, миелоидные индукторы в зависимости от того, какой процесс при их действии в клетках модулируется первично (дифференцировка или апоптоз), способствуют снижению или увеличению чувствительности опухолевых клеток к лизису ЕКК. В представленных экспериментах нами не были использованы лимфокинактивированные, в частности интер-лейкином-2, ЕКК. Поэтому работы в этом направлении связаны с перспективой дальнейшей разработки и совершенствования метода адоптивной иммунотерапии, поскольку цитолитическая активность лимфокинактивированных киллерных клеток превышает таковую неактивированных на порядок и выше.

Нами показано, что в реализации процессов апоптоза и дифференцировки клеток К562 непосредственное участие принимают клеточные каспазы. Переключение процессов клеточной дифференцировки с одного пути на другой, например, с эритроидного (индуцированного нуклеозидами) на миелоцитарный (индуцированный ФМА) влечет за собой переключение работы каспаз и модуляцию процессов апоптоза. Так, например, индуцированная нуклеозидами эритроидная дифференцировка в опухолевых клетках сопровождается активацией каспаз-3 и -9 и запуском апоптоза. В обработанных ФМА клетках К562 наблюдается индукция мо-ноцито-макрофагальной дифференцировки, повышение активности каспаз-3 и -6 без фрагментации ДНК. В то время как при использовании в сочетании с нуклеозидами ФМА, наблюдается внутриклеточная активация каспазы-6, значимое снижение активности каспазы-3, инактивация каспазы-9 и ингибирование ФМА нук-леозид-индуцированного апоитоза. Таким образом, не исключено, что, каспазы могут выступать в роли ключевых сигнальных молекул клетки, воздействуя на которые можно регулировать протекание того или иного клеточного процесса. Поскольку каспазы принимают непосредственное участие в процессах дифференци-ровки и апоптоза клеток, возможно, данная группа ферментов вносит определенный вклад в модуляцию чувствительности опухолевых клеток к цитотоксическому лизису ЕКК. Поэтому определение активности каспаз (и других протеаз) в опухолевых клетках может выступать в роли дополнительного биохимического маркера, в определенной степени позволяющего судить о прогрессии опухолевого процесса, в том числе лейкоза.

Нами предпринята попытка определить возможный механизм апоптогенного действия стирильных 7У-оксидированных производных хинолина и пиридина, а также производных тиазофосфола на клетки К562. Указанные соединения являются интересными с точки зрения их биологических эффектов на клетки, поскольку имеют в своем составе несколько центров функциональной активности, определяющих плейотропность действия реагентов. Показано, что клеточная активация данных гетероциклов происходит при участии микросомальных монооксигеназ; в реализацию программы апоптоза вовлечены каспазы-9, -3 и -8, а также митохонд-риальный цитохром С. Кроме того, установлено, что изучаемые ксенобиотики могут существенно модулировать реакции окисления и восстановления данного белка. В свою очередь, в зависимости от того, в какой форме (окисленной или восстановленной) цитохром С присутствует в цитоплазме клеток процессы микросо-мального окисления ксенобиотиков могут замедляться или ускоряться, что также вносит определенный вклад в реализацию апоптоза.

Не исключено, что обнаруженный нами апоптогенный эффект исследуемых гетероциклов на клетки К562 является частным случаем, свойственным только данной экспериментальной системе. Поэтому необходимы более детальные исследования этого вопроса с использованием клеток других линий того же гистогене-тического происхождения и иных линий клеток. В зависимости от происхождения опухолей, эффекты одного и того же соединения на клетки могут существенно различаться. Кроме того, многое зависит от способа обработки опухолевых клеток: in vivo или in vitro. Дозы реагентов и временные интервалы, используемые для инкубации клеток in vitro, практически не приемлемы для обработки опухолевых клеток in vivo. В последнем случае они могут оказаться либо слишком низкими, в результате желаемый терапевтический эффект не будет достигнут, либо слишком высокими, тогда трудно сказать, все ли нормальные клетки организма останутся жизнеспособными, в то время как опухолевые будут повреждены. Поэтому необходимо проведение сравнительного анализа протекания процессов апоптоза в разных клетках при обработке какой-либо группой химических индукторов. Таким образом, используемые нами гетероциклы (в частности, стиральные TV-оксидированные производные хинолина и пиридина, производные тиазофосфола) могут рассматриваться как перспективные химические соединения, на основе которых возможен синтез реагентов со специфической антиопухолевой активностью. Безусловно, из-за повышенной токсичности таких соединений наиболее реальным способом увеличения их избирательного действия можно рассматривать направленный транспорт реагентов к месту патологического очага или к клеткам-мишеням. В качестве «транспортных средств» следует рассматривать липосомы, наночастицы, специфические белковые (например, а-фетопротеин) или вирусные векторы, некоторые модульные рекомбинантные транспортеры. Другой формой воздействия данных соединений на опухолевые клетки могут являться их активные внутриклеточные метаболиты, выделенные и синтезированные de novo. Как правило, такой вариант соединений более эффективен и используется клетками с наименьшими потерями.

Результаты проведенного исследования являются перспективными в первую очередь с точки зрения изучения механизмов и комплексной оценки взаимосвязи процессов пролиферации, дифференцировки и апоптоза клеток опухолевых линий на уровне первичных, вторичных и третичных мессенджеров, а также рекомендаций по влиянию на указанные клеточные функции и их дальнейшей корректировке. Сбой программы апоптоза в генетически дефектных клетках становится причиной развития многих злокачественных опухолей, в том числе лейкозов, традиционные способы лечения которых в настоящее время являются недостаточно эффективными. Очень часто повышенной устойчивостью к антиопухолевым агентам обладают вторичные опухоли, развивающиеся на месте успешно вылеченных первичных. Подобный эффект вызван изменившейся структурой генома клеток, случайными мутациями в таких генах как р53, Ьс1-2, Ьах и других, а также аномалиями митоза, количество которых прогрессирует со временем (Александрова, 2004).

Нами показано усиление эритроидной дифференцировки и апоптоза клеток К562 с трансфецированным мутантным р53 (К562/{з-р53) по сравнению с родительскими клетками в условиях обработки тимидином, бутиратом натрия или ДМСО при 32°С. Кроме того, в индукции (и протекании) эритроидной дифференцировки клеток участвует каспаза-3, поскольку добавление ингибитора фермента приводит к снижению концентрации гемоглобина в опухолевых клетках. Спектр обнаруживаемых точечных мутаций в гене р53 довольно разнообразен, чаще всего поражаются кодоны 175, 248, 273 (Ьеуте й а1., 1994; НоМет й а1., 1994). Различия в спектре мутаций могут быть связаны с особенностями влияния определенных мутантных форм р53 на фенотип опухолевых клеток, поэтому данный факт очень важно учитывать в диагностике генетических вариантов злокачественных новообразований.

Другая проблема современной онкологии - это возникновение множественной лекарственной устойчивости опухолевых клеток к антинеопластическим препаратам, сопровождающееся амплификацией тс1г-генов и являющееся одной из основных причин неэффективной химиотерапии рака, поскольку дифференциров-ка и уровень фрагментации ДНК различаются в исходных опухолевых клетках и клетках, сохранившихся после воздействия цитостатика. В популяции клеток при действии цитостатика могут появиться редкие генетические варианты резистентных клеток, которые впоследствии размножаются, в том случае, если получают селективное преимущество.

В современной химиотерапии обычно используют комбинации лекарственных веществ, принадлежащих к разным классам и действующих на различные клеточные мишени. Поэтому МЛУ в данном случае характеризуют как устойчивость клетки не к одному, а к нескольким (или многим) препаратам с разным механизмом действия и разного химического строения. МЛУ нередко связана не только с действием препарата на клетку, но и с типом дифференцировки опухолевых клеток, с их локализацией в организме. Показано, например, что хромосомная транслокация ? (9 ; 22) и связанное с ней появление гибридного белка ВСЯ/АВЬ, определяющие хронический миелолейкоз, приводят к отмене апоптоза и, следовательно, могут определять МЛУ (Не1з1егкатр с1 а1., 1985; Ставровская, 2000).

Нами были получены и охарактеризованы по способности подвергаться индуцированным дифференцировке и/или апоптозу под действием структурно-различных химических реагентов клетки К562, резистентные к 2-И()0 и 4-Ы()0. Показано, что в клетках сублиний процессы эритроидной и миелоидной дифференцировки протекают более интенсивно по сравнению с клетками родительской линии. Однако к апоптоз-индуцирующему действию используемых соединений клетки К562!4-Ы()0 и К562!2-0()0 были резистентны. Чувствительность опухолевых клеток к терапии в большей степени зависит от того, что принято называть «клеточным контекстом» (сочетание особенностей регуляции жизненно важных процессов клетки, связанных с ее видовой, тканевой принадлежностью, а также с теми генетическими изменениями, которые произошли в клетке в ходе ее малигни-зации и прогрессии новообразования). Данный феномен также осложняет диагностику причин и изучение МЛУ опухолей (Меликсетян и др., 1997; Ставровская, 2000). В результате, при планировании методов, выявляющих резистентность, важно учитывать тот факт, что чаще всего устойчивость клеток к конкретному веществу определяется несколькими причинами.

Поэтому, успешное использование в современных исследованиях методов молекулярно-генетического анализа, в том числе ПЦР-анализа, позволит в дальнейшем более полно охарактеризовать протекание многих процессов жизнедеятельности опухолевой клетки, будет способствовать обогащению не только теоретической базы знаний о неоплазмах, но и эффективной диагностике злокачественных опухолей, поиску новых оптимальных схем антиопухолевой химиотерапии.
