Фундаментальная проблема в изучении регенерации — понять механизмы восстановительных процессов. Эффективный путь изучения механизмов регенерации — выявить и проанализировать структуру и функцию генов, контролирующих разные этапы регенерации органов и тканей у животных, обладающих ярко выраженными регенерационными потенциями. Анализ экспрессии генов при регенерации стал возможным в результате внедрения методов молекулярной биологии в исследования регенерации (Миташов и др., 1995).

За полувековой период исследований накоплен большой экспериментальный материал о процессах регенерации у позвоночных животных на клеточном уровне (Карлсон, 1986; Stocum, 1995; Tsonis, 1996; Gerandie, Ferretti, 1998). В лаборатории В.И. Миташова разработаны методы и приемы работы с уникальной моделью регенерации тканей глаза у взрослого тритона. Хрусталик и сетчатка регенерирует в результате трансдифференцировки клеток радужки и пигментного эпителия соответственно. В восстановлении сетчатки участвуют также клетки периферической цилиарной зоны, в которой локализованы мультипотентные клетки, обеспечивающие рост сетчатки в онтогенезе (Stone, 1950; Hasegawa, 1958; Миташов, 1968). Первые результаты по изучению экспрессии генов при регенерации хрусталика были опубликованы в 1992 г. (Mitashov е* а/., 1992). В дальнейшем это новое направление исследований регенерации получило развитие. Появились результаты исследований экспрессии генов при регенерации планарий, конечности, хвоста.

Открытие гомеобоксных генов, экспрессирующихся в процессе развития органов и тканей у всех изученных групп житвотных от дрозофилы до позвоночных послужило новой отправной точкой в современной; биологии и приблизило исследователей к анализу молекулярных механизмов развития. Показано, что существует каскад регуляторных генов, которые контролируют развитие глаза (Zhang et al., 2002а; Zuber et al., 2003). Первые представления о наличие таких генов были получены из исследований развития глаза дрозофилы (Bonini et al.,1997; Chen et al., 1999; Ohto et al., 1999). Позднее гомологи этих генов были идентифицированы в развивающемся глазу позвоночных животных. Гомеобоксные гены являются ядерными транскрипционными факторами, контролирующими экспрессию других генов. Анализ структуры этих генов показал, что в их состав входят консервативные домены, имеющие определённые функции. Каждое семейство гомеобоксных генов имеет специфический для каждого семейства консервативный мотив, который отвечает за белок белковые взаимодействия. Общие свойства гомеобоксных генов заложены в структуре особой консервативной последовательности — гомеодомена, который предназначен для взаимодействий с ДНК. Природа отдает предпочтение использованию одних и тех же путей для контроля специфической дифференцировки в ходе онтогенеза у различных групп животных.

Исследования экспрессии генов при регенерации хрусталика и сетчатки ещё находятся на начальной стадии. В процессе регенерации хрусталика у взрослых тритонов и сетчатки у взрослых рыб выявлена экспрессия гомеобокссодержащих генов Proxl и Рахб (Tsonis, Rio-Tsonis, 2004). Молекулярно-генетические механизмы восстановительных процессов сетчатки менее изучены. Приблизиться к пониманию механизмов регенерации сетчатки мы сможем, если идентифицируем факторы, стимулирующие трансдифференцировку клеток пигментного эпителия сетчатки, идентифицируем клетки, продуцирующие регуляторные морфогенетические факторы и их рецепторы; идентифицируем гены, контролирующие как последовательные стадии регенерации, так и морфогенез глаза.

В основе развития и регенерации лежат сходные процессы клеточной дифференцировки. Поэтому логично предположить, что в регенерации участвуют гомологичные гены, что и при развитии хрусталика и сетчатки. Модель регенерирующего глаза тритонов является уникальной моделью для исследования экспрессии генов, о которых известно, что они участвуют в развитии нервной системы и структур глаза. Сравнительный анализ спектра регуляторных генов, экспрессирующихся в пигментном эпителии, сетчатке и в регенератах сетчатки, позволит осветить молекулярные аспекты процесса регенерации.

Цель настоящей диссертационной работы состояла в идентификации регуляторных генов и локализации первичных продуктов их экспрессии в регенерирующих структурах глаза тритона (Pleurodeles waltl) на последовательных стадиях восстановительного процесса. В данной работе предпринимается попытка понять, что меняется на уровне экспрессии. генов при регенерациии. Мы сосредоточили наше внимание на генах из следующих семейств: Pax, Six, Prox и конкретно генах Рахб, Six3, Proxl. Этот выбор неслучаен, поскольку об этих генах известно, что они являются главными генами, которые определяют морфогенез глаза.

При выполнении этой работы использовался широкий спектр молекулярно-генетических методов: идентификации гомологичных последовательностей, клонирорвание ПЦР-продуктов, секвенирование, сравнительный ПЦР-анализ и in situ гибридизация.

Основные задачи нашего исследования заключались в следующем:

1) Разработка и адаптация молекулярно-биологических подходов к анализу экспрессии генов на модели регенерации хрусталика и сетчатки у представителей низших позвоночных животных — тритонов.

2) Идентификация генов, принадлежащих семействам Pax, Six, Prox (методом ПЦР с последующим секвенированием).

3) Сравнение экспрессии исследуемых генов в интактной сетчатке и на разных стадиях регенерации (методом ПЦР на отнормированных кДНК библиотеках).

4) Локализация экспрессии генов (Рахб, Six3, Proxl) в интактной сетчатке и на разных стадиях регенерации сетчатки и хрусталика (методом in situ гибридизации).

Выводы

1) С помощью молекулярно-генетических подходов впервые идентифицированы регуляторные гомеобоксные гены - Рахб, Proxl и Six3 в нативной и регенерирующей сетчатке тритона Pleurodeles waltl.

2) Сравнительный ПЦР-анализ уровня экспрессии генов Рахб, Proxl и Six3 в нативной и регенерирующей сетчатке тритона показал, что уровень экспрессии генов Рахб и Proxl увеличивается, а - гена Six3 уменьшается в ходе регенерации сетчатки.

3) Впервые методом in situ гибридизации локализована мРНК генов Рахб, Proxl и Six3 в нативной и в регенерирующей сетчатке тритонов: показана экспрессия исследуемых генов во всех слоях нативной и регенерирующей сетчатки, за исключением слоя фоторецепторов, наружного и внутреннего сетчатого слоя.

4) Экспрессия исследуемых генов Рахб, Proxl и Six3 обнаружена в эпителиальных клетках нативного и регенерирующего хрусталика.

5) При анализе результатов in situ гибридизации обнаружен градиент экспрессии: возрастание уровня экспрессии от центра сетчатки к периферической цилиарной зоне, где локализованы мультипотентные клетки.

6) Впервые показан высокий уровень экспрессии гена Proxl в периферической цилиарной зоне.

7) На основании анализа результатов исследования сделано заключение о двойной роли регуляторных генов Рахб, Proxl, Six3, контролирующих как пролиферацию клеток на ранних стадиях регенерации сетчатки, так и дифференцировку ганглиозных, амакриновых и горизонтальных клеток в восстановленной сетчатке.

4.8. Заключение

Анализ литературных данных и результатов собственных исследований позволил нам предположить, что последовательные стадии регенерации структур глаза контролируются линейными и перекрестными взаимодействиями сети регуляторных генов, ключевыми компонентами которой являются гены Рахб, Proxl и Six3. Характер взаимодействия и соответственно выполняемые генами Рахб, Proxl и Six3 функции в ходе регенерации сетчатки и хрусталика могут изменяться под воздействием неких индуцирующих и ингибирующих сигналов (являющимися продуктами других генов), продуцируемых пролиферирующими и дифференцированными клетками. В ходе развития и регенерации глаз принимают участие значительное количество гомеобоксных генов. Между ними обнаруживаются нелинейные взаимодействия, более того некоторые гены являются самоактиваторами. В настоящее время полагают, что работа гомеобоксных генов организована в виде иерархической сети (иерархической в том смысле, что не все ее участники равнозначны). Нужно признать, что полное понимание этой проблемы в данный момент отсутствует, в наших силах только выяснять роль отдельных элементов, в надежде из отдельных элементов впоследствии построить целостную систему.

Источником регенерации сетчатки глаза у взрослых тритонов служат две группы клеток: цилиарная зона и пигментный эпителий. В ходе наших исследований показано, что изучаемые нами гены Рахб, Proxl и Six3 участвуют в процессах регенерации. Вместе с тем остаётся открытым вопрос относительно того, что изучаемые гены запускают процесс регенерации. Ответить на этот вопрос в рамках данной диссертационной работы нельзя, нами только показана экспрессия этих генов в областях ответственных за регенерацию. Способность клеток цилиарной зоны обеспечивать рост сетчатки глаза в течение всей жизни организма является уникальным явлением. Более того, вместе с ростом сетчатки растёт сам глаз и весь организм, и как следствие такие животные не имеют строго определённой продолжительности жизни. Птицы и млекопитающие лишены подобной способности — расти в течение всей жизни. В этом случае допустима более широкая постановка вопроса, - какие гены (или взаимодействия каких генов) определяют способность организма расти в течение всей жизни. Способность организма расти всю жизнь, позволяет ему жить неопределённо долго. Такая постановка вопроса переводит решение проблемы регенерации из локальных задач в глобальную проблему, о решение которой человечество мечтает очень давно. Однако не все животные, обладающие цилиарной ростовой зоной, обладают способностью к полной регенерации сетчатки во взрослом состоянии. Так, например, рыбы и лягушки способны к восстановлению сетчатки только на ранних стадиях развития и источником регенерации служат клетки цилиарной зоны. Во взрослом состоянии их количества не хватает для полноценной регенерации, требуется обязательное участие пигментного эпителия. Следовательно, можно сделать очень важный вывод — трансдифференцировка пигментного эпителия и пролиферация клеток цилиарной зоны являются независимыми событиями и, возможно, контролируются различными механизмами.
