Актуальность работы. Возникновение и эволюция патогенности микроорганизмов является одной из фундаментальных проблем современных микробиологии и медицины. Исследование молекулярных основ этого процесса - ключевой подход к решению данной проблемы. В настоящее время известно, что патогенные бактерии вырабатывают вещества, как непосредственно повреждающие или убивающие клетки и структуры макроорганизма, так и способствующие их проникновению в организм, преодолевая его защитные системы. Эти вещества играют главную роль в развитии заболеваний, вызываемых бактериями (McDonel et al., 1986).

Среди многообразия таких веществ немаловажная роль в патогенезе принадлежит цитолитическим токсинам. Характерными примерами, подтверждающими этот тезис, являются такие цитолизины, как фосфолипаза С Clostridium perfringens - основной фактор, ответственный за развитие газовой гангрены (Stevens et al., 1988), тиол-активируемый токсин Listeria monocytogenes, нарушение продукции которого приводит к потере вирулентности (Cossart et al., 1989), альфа-токсин Staphylococcus aureus, ответственный за целый ряд клинических эффектов при стафилококковых инфекциях (Freer, 1988; Дерябин, 2000).

Не секрет, что патогенные свойства того или иного штамма в значительной мере определяются как спектром его структурных генов, кодирующих факторы патогенности, так и механизмами, обеспечивающими регуляцию этих генов. Отсюда возрастающее внимание медиков и биологов к системам, контролирующим экспрессию генов токсинов. На сегодняшний день описаны сложные регуляторные системы для многих микроорганизмов, например, для С. perfringens (Rood and Lyristis, 1995; Ba-Thein et al., 1996), S. aureus (Heinrichs J.H. et al., 1996; Deora et al., 1997; Chien Y. et al., 1998), L. monocytogenes (Sheehan et al., 1994; Huillet et al.,1999), Vibrio cholerae (DiRita, 1995; Pfau and Taylor, 1998) и ряда других распространенных патогенов.

Интерес к исследованиям цитолитических токсинов не ограничивается лишь медицинской сферой. Изучение этих белков стимулируется и тем, что цитолизины стали ценными инструментами при исследовании клеточной физиологии и функционирования клеточных мембран (Ahnert-Hilger and Weller, 1998; Hucho, 1995), а также являются превосходными модельными объектами для выяснения связи между структурой и функцией белка (Bayley, 1994; Gouaux, 1998).

Таким образом, изучение цитолитических токсинов и их генетических детерминант, включающее в себя клонирование, идентификацию, молекулярный анализ, гетерологичную экспрессию генов цитолизинов, выяснение путей регуляции их экспрессии, а также изучение механизмов действия этих токсинов представляется одним из наиболее перспективных направлений в исследовании молекулярных основ патогенности и вирулентности микроорганизмов.

Важной практической задачей в рамках данной проблемы является удачный выбор объекта исследований. Учитывая тот факт, что большинство бактерий, продуцирующих цитолитические токсины, принадлежат к числу опасных патогенов, удобно использовать в подобных исследованиях микроорганизмы, вырабатывающие цитолизины, но не являющиеся строго патогенными. С этой точки зрения внимание исследователей в течение многих десятилетий привлекает антракоидная группа микроорганизмов рода Bacillus, представители которой широко распространены в природе и имеют ярко выраженные филогенетическое родство и морфологическое и физиолого-биохимическое сходство на фоне широкого диапазона их патогенных свойств. Так, В. mycoides является сапрофитным микроорганизмом, В. thuringiensis - патогенным для насекомых. В. cereus относится к условно-патогенным для теплокровных. В. anthracis печально известен как опасный патоген, вызывающий сибирскую язву. Эти микроорганизмы продуцируют ряд цитолитических белков, рассматриваемых в качестве потенциальных факторов вирулентности. В. cereus, занимая по патогенному статусу промежуточное положение среди представителей данной группы бактерий и продуцируя широкий спектр цитолизинов, может служить прекрасной моделью для изучения этих токсинов.

Еще одной причиной, которая побуждает к активному изучению токсинов, вырабатываемых В. cereus (и его ближайшим родственником В. thuringiensis), является огромная значимость этих микроорганизмов для человека. Сейчас остро встает вопрос экологически чистого производства сельскохозяйственной продукции. При этом важная роль отводится применению микробиологических средств защиты растений, среди которых серьезное место занимают энтомопатогенные бактериальные препараты. Внедрение интенсивных технологий возделывания сельскохозяйственных культур неизбежно влечет расширение применения микробиологических средств защиты растений. Для производства инсектицидных препаратов в настоящее время широко используется В. thuringiensis, приводя к широкомасштабному внесению этого микроорганизма в окружающую среду. Другой важной проблемой является бактериологическое загрязнение производимых промышленностью продуктов питания, лекарственных и косметических препаратов. При этом В. cereus - один из основных бактериальных загрязнителей. В связи с вышеизложенным встает задача определения степени безопасности таких микроорганизмов, как В. cereus и В. thuringiensis, для окружающей среды, животных и человека. В рамках данной задачи изучение отдельных токсинов, продуцируемых этими бактериями, является необходимым звеном.

Хорошо известно, что В. cereus производит ряд внеклеточных токсинов, обладающих гемолитической активностью и рассматриваемых в качестве потенциальных факторов патогенности (Turnbull, 1986; Drobniewski, 1993). На настоящий момент в разной степени описаны цереолизин, сфингомиелиназа, цереолизин АВ, гемолизин BL, цереолизин-подобный гемолизин, гемолизины II, III и CytK. Спектр и свойства гемолитических белков, продуцируемых данным микроорганизмом, изучены далеко не полно. Это обусловлено сложностью получения индивидуальных препаратов данных белков из исходного микроорганизма. В связи с этим, природа одного из них - гемолизина II - до последнего времени оставалась под вопросом. Гемолитическая активность, позднее приписанная ему, была впервые обнаружена в культуральной жидкости одного из штаммов В. cereus еще в 1963 году (Fossum, 1963). Однако до сих пор не существовало данных, свидетельствующих о том, что гемолизин II является продуктом самостоятельного гена, а не произволен от других, уже хорошо описанных гемолитических белков. Активность, приписываемая гемолизину II, считалась обусловленной совместным действием сфингомиелиназы и фосфолипазы С или же она связывалась с проявлением других известных гемолитических факторов В. cereus. Таким образом, вопрос о самом существовании гемолизина II оставался открытым. Для выяснения подобных вопросов весьма эффективным является подход, основанный на клонировании отдельных генов, кодирующих цитолизины, и изучении данных цитолизинов в гетерологичных системах.

Несмотря на то, что гемолитические токсины В. cereus активно изучаются, на сегодняшний день очень мало известно об их генетической регуляции, так же как и о регуляции других факторов патогенности этого микроорганизма. Лишь недавно был обнаружен первый плейотропный регулятор, PlcR, который является активатором генов фосфолипаз, негемолитического и гемолитического энтеротоксинов, а также — целого ряда других генов В. cereus и В. thuringiensis (Lereclus et al., 1996; Agaisse, 1999; Gohar et al., 2003). Учитывая, что вопрос о регуляции генов токсинов неразрывно связан с регуляцией патогенных свойств микроорганизмов, а также ввиду малой изученности данного вопроса для В. cereus, весьма актуальным является поиск и изучение других локусов в геноме этой бактерии, контролирующих экспрессию генов токсинов.

Цели и задачи исследования. Данная работа выполнялась в рамках исследовательского направления, имеющего целью обнаружение и идентификацию генов цитолизинов В.cereus, их структурно-функциональный анализ и изучение функции данных цитолизинов. Особый интерес представляло клонирование из генома В.cereus гемолитических детерминант, не тождественных ранее клонированным. Целью настоящей работы являлось клонирование генетической детерминанты гемолизина II, определение ее первичной структуры, функциональной активности в гетерологичной системе и распространение среди бацилл цереусной группы, описание особенностей регуляции экспрессии гена гемолизина II.

Для достижения указанной цели последовательно ставились следующие задачи:

1. Создать библиотеку генов B.cereus и отобрать гемолитические клоны.

2. Идентифицировать генетическую детерминанту гемолизина II.

3. Определить нуклеотидную последовательность клонированной генетической детерминанты B.cereus и провести ее анализ. На основании анализа выведенной аминокислотной последовательности сделать предварительное заключение о механизме действия гемолизина II.

4. Исследовать особенности активности гемолизина II в гетерологичных системах (E.coli и B.subtilis).

5. Провести скрининг коллекции штаммов бацилл цереусной группы на наличие последовательностей ДНК, гомологичных области, содержащей ген гемолизина II, и на продукцию подобного гемолизина.

6. Определить роль нового гена hlyllR в регуляции экспрессии hlyll и характер (особенности) этой регуляции.

Данная работа выполнена в лаборатории генной систематики и ВНТК генной активности Института биохимии и физиологии микроорганизмов имени Г.К. Скрябина РАН.

Научная новизна работы. Результаты, полученные в ходе данных исследований, носят приоритетный характер. Впервые клонирован ген гемолизина II Bacillus cereus. Тем самым положен конец многолетней дискуссии о существовании этого гемолизина и доказано, что он является самостоятельным белком В. cereus, а не произволен от других гемолитических факторов этого микроорганизма. Впервые определена нуклеотидная последовательность гена гемолизина II и получены данные, позволяющие причислить этот белок к группе (3-складчатых олигомерных порообразующих цитолизинов. Впервые показано, что способность В. cereus продуцировать гемолизин II является штаммоспецифичным свойством по причине ограниченного распространения его гена среди штаммов этого вида. Впервые установлено, что многие штаммы В. thuringiensis производят гемолизин, гомологичный гемолизину II В. cereus и сходный с ним по свойствам. Впервые клонирован ген этого гемолизина В. thuringiensis. Обнаружен и клонирован новый регуляторный ген В. cereus и показана его функция как регулятора транскрипции гена гемолизина II.

Практическое значение работы. Полученные результаты вносят вклад в понимание феномена гемолитического фенотипа и молекулярных основ вирулентности В. cereus и близких видов микроорганизмов. Эти данные являются необходимым первоначальным этапом, закладывающим основу для дальнейших исследований гемолизина II: выяснения его участия в патогенезе, изучения его структурно-функциональных особенностей, что позволит в перспективе использовать этот объект при исследовании клеточной физиологии и функционирования мембран клеток. Тот факт, что гемолизин II обнаружен у В. thuringiensis, широко применяемого как биологический инсектицид, заставляет усилить внимание к тщательному анализу и отбору используемых штаммов на предмет их безопасности для человека и животных и заостряет вопрос о правомочности использования данного микроорганизма в качестве инсектицидного препарата. Обнаружение нового регуляторного локуса В. cereus, контролирующего транскрипцию гена гемолизина II, является шагом к выяснению сложных механизмов скоординированной регуляции генов, обеспечивающих вирулентность этого микроорганизма, что в перспективе позволит осуществлять контроль над его патогенными свойствами.

1. Обнаружен и впервые клонирован ген гемолизина II Bacillus cereus, определена его нуклеотидная последовательность и на основании анализа аминокислотной последовательности, выведенной из первичной структуры его гена, гемолизин II причислен к семейству р-складчатых порообразующих олигомерных цитолизинов.

2. Установлено, что способность продуцировать гемолизин II является штаммоспецифичным свойством В.cereus и В.thuringiensis.

3. Идентифицирован и клонирован новый ген В.cereus - hlyllR. Установлено, что экспрессия гена гемолизина II В. cereus контролируется белковым продуктом hlyllR, который взаимодействует с операторным участком, лежащим перед геном гемолизина

4. Показано, что HlyllR способен выступать как в роли позитивного, так и негативного регулятора экспрессии гена гемолизина II.
