Нейрональные кальциевые сенсоры (NCS, neuronal calcium senI sors) образуют семейство EF-hand-содержащих Ca -связывающих белков, как правило, высокоспецифичных для нервных клеток. Эти белки выполняют регуляторную роль в отношении различных внутриклеточных белков-мишеней таких, как: протеинкиназы, гуанилат-циклазы, аденилатциклазы и ионные каналы. Типичным представителем семейства NCS является рековерин, высокоспецифичный для I нейронов сетчатки. Рековерин участвует в Са -зависимой регуляции зрительной трансдукции, являясь Са2+-сенсором родопсинкиназы -фермента, десенситизирующего фотоактивированный родопсин путём его фосфорилирования. Общим свойством представителей семейства NCS является наличие в их структуре четырех потенциальных Са -связывающих центров типа EF-hand. При этом в большинстве случаев Са2+-связывающий центр EF1 не способен координировать катионы, поскольку нарушена его каноническая последовательность. Молекула рековерина, помимо нефункционального центра EF1, содержит в сво

2+ ей структуре Са -связывающий центр EF4, который также не способен связывать ионы кальция: таким образом, в его молекуле только два Са2+-связывающих центра, а именно EF2 и EF3, координируют ка

2+ тион. Наличие активного четвертого Са -связывающего центра в структуре молекулы белка характерно почти для всех представителей семейства NCS, что свидетельствует о необходимости центра EF4 для нормального функционирования белков этого семейства. Однако, поскольку ранее не было продемонстрировано влияние активного четвертого Са2+-связывающего центра молекулы рековерина на его функциональные свойства, мы попытались ответить на этот вопрос в настоящей работе, используя мутантные формы белка с «восстановленной» способностью координировать катион центром EF4.

1. Исследованы основные функциональные свойства мутантных форм рековерина с заменами в его Са2+-связывающих центрах:

- «EF+4» с заменами G160D, К161Е, K162N, D165G и K166Q в 4-м Са -связывающем центре, приводящими к появлению у этого центра способности связывать ионы кальция;

- «EF-2+4» с заменой E85Q во 2-м Са2+-связывающем центре, приводящей к потере у этого центра способности связывать ио

2+ ны кальция, и вышеуказанными заменами в 4-м Са -связывающем центре;

- «EF-3+4» с заменой E121Q во 3-м Са -связывающем центре, приводящей к потере у этого центра способности связывать ионы кальция, и вышеуказанными заменами в 4-м Са -связывающем центре.

2. Реконструкция Са -связывающего центра EF4 в рековерине не влияет на заполнение катионами его Са2+-связывающих центров EF2 и EF3.

Связывание ионов кальция в одном из Са -связывающих центров, локализованных в С-концевом домене рековерина, необходимо и достаточно для функционирования его Са -миристоильного переключателя и, соответственно, для взаимодействия рековерина с фоторецепторными мембранами и ингибирования родопсинкиназы.

5. Реконструкция 4-го Са -связывающего центра в рековерине, в ко

2+ тором 3-й Са -связывающий центр EF3 модифицирован и не способен связывать ионы кальция, функционально компенсирует отсутствие связывания ионов кальция в Са2+-связывающем центре EF3.

6. Методом поверхностного плазмон-резонанса показано, что 2-й Са2+-связывающий центр рековерина контролирует время нахождения белка на поверхности липидного бислоя.
