Открытие феномена прионов позволило обнаружить существование нового типа наследственности, на основе белка. Термин "прион" появился в связи с исследованием ряда нейродегенеративных заболеваний с неизвестной этиологией, таких как скрэйпи овец и болезнь Крейцфельда-Якоба у человека. Несмотря на то, что эти болезни известны довольно давно, природа их оставалась загадкой достаточно долго. Объяснение столь медленного хода исследований заключалось в необычном способе возникновения этих болезней: они могут возникать спонтанно и в то же время могут наследоваться, а также передаваться инфекционным путем.Долгое время комбинация этих трех свойств казалась необъяснимой. В 1967 была предложена так называемая "белковая" гипотеза, согласно которой инфекционный агент, вызывающий эти заболевания, позже получивший название "прион", представляет собой обычный клеточный белок, принявший особое конформационное состояние. Это состояние способно самоподдерживаться за счет автокаталитического механизма. На сегодняшний день эту гипотезу можно считать доказанной. Ее подтверждают практически все экспериментальные данные, а также тот факт, что до сих пор не удалось найти какую-либо нуклеиновую кислоту, связанную с этими инфекционными заболеваниями.За последние десять лет появились работы, показывающие существование прионоподобных белков у дрожжей Saccharomyces cerevisiae и гриба Podospora anserina. Если у млекопитающих существование прионов связано с патологией, то у низших эукариот наследуемое переключение активности белков, вызванное их переходом в прионное состояние, может иметь адаптивное значение. У дрожжей прионоподобные свойства белков лежат в основе механизмов эпигенетической наследственности и регуляции экспрессии генов на посттрансляционном уровне.Изучение прионов у дрожжей открывает новые перспективы в понимании сущности этого явления в целом. Феномен прионов относится к типу, так называемых, амилоидных белков, образующих фибриллярные агрегаты со специфической белковой укладкой. Похоже, что феномен прионов и амилоидов гораздо шире распространен в природе и играет существенно более важную роль у различных организмов, чем это предполагается на сегодняшний день. Поиск в банках генов выявляет значительное количество белков у различных организмов, структурно сходных с уже известными прионными белками у дрожжей. Это позволяет полагать, что белки, которые могут изменять свою конформацию и полимеризоваться, широко распространены и могут играть важную роль во внутриклеточной регуляции. Кроме того, до сих пор остается открытой проблема диагностики и лечения прионных болезней у человека и животных. Особое значение данная проблема получила в силу появившихся в последнее время сообщений о возможности передачи этих заболеваний от животных к человеку (при употреблении больных животных в пищу). Также, сходство свойств дрожжевых прионов и амилоидов млекопитающих выводит актуальность исследований в этой области на передний план.

6. выводы

1. Показано, что прионные полимеры белка Sup35 отличаются от прочих клеточных макромолекулярных комплексов нерастворимостью в SDS при 37°С, что характерно для амилоидных фибрилл Sup35, образующихся in vitro.

2. Разработан метод полуденатурирующего электрофореза в агарозном геле для определения размера прионных полимеров.

3. Показано, что прионные агрегаты Sup35 представляют собой комплексы, состоящие из прионных полимеров. Полимеры гетерогенны по размеру и содержат от 10 до 50 мономеров Sup35.

4. Гидрохлорид гуанидина полностью блокирует фрагментацию прионных полимеров Sup35 в клетках дрожжей. Подтверждено предположение о фрагментации прионных полимеров шапероном Hsp 104.

5. Обнаружено, что химерные белки Q70-Sup35MC, Q85-Sup35MC и QY76-Sup35MC способны к полимеризации in vivo. Внедрение остатков тирозина в полиглутаминовые домены улучшает фрагментацию образуемых ими полимеров.

6. Метод полуденатурирующего электрофореза может быть использован для выявления амилоидного состояния разнообразных белков. Способность образовывать полимеры амилоидного типа in vivo показана для дрожжевых белков Rnql и РиЫ, химерного Sup35, содержащего N-концевой домен Sup35 Pichia methanolica, а также для химерного белка, содержащего полиглутаминовую последовательность хантингтина человека.

5.9 ЗАКЛЮЧЕНИЕ

Разработанный в настоящей работе электрофоретический подход позволил выявить двухуровневую организацию прионных частиц белка Sup35 в клетках дрожжей. С использованием данного метода были получены убедительные доказательства способности шаперона Hsp 104 фрагментировать прионные полимеры Sup35. Продемонстрировано, что метод может быть использован для исследования различных прионо- и амилоидоподобных белков как в клетках дрожжей, так, по-видимому, и в клетках других организмов, например, млекопитающих. Можно полагать, что разработанный нами метод может быть в дальнейшем использован для выявления новых белков, проявляющих прионогенные или амилоидогенные свойства in vivo, а также для выявления воздействий ингибирующих амилоидогенез, но не влияющих на основные параметры физиологии клетки.
