Экспрессия гена регулируется при помощи различных цис- и транс-регуляторных элементов. К цис-регуляторным элементам относятся последовательности ДНК, непосредственно прилежащие к генам, так называемые промоторы, и дистально расположенные энхансеры и сайленсеры. Большинство энхансеров не обладает специфичностью по отношению к промоторам, следовательно должна существовать система, ограничивающая активность энхансера только «правильным» промотором. Предполагается, что в функционировании данной системы значительную роль могут играть инсуляторы. Инсулятор представляет собой последовательность ДНК, которая, находясь между энхансером и промотором, ограничивает их взаимодействие. Таким образом, инсуляторы выполняют важную функцию регуляции транскрипции, определяя какой энхансер должен активировать данный промотор.

На настоящий момент механизм действия инсуляторов полностью не изучен. Существует несколько моделей их функционирования, каждая из которых имеет свои преимущества и недостатки, но практически все они предполагают связь работы инсулятора с ядерной периферией и белками ядерного матрикса. К сожалению, до сих пор не выявлены конкретные белки, входящие в эти ядерные структуры и одновременно влияющие на работу инсулятора. Это направление исследований представляет большой интерес, так как рассматривает инсуляцию как составную часть глобального процесса регуляции транскрипциии в ядре в целом. Выяснение функциональных связей между процессом инсуляции и белками ядерного матрикса позволит понять роль инсуляторов в образовании различных областей ядра: с повышенной и пониженной активностью транскрипции.

В представленной работе было продолжено изучение широко известного Su(Hw)зависимого инсулятора, впервые обнаруженного в последовательности ретротранспозона

МДГ4 у Drosophila melanogaster . Известно, что инсулятор Su(Hw) способен блокировать более 30 известных энхансеров, работающих в разных тканях и на разных этапах развития. Белковый комплекс инсулятора Su(Hw) состоит из трех известных на данный момент белков: связывающегося с ДНК белка Su(Hw), белка Mod(mdg4) и недавно открытого белка CP 190. Мутация в гене mod(mdg4) нарушает связывание кодируемого им белка Mod(mdg4)67.2 с инсуляторным комплексом. Отсутствие белка Mod(mdg4)67.2 приводит к нарушению инсуляции в различных модельных системах. Недавно в нашей лаборатории была обнаружена группа генов su(mg) {suppressor of mod(mdg4)), мутации в которых приводили к восстановлению 8и(Нш)-зависимой инсуляции даже при отсутствии белка Mod(mdg4)67.2. Один из клонированных генов группы su(mg) кодирует белок EAST, который, как предполагается, является компонентом ядерного матрикса. Изучение свойств и функций этого белка позволит более детально понять связь между инсуляцией, ядерным матриксом и транскрипцией.

Цель и задачи исследования

Основной целью данной работы является изучение структуры и функций гена из семейства su(mg), который кодирует белок EAST.

В работе были поставлены следующие задачи:

1. Изучить генетические взаимодействия между геном east и генами, кодирующими белки, входящие в состав инсуляторного комплекса Su(Hw).

2. Найти в составе ретортранспозона МДГ4 последовательность, отвечающую за проявление эффектов гена east.

3. Доказать существование молекулярных взаимодействий между белками EAST, СР60 и белками инсуляторного комплекса.

5. Выводы

1. С помощью различных генетических модельных систем, как нативных, так и искусственно созданных, впервые была показана функциональная взаимосвязь между инсуляторными и репрессионными свойствами инсулятора Su(Hw) и уровнем экспрессии генов east и србО. Было показано, что изменение уровня экспрессии генов, кодирующих входящие в инсуляторный комплекс белки, в сочетании с мутациями в генах east и србО, также меняло свойства инсулятора.

2. Установлено, что для проявления эффектов, связанных с экспрессией гена east, необходимы последовательности инсулятора Su(Hw) и длинного концевого повтора из ретротранспозона МДГ4.

3. Впервые показано прямое взаимодействие между белком СР60 и белком СР190, компонентом инсуляторного комплекса, а также взаимодействие между белком СР60 и белком EAST, предполагаемым компонентом ядерного матрикса. Способность белка СР60 связываться с последовательностью инсулятора Su(Hw) и коиммунопреципитация белка СР60 с основным компонентом инсулятора Su(Hw) -белком Mod(mdg4)67.2 подтверждает, что белок СР60 также является компонентом инсуляторного комплекса.
