ВВЕДЕНИЕ

Изучение структуры вирионов фитовирусов и вирусных белков оболочки, а также возможностей их модификации и структурной перестройки является важной задачей современной молекулярной вирусологии. Структурная модификация частиц вирусов растений может быть не только направлением защиты сельскохозяйственных растений, но также открывает возможности создания новых биотехнологий в медицине и ветеринарии.

Сегодня вирусы можно рассматривать не просто как инфекционные агенты, а как функциональные наночастицы, позволяющие получать новые материалы. Вирусы и вирусоподобные частицы (макромолекулярные структуры белка оболочки вируса, не содержащие инфекционный материал) широко применяют в различных областях биотехнологии: создание кандидатных вакцин, адресная доставка целевых веществ,

биоконтрастирование, микроэлектроника (Yusibov et al, 2006; Lico et al, 2008; Soto and Ratna, 2010; Leong et al, 2010; Liepold et al, 2007; Aljabali et al., 2012; Bruckman et al., 2013). При этом вирусы растений обладают безусловным преимуществом при получении новых функциональных и биологически активных материалов, в частности кандидатных вакцин, так как они не патогенны для млекопитающих, в том числе для человека (Lico et al, 2009). Большинство современных вакцин создаются на основе ослабленных и инактивированных форм инфекционных агентов. Живые вакцины обладают высокой протективной активностью, однако при этом, остается возможность их реверсии к патогенной форме. Инактивированные вакцины значительно безопаснее, но для индукции эффективного иммунного ответа требуется несколько актов ревакцинации (Jennings and Bachman, 2008). Субъединичные вакцины, основанные на индивидуальных пептидах и белках инфекционного агента, более безопасны, но существует проблема их низкой иммуногенности. Связывание антигенов патогенов с высокомолекулярным носителем

(платформой) может существенно повысить иммунный ответ и обеспечить защиту организма от инфекции (Ысо et а1., 2008).

Настоящая работа посвящена изучению структуры и свойств сферических частиц, образующихся при термической перестройке вируса табачной мозаики и его белка оболочки, и исследованию возможности их применения в качестве платформы для презентации биологически активных молекул, в частности антигенов инфекционных агентов человека, животных и растений.

1. Продемонстрировано, что при нагревании вируса табачной мозаики (ВТМ) до 94°С происходит термическая двухступенчатая перестройка белка оболочки (БО) вириона, и образуются биодеградируемые сферические частицы (СЧ) контролируемого размера, свободные от РНК. СЧ могут быть получены при термической денатурации различных форм БО ВТМ, не содержащих РНК вируса.

2. Получены сведения о том, что термическая перестройка БО вириона сопровождается переходом значительной части а-спиральных структур белка в неупорядоченные или ^-структуры и увеличением плотности образующихся частиц по сравнению с вирионами ВТМ.

3. Выявлена высокая стабильность СЧ к воздействию различных внешних факторов.

4. Показано, что СЧ обладают значительно более высокой иммуногенностью по сравнению с нативным вирусом.

5. Продемонстрированы уникальные адсорбционные свойства СЧ. На основе СЧ in vitro созданы антигенно активные комплексы с антигенами патогенов человека, животных и растений.

6. Установлено, что СЧ проявляют свойства эффективного адъюванта и активно стимулируют иммунный ответ на антиген, адсорбированный на их поверхности.

ЗАКЛЮЧЕНИЕ

Показано, что при нагревании спиральных частиц ВТМ до 94°С происходит перестройка вирусного белка оболочки и формируются сферические частицы, контролируемого размера, отдаленно антигенно родственные вирионам ВТМ, обладающие более высокой плотностью, чем вирионы фитовирусов со спиральной структурой, свободные от вирусной РНК и биодеградируемые, в отличие от ВТМ. Обнаружено, что сферические частицы могут быть получены из различных форм БО, не содержащих генетический материал вируса, в том числе из препаратов А-белка и мономерной формы БО. Это позволяет утверждать, что спиральная структура БО в вирионе или вирусоподобных частицах не является определяющей при термической перестройке БО в СЧ. С использованием ряда физико-химических методов получены данные о структурных различиях белка оболочки ВТМ в составе нативного вируса и белка, входящего в состав сферических частиц. Продемонстрировано, что в отличие от нативного БО ВТМ белок, из которого состоят сферические частицы, имеет в своем составе меньшее количество ос-спиралей и большее количество Р-структур. Кроме того, было показано, что СЧ взаимодействуют с тиофлавином Т, что может являться признаком амилоидоподобной структуры сферических частицах.

Продемонстрирована высокая стабильность СЧ к воздействию различных внешних факторов. Сферические частицы не изменяют формы, размеры и степень агрегации, по сравнению с исходными, при: 1. замораживании (-20 °С) и последующем оттаивании; 2. повторном нагревании до 98°С и охлаждении; 3. при осаждении центрифугированием (до 10 ООО g) и ресуспендировании осадка; 4. при хранении в течение года (4°С) в воде. Показано, что СЧ обладают более высокой иммуногенностью по сравнению с нативными вирионами ВТМ. Можно утверждать, что СЧ уникальны по своим свойствам и структуре и не имеют аналогов среди вирусов, вирусоподобных частиц и других биологических объектов.

Впервые показано, что СЧ обладают исключительными адсорбционными свойствами. На основе СЧ in vitro созданы антигенно и иммуногенно активные комплексы с антигенными детерминантами патогенов человека, животных и растений, отличающихся по аминокислотному составу и молекулярной массе, в том числе с антигенами таких актуальных для медицины инфекционных агентов, как вирус гриппа и вирус краснухи. Получены сведения об определяющей роли электростатических и гидрофобных взаимодействий при образовании комплекса СЧ-антиген. Продемонстрировано, что связанные с поверхностью СЧ чужеродные белки способны реагировать со специфическими антителами и, следовательно, сохраняют свои антигенные свойства в составе полученных комплексов. Установлено, что СЧ проявляют свойства эффективного адъюванта и активно стимулируют иммунный ответ на антиген, адсорбированный на их поверхности. Иммуногенные комплексы, полученные на основе СЧ, могут быть использованы для создания кандидатных вакцин нового типа, которые будут полностью биобезопасными, так как они создаются на основе белковых структур фитовируса, а также для получения дешевых диагностических антител.

На основании опубликованных по данной работе статей, недавно группой Dr. Steinmetz были получены комплексы СЧ-гадолиний, которые обладают значительно более высокой релаксационной эффективностью в сравнении с другими вирусными платформами, модифицированными гадолинием (Gd), что позволяет рассматривать их как перспективную платформу для разработки парамагнитных контрастных веществ для магнитно-резонансной томографии (Bruckman et al., 2013).
