1.1. Актуальность проблемы

Апоптоз — генетически запрограммированная гибель клеток, которая приводит к "аккуратной" разборке повреждённых клеток и их удалению. Очень важно, что при апоптотической гибели клетки не развивается воспалительный процесс. Зачастую возникновение и развитие опухоли связано с нарушением в работе апоптотической системы, поэтому индукцию апоптоза используют как один из подходов в терапии канцерогенеза и ряда других заболеваний.

В клетке одной из систем деградации белков являются протеасомы. Убиквитин-протеасомная система (УПС) осуществляет программированный протеолиз и процессинг различных регуляторных белков, участвующих во множестве клеточных процессов, включая регуляцию транскрипции, репарацию ДНК, продвижение клетки по клеточному циклу, иммунный ответ, апоптоз (Konstantinova et al., 2008; Моисеева и др., 2010; Цимоха, 2010). Убиквитин-зависимому протеолитическому пути деградации подвергаются многие белки-регуляторы апоптоза (Drexler 1998; Wojcik, 2002) включая многие опухолевые супрессоры и онкогенные белки. Поэтому нарушения в УПС, вызывают, в частности, накопление в клетке онкобелков, что приводит к реализации их онкогенного потенциала и, как следствие, к образованию новых очагов опухолевого роста (Naujokat, Hoffmann, 2002; Herrmann et al., 2004).

Согласно современным представлениям, протеасомы в клетке вездесущи: они находятся и в ядре, и в цитоплазме клеток. В последнее годы исследований протеасом в литературе появились данные об их присутствии во внеклеточном пространстве (Lavabre-Bertrand et al., 2001; Stoebner et al., 2005; Sixt et al., 2007, 2009; Sixt, Dahlmann, 2008; Albright et al., 2009; Henry et al., 2009; Sixt, Peters, 2010). С помощью метода электронной микроскопии было показано, что внеклеточные протеасомы имеют структуру, аналогичную внутриклеточным частицам (Zoeger el al., 2006). Биологические функции внеклеточных протеасом до сих пор неясны, однако были выявлены различия в количестве экспортируемых из клеток в плазму протеасом при опухолевой трансформации клеток (Lavabre-Bertrand et al., 2001; Stoebner et al., 2005; Henry et al., 2009). Кроме того, наблюдается увеличение концентрации протеасом во внеклеточном альвеолярном пространстве при дыхательной недостаточности и во время воспалительных процессов в лёгких (Sixt et al., 2007, 2009; Albright et al., 2009). Интересно, что не наблюдалось никакой корреляции между клеточной гибелью и появлением протеасом во внеклеточном пространстве (Sixt et al., 2007). Предполагается, что накопление протеасом в межклеточном пространстве связано, прежде всего, с необходимостью «расчистки территории» — избавление от накапливающихся во внеклеточном пространстве белков и активация секретируемых клеткой белков-предшественников, а также процессинг антигенов (Sixt, Peters, 2010).

В упомянутых выше исследованиях была сделана оценка лишь количества и удельной протеолитической активности внеклеточных протеасом. Известно, однако, что активность и стабильность белка, так же как его функционирование в клетке, определяется, в том числе, его посттрансляционными модификациями (ПТМ), поэтому описывая такой многофункциональный белковый комплекс, как протеасома, крайне важно получить описание паттерна его ПТМ. На сегодняшний день не был сделан анализ паттерна ПТМ протеасомных субъединиц и субъединичного состава внеклеточных протеасом. Не изучались изменения пептидазных активностей протеасом при выходе их из клеток во внеклеточное пространство, а также при индукции апоптотической гибели в этих клетках.

Многие процессы внутри клеток регулируются белками, которые осуществляют свою регуляторную функцию, в том числе за счет специфического связывания с другими белками, которые, как правило, являются специфическими ферментами. В силу этого важно исследование ассоциированных с внеклеточными протеасомами белков, как возможных кандидатов в регуляторы протеасомной секреции во внеклеточное пространство.

В свете вышесказанного весьма актуальным представляется исследование специфических изменений паттерна ПТМ субъединиц и пептидазных активностей внеклеточных протеасом до и после индукции апоптоза в клетках проэритролейкемии человека линии К562 при помощи противоопухолевого препарата доксорубицина (ДР). Кроме того, важно исследовать субъединичный состав внеклеточных протеасом в сравнении с внутриклеточными частицами, получить первичный скрининг белков, ассоциированных с протеасомами.

ВЫВОДЫ

1. При выходе протеасом из клеток во внеклеточное пространство происходит изменение паттерна ПТМ их субъединиц.

2. Вне- и внутриклеточные протеасомы отличаются по пептидазным активностям: внеклеточные протеасомы менее активны по типу трипсина, чем цитоплазматические комплексы.

3. В составе внеклеточных протеасом присутствует регулятор РА200, участвующий в репарации ДНК, количество 198 регуляторных комплексов в сравнении с внутриклеточной популяцией заметно меньше.

4. Воздействие на клетки К562 индуктора апоптоза ДР вызывает специфические изменения паттерна ПТМ субъединиц внеклеточных протеасом, а также приводит к снижению трипсин- и каспаза-подобных типов пептидазной активности внеклеточных протеасом и не изменяет химотрипсиновую активность.

5. Выявлены новые сайты убиквитинилирования и ацетилирования субъединиц протеасом а2 (К196), а4 (К189 и К234), аб (К217) и ЯрпП (А2).
