Липиды являются одним из основных классов биомолекул, интерес к которым обусловлен целым рядом важных обстоятельств. Эти близкие по своей химической природе вещества участвуют в построении биологических мембран всех клеточных систем, обеспечивая универсальность их функционирования. Будучи структурной основой клеточных мембран, липиды определяют степень их жидкостности, латеральную и трансбислойную асимметрию и другие параметры.

In vivo липиды также чрезвычайно интересны. Они участвуют практически во всех физиологических процессах (иммунном ответе, передаче нейрональной информации, регуляции сосудистого и мышечного тонуса, гомеостазе, воспалении и т.д.) [1] и имеют практическое применение. Например, изменение в составе липопротеинов человеческой крови свидетельствует о развитии сосудистых заболеваний и используется для их диагностики. Диацилглицериды выполняют функции молекул-мессенджеров, участвующих в механизмах передачи сигналов [2]. Лизофосфолипиды, концентрация которых в клеточных мембранах при стрессовых ситуациях значительно возрастает, являются важными маркерами некоторых заболеваний, таких как атеросклероз [3] и ревматоидный артрит [4] (Механизм образования лизофосфатидов в разных случаях может иметь различные биохимические сочетания, но всегда должен включать активизацию фосфолипазы Аг, гидролизующей ФЛ [5]. Присутствие лизофосфатидов в мембране делает ее непрочной и увеличивает вероятность нарушения целостности внутренней среды клетки.).

Многогранность функций липидов связана с разнообразием их состава. Липиды подразделяют на полярные и нейтральные. К полярным липидам относятся фос-фолипиды и холестерин, которые являются основными структурными составляющими биологических мембран и во многом определяют свойства последних. Молекулы ФЛ имеют единую структурную организацию, при которой полярная и гидрофобная области оказываются разделенными. Полярная и гидрофобная (углеводородная) части молекулы, как правило, обладают достаточной гибкостью и подвижностью, что позволяет им принимать наиболее выгодную форму, соответствующую минимуму энергии, а жидкому бислою - сохранять барьерные свойства при изменении в широких пределах липидного состава, температуры, ионной силы водной среды и т.д.

Нейтральные липиды являются наиболее доступными источниками энергии, обеспечивающими обменные процессы. Запасы нейтральных липидов могут служить источником структурных элементов - глицерина, ЖК, холестерина, являющихся исходными продуктами при биосинтезе многих других необходимых для жизнедеятельности соединений: углеводов, мембранных липидов, желчных кислот, простаг-ландинов, стероидных гормонов и др.

Функциональные особенности липидов во многом определяются их жирнокис-лотным составом. Существует большое число разнообразных молекул ЖК: нормального строения и разветвленных, с четным и нечетным числом углеродных атомов в алифатической цепи, насыщенные и ненасыщенные, с разным позиционным расположением двойных связей и с различной длиной цепи. ЖК, кроме обеспечения целостности мембран и оптимального уровня ненасыщенности, обусловливают включение и диффузионное перемещение мембранных компонентов, активность мембраносвя-занных ферментов, мембранную проницаемость и транспортные свойства.

Большой прогресс в липидологии произошел после объединения исследований химии и биологии липидов в одно направление, которое получило название «lipidom-ics» (липидомика) [6]. Хотя по количеству статей (около 200) липидомика пока значительно уступает геномике (около 25000) и протеомике (около 13000) [7], интерес к липидам и их анализу в последние 10 лет значительно увеличился [8]. В настоящее время всесторонний анализ липидных молекул, липидомика, в дополнение к геномике и протеомике, является одним из важнейших условий для понимания клеточной физиологии и патологии.

Важность липидных молекул в осуществлении клеточных функций и патологии, их многообразие породили настоятельную потребность в создании хорошо организованной базы данных по липидам и их классификации. В 2005 г. Международный комитет по классификации и номенклатуре липидов предложил ввести в действие Всеобщую систему классификации липидов (Comprehensive Classification System for Lipids) (см. [82]), основанной на четких химических и биохимических принципах, которые учитывают различия в составе и структуре гидрофобных и гидрофильных доменов липидных молекул. Единый способ представления химических структур индивидуальных липидов и их производных, цифровые идентификаторы для каждой уникальной липидной молекулы делают предлагаемую систему совместимой с требованиями информатики [9] и с базами данных других макромолекул. В 2009 г. опубликована усовершенствованная версия этой классификации, более полно представляющая структурное разнообразие липидов и их производных [10].

Многочисленными исследованиями показана важная роль липидов в развитии приспособительных реакций у организмов при экологических модуляциях. К настоящему времени как в отечественной, так и в зарубежной литературе накоплен значительный материал по вопросам, касающимся участия липидов в адаптивных реакциях организмов в ответ на изменение разнообразных условий окружающей среды [11]. Адаптации осуществляются преимущественно за счет модификаций мембранных липидов и углеводородных радикалов их ЖК. Ведущая роль в запуске адаптационных механизмов отводится физическому состоянию мембран, зависящему от степени ненасыщенности ЖК в мембранных липидах.

Наряду с общими закономерностями адаптационных механизмов на уровне липидов должны существовать и специфические реакции для разных видов, позволяющие особи поддерживать жизнедеятельность в разнообразных условиях, которые в полной мере еще не изучены. Бактерии, структурной единицей которых является про-кариотическая клетка, представляют собой обширную группу живых организмов, населяющих нашу планеты. В отличие от животных и растений микроорганизмы чрезвычайно разнообразны по своей физиологии. Структурная организация мембран микроорганизмов, во многом обусловленная составом липидов, которые у бактерий характеризуются чрезвычайно высоким разнообразием [12], и соотношением их с белками, определяет разнообразные функциональные свойства бактериальных клеток, такие как механизмы транспорта, регуляция активности ферментов и метаболических процессов, вирулентность, резистентность и др. Благодаря высокой скорости размножения, бактерии быстро эволюционизируют и в течение короткого времени могут приспособиться к жизни даже в мало пригодных для этого условиях.

До недавнего времени почти весь научный и практический интерес был сосредоточен, что вполне понятно, на изучении микроорганизмов, использующих человека и животных как среду обитания. Существенно меньший интерес вызывала интерес группа микроорганизмов, часто родственных тем, о которых говорилось выше, но обитающих в иных экологических условиях и являющихся сапрофитами. С точки зрения экологии они являются факультативными психрофилами с температурным диапазоном существования от 0 до 30 °С. Многие сапрофитные организмы при изменении условий среды переходят к паразитическому существованию и инфицируют различные виды рыб, теплокровных животных и человека. Остается неясным, как изменяется качественный состав и количественные соотношения липидных компонентов у таких микроорганизмов под влиянием разных факторов окружающей среды и при переходе от одной среды обитания к другой. Поэтому исследование приспособительных реакций на молекулярном и физиологическом уровнях организации у организмов разной таксономической принадлежности, в том числе у микроорганизмов, остается актуальной задачей экологической и эволюционной физиологии и биохимии.

185 ВЫВОДЫ

1. Изучено влияние экзогенных (температура, способ культивирования, фаза роста, источник углерода) и эндогенных (плазмидные ДНК) факторов роста и условий обитания на липидный состав некоторых морских и наземных грамотрицательных бактерий.

2. Показано, что холодовые культуры Yersinia pseudotuberculosis, в отличие от культур, выращенных при 37 °С, имеют более высокое общее содержание фосфоли-пидов и липополисахаридов, более высокий индекс ненасыщенности жирных кислот и более высокое значение отношения ФЭ/ФГ+ДФГ. Липополисахариды «холодовых» вариантов бактерий, в сравнении с липополисахаридами «тепловых» вариантов, имеют более длинные О-специфические углеводные цепи и более высокую степень аци-лирования остатков 3-гидрокситетрадекановой кислоты. Липид А бактерий, выращенных на холоду, имеет, в основном, пентаацильный тип структуры, тепловые культуры бактерий синтезируют тетраацильный липид А. Терморегуляция состава липидов контролируется хромосомными генами и подавляется в присутствии плазмиды pVM82. Плазмида вирулентности не влияет на состав липидных компонентов бактерий псевдотуберкулеза.

3. Показано, что присутствие глюкозы в питательной среде оказывает существенное влияние на состав фосфолипидов Yersinia pseudotuberculosis, характер которого зависит от температуры: на холоду (8 °С) глюкоза стимулирует синтез фосфолипидов, при 37 °С — ингибирует этот процесс.

4. Найдено, что колониальные культуры Yersinia pseudotuberculosis содержат больше общих липидов и фосфолипидов и имеют более высокий индекс ненасыщенности жирных кислот, чем суспензионные. Липополисахариды, выделенные из колониальных культур, в сравнении с липополисахаридами суспензионных культур, имеют более длинные углеводные цепи. При обоих способах культивирования липополисахариды с максимальной длиной цепи О-полисахаридов синтезируются в стационарной фазе роста.

5. Показано, что низкая температура и рост в виде колоний являются факторами, усиливающими патогенный потенциал бактерий псевдотуберкулеза: липополисахариды колониальных культур, выращенных на холоду, в сравнении с липополисахарида-ми «тепловых» вариантов суспензионных культур, обладают более высокой острой токсичностью для теплокровных животных.

6. Показано, что липиды А морских бактерий имеют преимущественно пентаа-цильный тип структуры и часто содержат одну фосфатную группу. Установлена корреляция между хроматографическими (ТСХ) профилями липидов А, составом их жирных кислот, с одной стороны, и таксономическим положением бактерий, с другой.

7. Показано, что липиды А морских бактерий Marinomonas communis АТСС 27118т и Marinomonas vaga АТСС 27119 представляют собой 1-фосфаты р-Г—>6-связанного дисахарида D-глюкозамина, ацилированного остатками редких короткоцепочечных (-Я)-З-Додеканоилоксидекановой (или (-Я)-З-деканоилоксидекановой) и С#)-3-гидроксидекановой кислот в положениях 2 и 3 соответственно и (R)-3-{(R)-3-гидроксидеканоил}оксидекановой кислоты в положении 2'. Липид А бактерий Mari-nomonas mediterranea АТСС 700492 не содержит нормальных жирных кислот и представляет собой 1,4'-дифосфат глюкозаминобиозы, ацилированный остатками (R)-3-{(7?)-3-гидроксидеканоил}оксидекановой кислоты в положениях 2, 2' и (R)-3-гидроксидекановой кислоты в положении 3.

8. Показано, что липиды А бактерий рода Marinomonas - первые представители соединений этого класса, содержащие необычную 3-ацилоксиалкановую кислоту, состоящую из двух остатков (/?)-3-гидроксидекановой кислоты.

9. Впервые из природных штаммов бактерий (морские бактерии Chryseobacterium т т* indoltheticum CIP 103168 и Chryseobacterium scophthalmum CIP 104199 ), выделен липид А, структурно подобный моносахаридному биосинтетическому предшественнику липида А мутантных клеток Escherichia coli. Показано, что липиды А этих бактерий представляют собой 1-фосфаты D-глюкозамина, ацилированные остатками (R)-3-гидрокси-шо-пентадекановой и (7?)-3-гидрокси-мзо-гептадекановой кислот в положениях 2 и 3 соответственно.

10. Изучены эндотоксические свойства липидов А и липополисахаридов ряда морских бактерий. Показано, что многие из них являются слабо токсичными соединениями и не индуцируют синтез фактора некроза опухоли альфа клетками периферической крови человека при всех изученных концентрациях.

11. На примере липополисахарида Marinomonas communis показано, что липиды А и липополисахариды морских бактерий проявляют свойства антагонистов эндотоксинов универсального типа действия и могут представлять интерес для терапии грамот-рицательного сепсиса и эндотоксикоза.
