ВВЕДЕНИЕ

Как известно, общий план строения развивающегося организма устанавливается в процессе гаструляции. В это время предетерминированные к различной судьбе клетки, мигрируя, формируют различные зародышевые листки. Клетки, образующие впоследствие энтодермальные и мезодермальные ткани, попадают внутрь зародыша, а будущие эктодермальные производные и зачатки нервной ткани располагаются по его поверхности. У позвоночных мезодерма является наиболее рано специфицирующейся тканью, которая одновременно подвергается достаточно детальной регионализации. Под регионализацией подразумевается то, что клетки в разных участках мезодермы коммитируются (предназначаются) к дифференцировке в разные специализированные типы. Из дорсальной области мезодермы, в частности, образуется хордомезодерма, которая становится "организатором", формирующим ось тела будущего зародыша. Клеточной спецификации мезодермы предшествует активация ранних зиготических генов Wnt 11, no tail, goosecoid, sna-1, eve-1 и др., некоторые из которых начинают работать уже на бластуле. Интересно, что клетки, активно экспрессирующие эти гены, расположены в краевой области бластодермы (Stachel et al., 1993; Hammerschmidt and Nusslein-Volhard , 1993; Thisse et al., 1993; Schulte-Merker et al., 1994; Talbot et al., 1995), причем экспрессия этих генов внутри краевой области подвергается пространственно-временной регуляции. На сегодняшний день точно известно, что молекулярная ассиметрия развития наступает еще до морфологической. Так, в бластодерме имеется ассиметрия в локализации таких ранних мезодермальных генов рыбы, как goosecoid ( Stachel et al., 1993; Schulte-

Merker et al., 1994 ) и fork head (Jesuthasan and Strahle, 1996) уже на стадии сферы. Показано для zebrafish, что на стадии сферы бластулы становится различим интенсивный сигнал zbmp-2 (Nikaido et al., 1997), локализация которого в бластодерме ассиметрична.

В последние десятилетия были обнаружены и молекулы, регулирующие экспрессию генов, участвующих в формировании мезодермального препаттерна

(работы проводились в основном на амфибиях). К ним относятся белковые факторы из семейств TGF-b, Wnt, FGF и др. (Sive et al., 1993; Kessler and Melton,1994; Slack et al., 1994; Doniach et al., 1995). Аналогичные факторы были обнаружены и у костистых рыб ( Cho et al., 1991; Lemaire et al., 1995; Talbot et al., 1995; Fisher et al., 1997; Furthauer et al., 1997). С каждым годом число обнаруженных морфогенов увеличивается. Существует, по-видимому, значительно большее количество факторов, пока еще неидентифицированных, участвующих в формообразовательных процессах и имеющих сложную взаимную регуляцию (Lemaire et al., 1996). Результатом их деятельности в конечном счете является формирование определенной препаттерной дифференцировки, в частности, мезодермальной.

Идентичности молекулярных механизмов развития мезодермы у амфибий и костистых рыб противостоит несхожесть процессов морфогенеза, в основе которых лежат различия. , между дискоидальным типом дробления рыб и меробластическим типом дробления амфибий. То обстоятельство, что зародыши рыб в последнее время становятся наиболее широко изучаемыми на ' молекулярном уровне объектами биологии развития, говорит о необходимости

проведения детального анализа их клеточных механизмов. Нашей основной задачей было изучение роли межклеточных взаимодействий в формировании препаттерна мезодермы у зародышей рыб и изучение его развития на стадиях вплоть до раннего сомитогенеза. На первом этапе работы нас интересовало взаимодействие бластомеров с желточной клеткой. Изучение взаимодействия бластомеров с желтком представляется нам важным потому, что зиготические гены, связанные с мезодермальной дифференцировкой, локализуются на стадии поздней бластулы в базальном слое бластодермы, примыкающем к желтку (Schulte-Merker et al., 1992; Stachel et al., 1993; Joly et al., 1993; Schulte-Merker et al., 1994). В процессе развития зародыша возможны два механизма взаимодействия бластомеров с желточной клеткой: переход из нее по цитоплазматическим связям в базальный слой бластодермы материала желтка (материнских факторов) еще до формирования желточного синцитиального слоя (Игнатьева, 1979), и переход низкомолекулярных веществ в бластомеры через высокопроницаемые межклеточные контакты - gap junctions или щелевые контакты (ЩК) (Potter et al., 1966) после формирования ЖСС. Возможно, что именно взаимодействия бластомеров с желтком посредством перечисленных двух механизмов лежат в основе формирования препаттерной генной активности. Важное значение желточной клетки в морфогенезе мезодермы и раннее предполагалось в работах по культивированию изолированных бластодерм рыб (Oppengeimer, 1936; Kostomarova, 1969; Rott et al., 1978 и др.). О важной роли желточной клетки в формировании дорсо-вентральной ассиметрии сообщали и другие работы (Long, 1983; Bozhkova et al., 1994; Mizuno et al.,1996). В недавней

работе Страла и Джисутасана (Jesuthasan and Strahle, 1996) были обнаружены субклеточные механизмы ассиметрии желточной клетки. Так, в желточной клетке различные вещества могут транспортироваться по микротрубочкам, причем процесс этот ассиметричен и в его основе лежит ассиметрия организации микротрубочек кортекса. Показано, что микротрубочки желтка на стадиях дробления входят в краевые бластомеры. Такая транспортировка веществ из желтка возможна, по данным авторов, до стадии 32-х клеток. Однако, цитоплазматические потоки в желтке у зародышей вьюна и данио наблюдаются не только в период дробления, но и позже, вплоть до средней гаструлы (Светлов, 1962). Многие детали процессов взаимодействия бластомеров с желтком не известны. Неясно также, когда заканчивается перенос морфогенетических веществ из желтка в зародыш по цитоплазматическим тяжам.

Трансплантационные эксперименты на рыбах показывают, что клетки становятся детерминированными к мезодермальной дифференцировке не ранее стадии средней гаструлы (Но and Kimmel, 1993). Только детерминированным клеткам свойственно автономное развитие, до этой стадии развитие клеток зависит от окружающих их тканей, как сообщают упомянутые авторы. После прекращения цитоплазматических связей большое значение для клеток приобретают межклеточные взаимодействия через молекулы адгезии и по каналам ЩК. Среди молекул клеточной адгезии, по-видимому, кадгеринам принадлежит ведущая роль при объединении клеток в ткани (Fujimono et al., 1990). Так, для зародышей амфибий было показано участие в адгезии клеток материнского ЕР-кадгерина (Heasman et al., 1994b ). Вполне вероятно участие в

этом процессе белков Wnt семейства и белков катенинового типа (TGF семейства) (Olson and Moon, 1992; Bradley et al, 1993; Kelly et al., 1995). Для зародышей рыб адгезионные молекулы еще не идентифицированы.

Известно, что у большинства зародышей уже ранние бластомеры связаны ЩК, в результате чего небольшие молекулы и ионы (менее 1000 Да) свободно проходят между клетками (см. лит. обзор, ниже). Если каналы ЩК еще не компактизированных зародышей мыши блокировать соответствующими антителами, то несмотря на продолжающиеся деления, клетки теряют способность компактизоваться (Lee et al., 1987). Похоже, что ЩК играют важную роль в синхронизации морфогенетических процессов на клеточном уровне. Однако, их влияние на механизмы формирования мезодермы и ее регионализации все еще далеки от понимания. Какая роль отводится индукционным потокам, обеспечиваемым лиганд-рецепторными взаимодействиями в этом процессе, а какая клеточным взаимодействиям через ЩК, до конца не ясно.

Предполагая, что на самых ранних стадиях потоки цитоплазмы из желточной клетки в бластомеры обеспечивают пространственное распределение материнских детерминант, необходимых для активации зиготических генов, индукции и регионализации мезодермы, мы изучили, во-первых, в течение какого периода развития существуют цитоплазматические связи между бластомерами и ЖК у зародышей вьюна и данио. Мы также описали динамику и пространственную картину процесса разобщения бластомеров с ЖК и выяснили, что этот процесс завершается только на поздней бластуле. Во-вторых, прерывая

цитоплазматические потоки, микрохирургически отделяя бластодерму от желточной клетки, мы проанализировали состояние бластодерм после изоляции в отношении наличия в них потенции к развитию хордомезодермы. Наши данные по изоляции бластодерм дополнили раннее проведенные работы (Kostomarova, 1969; Ротт и др., 1978) и позволили дать им альтернативную трактовку. В частности, мы показали, что повреждение нижнего слоя клеток, целостность которых нарушалась при механическом отделении от желтка, и последующая инкубация в натриевом физиологическом растворе существенно сказываются на их потенциях к развитию. При попытке сохранить эти клетки инкубацией в растворе KCl выяснилось, что процент дифференцированных структур в инкубируемых бластодермах увеличивался. Эти же опыты по культивированию позволили уточнить область бластодермы, в которой локализуются хордоме-зодермальные детерминанты после переноса их из желтка. Мы показали, что детерминанты достигают бластодерму уже на стадии 32 бластомеров. До этого момента дифференцировка в эксплантатах отсутствовала даже при инкубации бластодерм в растворе KCl. Таким образом, наши данные свидетельствуют в пользу того, что мезодермальные детерминанты попадают из желтка в бластомеры по цитоплазматическим связям в период от первых делений дробления до, по крайней мере, средней бластулы. С помощью маркирования флуоресцеин -декстраном (ФД) клеток, имеющих цитоплазматические связи с ЖК, мы показали, что окрашенные на ранней бластуле клетки являются потомками многих тканей, в том числе и потомками презумптивной хорды, однако после стадии 6,5-7 часов развития (13-14 т0) хордальный зачаток развивается уже

более автономно от желтка, тогда как включение меченных клеток в параосевую (сомитную) мезодерму еще продолжается. Похоже, что поступление хордоме-зодермальных детерминант из желтка в бластомеры заканчивается на этой стадии. Это предполагает, что передаваемая по цитоплазматическим связям информация может участвовать в регионализации мезодермы.

Изучая свойства ЩК методом переноса флуоресцентных красителей, различающихся величиной электростатического заряда: флуоресцеина (ФЛ), люциферового желтого (ЛЖ) и диамидино-2-фенилиндола (ДАФИ), мы впервые обнаружили, во-первых, что каналы ЩК могут избирательно пропускать вещества, различая их по заряду. Во- вторых, мы показали, что свойства ЩК различны в разных областях зародыша на стадиях бластулы и гаструлы. Так, будущая мезодерма в целом отличается по свойствам ЩК от будущей эктодермальной области. Имеются и дорсо-вентральные отличия в ЩК внутри мезодермы. При этом, однако, участок презумптивной хорды не образует ЩК с желточной клеткой и близлежащими клетками параосевой мезодермы и представляет собой изолированный компартмент по всем изученным красителям. Эти данные предполагают, что в процессе детерминации клетки мезодермы проиобретают особые свойства ЩК, способствующие поддержанию их специфичности.

Надеемся, что в целом наши данные окажутся полезными для дальнейших исследований раннего эмбриогенеза рыб и их мезодермы, в частности.

Выводы:

1. Наблюдение в базальном слое бластодермы зародышей вьюна ^¡БдигпиБ йзвБШв) клеток, формирующих цитоплазматические связи с желточной клеткой в период ранней-поздней бластулы, показало, что процесс отделения бластомеров является длительным и занимает весь период бластуляции. На стадии 10 х0 только около 20% клеток базального слоя изолированы от желтка плазматической мембраной. Полная изоляция бластодермы от желточной клетки происходит к стадии 17 То (к поздней бластуле). В это же время формируется и многоядерный желточный синцитиальный слой.

2. Отделение бластомеров от желточной клетки не является строго пространственно детерминированным. Выделение бластомеров происходит случайным образом и повсеместно, как в краевой зоне бластодермы, так и в ее центральной части. Статистически, тем не менее, более длительное время связанными с ЖК оказываются клетки краевой бластодермы (презумптивной мезодермы).

3. Опыты по механическому прерыванию связи бластомеров с желточной клеткой и последующей инкубацией эксплантатов показали, что уже со стадии 6 х0 около 40% бластодерм способны формировать зачаток хорды. Изоляты со стадии 12 т0 формировали зачаток хорды в 90% случаев. Потенции к дифференцировке со-митообразующей мезодермы обнаруживались в изолятах более поздних зародышей, начиная со стадии 10 х0.

4. Повреждение клеток базального слоя бластодермы, цитоплазматически связанных с желтком, после изоляции и инкубирования в разных солевых растворах приводило к значительному уменьшению их потенциий к мезодермальной диффе-ренцировке. Повреждаемость таких клеток у бластодерм, изолированных на стадиях 6-10 т0, была ниже в растворе с высоким содержанием калия, чем в растворе с высоким содержанием натрия и, соответственно, возрастала способность к дифференцировке. Дифференцировка бластодерм становилась нечувствительной к условиям инкубации на стадиях поздней бластулы, когда большинство бласто-меров было уже отделено от желточной клетки плазматической мембраной.

5. Предполагается последовательное послойное поступление мезодермальных детерминант или других активаторов дорсальной мезодермальной дифференци-ровки по цитоплазматическим тяжам из желтка зародыша в период ранней бластулы.

6. После образования желточного синцитиального слоя в зародышах рыб образуется два морфологически раздельных компартмента-желточная клетка и бластодерма. С помощью инъекций в желточную клетку флуоресцентных красителей с молекулярной массой менее 500 Да и высокомолекулярного красителя флуо-ресцеин-изотиоционат-декстрана с молекулярной массой 17 ООО Да нами было показано, что связь между этими областями осуществляется по щелевым контактам.

7. Впервые подробно изучена проницаемость щелевых каналов у зародышей вьюна с помощью трех разных флуоресцентных проб близкой молекулярной массы (люциферового желтого, флуоресцеина и 4,6-диамидино-2-фенилиндола) на стадиях от средней бластулы до поздней гаструлы. Показано существование в зародыше областей, различающихся по свойствам проницаемости щелевых контактов. На стадии ранней гаструлы в базальном слое бластодермы было обнаружено три коммуникационных компартмента (по связи бластомеров с желточной клеткой): область презумптивной эктодермы, область презумптивной дорсальной мезодермы и область презумптивной вентролатеральной мезодермы. На стадии средней гаструлы к ним добавляется компартмент презумптивной хорды. Специфика проницаемости каналов щелевых контактов разных областей предшествует их детерминации.

8. Впервые ¡n vivo показано существование щелевых контактов с анионной селективностью, не пропускающих катионный краситель (4,6-диамидино-2-фенилиндол), но высокопроницаемых для анионов (люциферового желтого и флуоресцеина). Такие контакты формировались в вентролатеральной области краевой бластодермы (презумптивной мезодермы) вьюна на стадиях ранней-средней гаструлы. Клетки дорсальной области краевой бластодермы, наоборот, формировали неселективные щелевые контакты, одинаково хорошо проницаемые для катионных и анионных красителей.

9. Впервые обнаружено, что область презумптивной хорды у зародышей рыб на стадии средней гаструлы представляет собой отдельный компартмент, который в отличие от других областей мезодермы не связан с желточной клеткой щелевыми контактами, пропускающими хотя бы один из использованных красителей. Показано с помощью флуоресцентных красителей, что область презумптивной хорды до стадии 50% эпиболии находится в глубоких слоях бластодермы.

10. Показано, что обработка зародышей вьюна на ранней бластуле в течение 510 минут 0,3 М ЫС1 сдвигает их последующее развитие в сторону дорсализации. При этом все клетки, в норме предетерминированные к вентральной судьбе, приобретают вместе с дорсальной спецификацией и щелевые контакты, селективность которых соответствует клеткам дорсальной спецификации.

Заключение

Как и у других видов костистых рыб (Lentz and Trinkaus, 1967; Long, 1980), у вьюна на стадии бластуляции из ранее цитоплазматически связанных перибласта и желтка образуется два различных функциональных компартмента: бластомеры и желток. Эти компартменты различаются и по темпу цитокинеза (Капе et al., 1992). Морфологическое отделение бластомеров от желточной цитоплазмы происходит за счет формирования плазматических мембран на нижней границе ба-зальных бластомеров, т.е. над поверхностью цитоплазмы желтка, что приводит к окончательному отделению бластодермы от желтка и формированию ЖСС. С этого момента желток, заключенный в цитоплазматическую мембрану, представляет собой в функциональном смысле внезародышевый орган - желточный мешок, а в цитологическом - желточную клетку - ЖК. Как показали наши опыты с инъекциями ФД в ЖК на зародышах вьюна, процесс их морфологического разделения оказывается более длительным, чем ранее предполагалось (Kimmel and Law, 1985), и завершается только к началу эпиболии (рис. 2А). Это же было показано нами ранее и для зародышей данио (Розанова, Божкова, 1995). Наш результат совпадает также с данными, полученными на Catostomus, у которых ЖСС тоже формируется только к началу эпиболии (Long, 1980). Гистологические исследования зародышей вьюна, как и данные, приведенные Киммелом и Лоу (Kimmel and Law, 1985) для данио, показали определенную пространственную закономерность потери цито-плазматических связей, когда более длительно связанными с цитоплазмой ЖК оказывались краевые бластомеры. Краевые и центральные бластомеры, таким образом, различаются по времени их образования. С другой стороны, они включают в себя и разную по составу цитоплазму. В течение всего времени отделения бластомеров от ЖК (от 10-со до 17то) происходит поступление в ЖСС (Long, 1980) большого количества рибосом (Айтхожин и др., 1964; Игнатьева, 1979; Thomas, 1968; Hagenmeier, 1969; Long 1983) и, повидимому, других морфогенети-ческих веществ, что предполагает важную роль ЖСС в предетерминации мезодермы. Пока из материнских факторов, определяющих регионализацию мезодермы у амфибий и рыб, наиболее исследован ß-катенин (Heasman et al., 1994; Sokol et al.,1995; Funajama et al., 1995; Kelly et al., 1995; Schneider et al., 1996; Heasman et al., 1997 др.), компонент Wnt-сигнальной системы. Он временно накапливается в ядрах клеток вегетативной части зародышей и ЖК на дорсальной стороне на средней бластуле и его локализация предшествует экспрессии дорсально специфических генов siamois и goosecoid (Cho et al., 1991; Lemaire et al., 1995). Что касается времени поступления мезодермальных детерминант в бластодерму рыб, то наши данные показывают, что дифференцировка мезодермы у зародышей вьюна также, как и у зародышей Fundulus (Oppenheimer, 1936), может идти автономно от желтка уже со стадии 32 клеток. Часть бластодерм, изолированных на стадии 5-6 деления (6-7 то), т.е. существенно ранее наступления активности морфогенетиче-ской функции ядер, которая у вьюна происходит на стадии 12-13 то (Нейфах и Тимофеева, 1977), не только образует выросты, характерные для мезодермальной ткани, но и развивает участки хордомезодермы. Особенно отчетливо эта способность обнаруживается в растворе 2, что согласуется с данными и др. авторов (Ротт и др., 1978). Наши данные имеют расхождение с данными Костомаровой (Kostomarova, 1969), не обнаружившей дифференцировки у бластодерм вьюна, изолированных до средней бластулы. Объяснить эти расхождения можно различием методов изоляции бластодерм и анализа их дифференцировок. Мы предполагаем, что в опытах Костомаровой (1969) использование для изоляции бластодерм метода центрифугирования приводило к большему повреждению «клеток без дна» и к большей потери из них морфогенетических веществ, чем в случае микрохирургической изоляции бластодерм, использованной в наших опытах и опытах Оппенгеймер (1936). Мы, как и Оппенгеймер (1936), не смогли однако обнаружить признаков дифференцировки у бластодерм, изолированных до стадии 32 бластомеров. Это совпадает с данными Джесутасана и Страхла (Jesuthasan and Strahle, 1996) показавших, что гипотетические дорсализующие материнские факторы могут транспортироваться из желтка в бластодерму данио по системе кортикальных микротрубочек в этот период, до стадии 32 бластомеров, и эти факторы необходимы для активации ранних мезодермальных генов goosecoid и forkhead 2. Механизмы движения этих веществ по микротрубочкам пока только предполагаются. Известно также, что определенную роль в переходе цитоплазмы из ЖК в область бластодиска может играть кортикальный актиновый цитоскелет (Ivanenkov et al., 1987). Как у амфибий, так и у рыб предполагаеся, что в формировании мезодермы участвуют гены, активируемые как материнскими факторами, так и внеклеточными сигналами индукционного типа. У амфибий индукционной активностью обладают бластомеры вегетативной области зародыша, а у рыб -желточная клетка (Gerhart et al., 1986; Mizuno et al., 1996). Вопрос о других механизмах, требует специального изучения. После того как непрерывная цитоплазма-тическая связь бластомеров с ЖК прекращена и сформировался ЖСС, из него в бластомеры, как отмечалось ранее, могут проходить низкомолекулярные вещества по ЩК (Potter et.al., 1966). Исследование специфики ЩК в базальном слое бластодермы было проведено во второй части работы. Основным результатом этого исследования стало обнаружение компартментов в пределах базального слоя бластодермы (презумптивной мезодермы и эктодермы), клетки которых имеют разные свойства ЩК, формируемые ими с ЖК: ЩК, пропускающие анионы, но не пропускающие катионы, ЩК, неселективные по ионам, но обладающие высокой проницаемостью и слабо проницаемые ЩК. Данные по изучению проницаемости ЩК между бластомерами ЖК у вьюна согласуются с результатами более ранних работ (Божкова и Воронов, 1995; Божкова, 1997; Bozhkova and Voronov,1997) и дополняют ранние исследования новыми фактами. Впервые обнаружены у зародышей рыб ЩК с анионной селективностью. Они появляются на стадии начала формирования зародышевого кольца у клеток вентролатеральной области презумптивной мезодермы. Из семи исследованных на настоящее время коннексинов считается, сто Сх45 и Сх26 могут формировать катион-, а Сх43 и Сх31 анионсе-лективные каналы (Brissette et al., 1994; Steinberg et al., 1994; Veenstra et al., 1994; Elfgang et al., 1995; Waltzman and Spray, 1995; Koval et al., 1995). Однако, эти данные были получены на искуственных системах, на клетках, трансфецированных экзогенными генами. До настоящего времени практически не имеется работ, в которых бы сравнивалась проницаемость ЩК для молекул разного заряда в клетках in vivo. В некоторых, из проведенных, работах не было обнаружено ионной селективности ЩК (Flagg-Newton et al., 1979; Imanaga et.al., 1987). По нашим данным, к клеткам презумптивной мезодермы, ЩК которых не обладают ионной селективностью, относятся клетки передней области зародышевого щитка и клетки центральной части базального слоя бластодермы (презумптивной эктодермы). Но вместе с тем, наша работа впервые показывает существование ион-селективных ЩК. Такие каналы формировались между клетками вентролатеральной области зародышевого кольца и ЖК. На сегодняшний день не имеется универсальной гипотезы о роли ЩК в дифференцировке. Существует только общее представление о том, что дифференцирующиеся клетки, которые в дальнейшем станут разнотипными, остаются связанными друг с другом диффузионными каналами. Клетки, прошедшие дифференцировку, теряют связь с клетками другого типа. Существование коммуникационных компартментов, пространственно совпадающих с областями детерминации, было показано для млекопитающих и асцидий (Kimmel et al., 1984; Goodall, Johnson, 1984; Serras et al., 1985; Van den Biggelaar, Serras, 1988). Наши наблюдения на рыбах дополняют эти результаты и показывают, что в ранних зародышах рыб изменения свойств ЩК сопровождают предетерминацию клеток разных областей мезодермы. Причем разные свойства ЩК могут обеспечивать условия для пространственного распределения сигналов к дифференцировке.
