1.1 Функциональное разделение генетических факторов Telomere elongation (Tel) и Enhancer of terminal gene conversion (E(tc)).

Последовательность от -69 до -100 п.н. необходима для стимуляции экспрессии yellow в теле и крыльях.

Последовательность от -69 до -100 п.н. не влияет на базовую активность промоторов, но она необходима для коммуникации между энхансером и находящимся от него на расстоянии промотором.

Последовательность от -69 до -100 п.н. необходима для репрессии промотора yellow gypsyинсулятором на фоне мутации mod(mdg4)ul.

Роль последовательности -69 - -100 п.н. в энхансер-промоторных взаимодействиях.

2.2 Функциональное взаимодействие между Su(Hw) инсуляторами, находящимися на большом расстоянии друг от друга, может регулировать энхансер-промоторные взаимодействия у Drosophila melanogaster.

Теломеры - это специализированные ДНК-белковые комплексы, находящиеся на концах линейных хромосом. Теломеры предохраняют концы хромосом от слипания, деградации, узнавания системой репарации ДНК. Таким образом, основной функцией теломер является обеспечение стабильности эукариотического генома. В настоящее время доказано, что поддержание строго определенного размера теломер критично для жизнедеятельности организма. Изменение длины теломер тесно связано с опухолеобразованием и старением клетки. У большинства высших эукариот теломеры состоят из простых повторов G-богатой последовательности, а их удлинение обеспечивается специальным ферментом - теломеразой. У Drosophila melanogaster теломеры состоят из мобильных элементов типа LINE, ориентированных «голова к хвосту» - НеТ-А, TART и TAHRE. В результате проведенных в нашей лаборатории исследований было показано, что удлинение теломер дрозофилы может происходить по трем механизмам: с помощью транспозиции мобильных элементов на конец хромосомы; с помощью генной конверсии и с помощью рекомбинации между теломерными повторами. Основными структурными единицами нормальной теломеры являются: 1) терминальный комплекс - это белковый комплекс, формирующийся на конце хромосомы и защищающий его от ферментов репарации; 2)теломерный хроматин, который формируется на последовательностях теломерной ДНК. Данные структуры играют основную роль в процессе регуляции длины и стабильности теломер. Важной особенностью дрозофилы, является то, что у нее теломерный хроматин может формироваться на любой неспецифичной последовательности ДНК, в то время как у теломеразозависимых организмов теломерная структура формируется только при наличии строго определенных последовательностей, которые представляют собой сайты связывания для белков, формирующих теломерный хроматин. Необходимо отметить, что несмотря на различия в структуре теломер, у теломеразозавимых организмов и у дрозофилы существуют общие механизмы, обеспечивающие поддержание стабильного размера теломер. Во-первых, эксперименты, проведенные на дрожжах и млекопитающих, доказали, что при инактивации теломеразы у данных организмов индуцируется альтернативный механизм удлинения теломер, связанный с процессами конверсии\рекомбинации. В том числе, альтернативные механизмы удлинения теломер часто реализуются в раковых клетках. Во-вторых, в последнее время было найдено, что некоторые консервативные белки, участвующие в репарации ДНК, также принимают участие в формировании терминального комплекса, как у дрожжей и млекопитающих, так и у дрозофилы. Поэтому данные, полученные при изучении теломер дрозофилы помогут понять закономерности процесса регуляции длины теломер у различных эукариотических организмов и выявить основные белки, формирующие теломерный комплекс.

Также, актуальным направлением исследований современной молекулярной генетики является изучение функциональных взаимодействий между регуляторными элементами генома, расположенными на большом расстоянии друг от друга, и роли таких взаимодействий в регуляции транскрипции. Точная временная и тканеспецифичная транскрипция генов высших эукариот обеспечивается с помощью ряда регуляторных элементов. В настоящее время наиболее изучены различные типы энхансеров, усиливающих транскрипцию, сайленсеров, ослабляющих транскрипцию, и промоторов, обеспечивающих базовый уровень транскрипции. Часто регуляторные элементы находятся на очень больших расстояниях от промоторов и могут отделяться от них другими генами. Существует несколько моделей, объясняющих механизм дистанционных взаимодействий между регуляторными элементами. Наиболее популярна модель «выпетливания». Эта модель предполагает, что активация транскрипции происходит в результате прямого взаимодействия между транскрипционными факторами, связанными с энхансером и промотором, при этом ДНК между ними выпетливается. В образованной хроматиновой петле находятся не активные на данном этапе развития гены. Таким образом, модель «выпетливания» поднимает актуальный вопрос: как потенциально разнородные регуляторные элементы, входящие в состав с/л-регуляторной области, игнорируют близкие к ним промоторы и взаимодействуют с промотором только своего гена-мишени? На энхансер-промоторные взаимодействия может влиять еще один тип регуляторных элементов, названных инсуляторами. Классическим свойством инсуляторов является их способность, располагаясь между энхансером и промотором, блокировать взаимодействие между ними. Но уже неоднократно, в том числе и в данной работе, показано, что взаимодействуя между собой, инсуляторы способны реорганизовывать пространственную структуру ДНК. Таким образом, они могут модулировать взаимодействия между энхансерами и промоторами и даже способствовать взаимодействию между регуляторными элементами, расположенными на расстоянии друг от друга.

Представленная работа посвящена выявлению молекулярно-генетических факторов, участвующих в регуляции длины теломер дрозофилы, исследованию свойств теломерного хроматина, а также изучению регуляторных последовательностей и белков, участвующих в организации и в поддержании дистанционных взаимодействий, влияющих на транскрипцию.

выводы

1. Проведено функциональное сравнение генетических факторов Tel и Eft с), влияющих на удлинение теломер. Установлено, что мутация Tel не приводит к предпочтению транспозиционного механизма удлинения теломер, как предполагалось ранее. В линиях с терминально делетированными хромосомами Tel увеличивает как частоту присоединений HeT/A/TART элементов к концу хромосомы, так и частоту терминальных генных конверсий.

2. Впервые показано, что белки НР1 (Heterochromatin Protein 1), Ku70 и Ku80 участвуют в негативной регуляции удлинения теломер у Drosophila melanogaster и играют важную роль в защите концов хромосом.

3. Показано, что на концевых последовательностях ДНК длиной 4-5 т.п.н. формируется особый теломерный хроматин, который негативно влияет на Ро1усошЬ-зависимую репрессию, активность транспозазы Р-элемента, а также подавляет генную конверсию, которая потенциально возможна при возникновении терминальной делеции. Продемонстрировано, что существует антагонизм между теломерным и субтеломерным (Pc-G-зависимым) хроматином.

4. Выявлено, что теломерный хроматин способен подавлять дистанционные взаимодействия между промотором yellow и полностью различными регуляторными элементами, такими как yellow энхансеры и gypsy инсулятор.

5. Предложена новая модель, описывающая механизм удлинения хромосом и контроля их длины у Drosophila melanogaster, предполагающая формирование Т-петли на конце хромосомы и основанная на антагонизме между двумя типами хроматина, формируемого на теломерах и TAS.

6. Впервые в предпромоторной области гена yellow обнаружены комуникаторные элементы, обеспечивающие как дистанционное взаимодействие между энхансером и промотором, так и gypsy (Su(Hw)) -зависимую репрессию.

7. В модельной системе гена yellow продемонстрировано, что расстояние между инсуляторами Su(Hw), окружающими энхансеры, влияет на способность инсуляторов блокировать взаимодействие между этими энхансерами и промотором. Когда расстояние между взаимодействующими инсуляторами достаточно мало, энхансеры, расположенные между ними, не могут взаимодействовать с изолированным промотором. При увеличении расстояния между инсуляторами взаимодействие между энхансерами и промотором восстанавливается.

8. Показано, что компонент Su(Hw) инсулятора белок Mod(mdg4)-67.2 негативно влияет на частоту Р-транспозиций, если Su(Hw) инсулятор входит в состав Р-транспозона.

9. Доказано, что в отличие от результатов, полученных в гетерологичных модельных системах (дрожжах и культуре клеток человека), в дрозофиле сайты связывания GAF из промоторов генов hsp26 и hsp70 не способны обеспечивать коммуникацию между регуляторными элементами, расположенными на большом расстоянии друг от друга.

10. Впервые показано, что взаимодействие между GAGA фактором и белком Mod(mdg4)-67.2 позволяет преодолеть активность Su(Hw) инсулятора у Drosophila melanogaster. Белки GAF и Mod(mdg4)-67.2 взаимодействуют in vitro и in vivo.

11. Доказано, что белок Zeste необходим для дистанционных взаимодействий между энхансером и промотором и преодоления энхансерами блокирующего действия инсуляторов в модельной системе гена white у Drosophila melanogaster.
