На протяжении почти столетия митохондрии рассматривались как энергетические станции, обеспечивающие клетку энергией. Однако результаты, полученные в последнее время, показали, что окислительное фосфорилирование не единственная функция митохондрий. Помимо производства энергии, эти органеллы в ходе ретроградной регуляции могут регулировать экспрессию ядерных генов (Rhoads et al., 2006). Механизм, с помощью которого митохондриальный и ядерный геномы взаимодействуют друг с другом, еще не достаточно изучен. Предполагается, что важную роль в этом процессе играет повышение генерации активных форм кислорода (АФК) и изменение концентрации внутриклеточного кальция (Юрина, Одинцова, 2010). Митохондрии, выступая как один из источников образования АФК (Blokhina, Fagerstedt, 2010), и, являясь депо внутриклеточного Са2+ в клетке (Logan, Knight, 2003; Subbaiah et al., 1998; Медведев, 2005), могут регулировать содержание этих вторичных мессенджеров в цитозоле и, тем самым, влиять на экспрессию ядерных генов (Тарчевский, 2002).

Другим важным откровением последних лет явилось понимание, что митохондрии играют активную роль в активации программированной гибели клеток (ПГК) (Wang, Youle, 2009). Гибель клеток в процессе развития ПГК, опять же, зависит от нарушения гомеостаза внутриклеточного кальция (Szabadkai, Duchen, 2008) и усиления митохондриальной продукции АФК (Perrone et al., 2008). Тепловой шок может инициировать развитие ПГК в клетках животных и растений (Gabai, Sherman, 2002; Reape, McCabe, 2010). Показано, что развитие ПГК может наблюдаться в клетках дрожжей после действия самых различных воздействий (Skulachev, 2002; Гордеева и др., 2004; Carmona-Gutierrez et al., 2010), но неизвестно, может ли такое же событие происходить после теплового шока.

Предполагается, что основной причиной гибели после теплового шока является денатурация и агрегация клеточных белков (Kampinga, 2006; Kubota et al., 2009). Очевидно, что усиление генерации АФК также вносит существенный вклад в активацию процесса гибели как растительной (Suzuki, Mittler, 2006), так и дрожжевой клетки (Davidson et al., 1996) при повышении температуры. При этом механизм, в результате которого усиливается генерация АФК, остается совершенно неизвестным. Повышение концентрации внутриклеточного Са2+ стимулирует генерацию АФК (Гордеева и др., 2003). Поэтому повышение уровня кальция в цитозоле также, вероятно, имеет большое значение в запуске механизма гибели клетки после теплового шока. Поэтому можно предположить, что митохондрии дрожжевой и растительной клетки, генерируя АФК, и участвуя в регуляции уровня Са2+ в клетке, играют важную роль в активации механизма гибели при тепловом шоке, а сам процесс гибели происходит программируемым образом.

Чтобы защитить себя от повреждающего воздействия теплового шока, клетки синтезируют белки теплового шока (БТШ). Повышение уровня БТШ приводит к повышению термотолерантности организма не только к жесткому тепловому шоку (Richter et al., 2010). Принято считать, что БТШ, действуя в качестве молекулярных шаперонов, восстанавливают поврежденные при нагревании белки или, если это невозможно, способствуют их деградации путем протеолиза (Kubota et al., 2009). Но, вероятно, БТШ выполняют в клетке и другие функции. Показано, что повышение уровня БТШ помогает растительной и дрожжевой клетке переживать отрицательные последствия усиления генерации АФК (Flower et al., 2005; Qi et al., 2011). Известно также, что БТШ, модулируя конформационное состояние определенных белков, могут подавлять, либо, наоборот, активировать развитие ПГК в клетках животных (Arya et al., 2007). Когда данная работа начиналась, было неизвестно, могут ли БТШ растений и дрожжей ингибировать развитие ПГК и защищать клетки не только от термической денатурации и агрегации, но и от последствий повышения генерации АФК.

Другой важный вопрос, на который до сих пор нет четкого ответа, как активируется экспрессия БТШ? Описаны транскрипционные факторы теплового шока (ФТШ) в клетках растений и дрожжей, которые активируют экспрессию БТШ. Известны промоторные последовательности в генах БТШ, с которыми эти ФТШ взаимодействуют. Но можно только догадываться, что же является первопричиной активации генов БТШ. Классическая модель регуляции экспрессии генов БТШ при повышении температуры основана на появлении в клетке денатурированных белков, которые в результате взаимодействия с Hsp90p или с Hsp70p активируют ФТШ (Voellmy, Boellmann, 2007). Однако эта модель не объясняет, почему обработка рядом агентов, которые не вызывают появления денатурированных белков, тем не менее, активирует экспрессию генов БТШ (Balogh et al., 2005; Saidi et al., 2011; Finka et al., 2011). Более того, непосредственно продемонстрировано, что активация экспрессии генов БТШ у мха Physcomitrella patens при повышении температуры может происходить независимо от появления денатурированных белков (Saidi et al., 2009). Биоинформационный анализ, проведенный Finka et al. (2011), показал, что появление в клетке денатурированных белков является достаточным, но не обязательным условием для активации экспрессии генов БТШ. С другой стороны, есть основания полагать, что усиление генерации АФК и кратковременное повышение уровня кальция в цитозоле может как непосредственно, так и косвенным образом оказывать значительное влияние на транскрипционную активацию БТШ (Balogh et al., 2005; Saidi et al„ 2011).

Таким образом, известные к настоящему времени литературные данные, позволяют предполагать, что усиление продукции АФК и повышение уровня внутриклеточного кальция, может, с одной стороны, являться важным фактором в запуске механизма гибели клеток дрожжей и растений при тепловом шоке, а, с другой стороны, активировать экспрессию генов БТШ, которые защищают клетки от гибели (Креславский и др., 2012). Поскольку митохондрии являются основным источником АФК и активно влияют на концентрацию кальция в клетке, можно предполагать, что эти органеллы играют критическую роль в судьбе клетки при тепловом шоке. В зависимости от интенсивности теплового воздействия митохондрии определяют: либо клетка погибнет, либо выживет. Несмотря на то, что за последнее десятилетие достигнут значительный прогресс в понимании механизма участия митохондрий в регуляции ПКГ и их роли в ретроградной регуляции экспрессии ядерных генов, было неизвестно какую роль выполняют эти органеллы в запуске гибели клетки дрожжей и растений при тепловом шоке. Также не известно, может ли митохондрия принимать участие в регуляции экспрессии генов БТШ.

Данная работа выполнялась в период с 1999 г. по 2011 г. в лаборатории физиологической генетики СИФИБР СО РАН. Отдельные эксперименты проводились в в лаборатории доктора Н. Iida (Tokyo Gakugei University, Япония). Ряд экспериментов выполнен в сотрудничестве с доктором Т. Tanaka (Университет Осака, Япония).

Цель и задача исследования. Целью настоящей работы являлось изучение роли митохондрий растительной и дрожжевой клетки в запуске программы гибели и в активации экспрессии генов белков теплового шока в ответ на тепловое воздействие.

Для достижения поставленной цели необходимо было решить следующие задачи:

1. Изучить зависимость между способностью дрожжей расти при температурах выше оптимальной и способностью выдерживать кратковременное повреждающее действие теплового шока.

2. Сравнить базовую термотолерантность дрожжей, различающихся по типу энергетического метаболизма.

3. Изучить эффект нарушения функционирования дыхательной цепи на изменение базовой термотолерантности и генерацию активных форм кислорода (АФК) при тепловом шоке в клетках дрожжей.

4. Установить зависимость между продукцией АФК в клетках Saccharomyces cerevisiae и Arabidopsis thaliana при тепловом шоке и гиперполяризацией внутренней митохондриальной мембраны.

5. Выявить признаки развития программированной гибели клеток (ПГК) после действия теплового шока различной интенсивности в клетках сегеушяе и А. ЛаНапа.

6. Определить эффект повышения уровня белков теплового шока (БТШ) на развитие признаков ПГК в клетках 5. cerevisiae и А. гИаНапа после действия теплового шока.

7. Изучить зависимость между действием митохондриальных ингибиторов и разобщителей на развитие индуцированной термотолерантности, экспрессию генов белков теплового шока и изменение потенциала на внутренней митохондриальной мембране в клетках 5*. сегеушае и А. ЛаИапа.

8. Изучить механизм повышения уровня кальция в цитозоле Б. сегеу1з1ае при тепловом стрессе и установить связь между этим явлением и функционированием митохондрий.

Основные положения, выносимые на защиту.

1. Дрожжи реализуют различные стратегии адаптации, чтобы существовать в условиях длительного повышения температуры и выдерживать повреждающее воздействие кратковременного теплового шока.

2. Механизмы гибели клеток дрожжей и растений при тепловом шоке существенно различаются в зависимости от интенсивности теплового воздействия. Гиперполяризация внутренней митохондриальной мембраны при умеренном тепловом шоке сопровождается повышением митохондриальной продукции активных форм кислорода, что является одной из причин гибели.

3. При умеренном тепловом шоке гибель клеток дрожжей и растений имеет признаки программированной гибели, которая подавляется в результате повышения количества БТШ.

4. Митохондрии растительной и дрожжевой клетки играет активную роль в развитии индуцированной термотолерантности к повреждающему тепловому шоку. Гиперполяризация внутренней митохондриальной мембраны при мягком тепловом стрессе является необходимым условием для экспрессии генов БТШ.

Научная новизна работы. В результате сравнения способности различных видов дрожжей, а также мутантов 5. сеге\>'тае переносить повреждающее действие теплового шока и расти при температуре выше оптимальной, впервые продемонстрировано, что между этими двумя явлениями отсутствует зависимость. Клетки дрожжей, неспособные поддерживать рост при повышенной температуре, могут лучше сохранять жизнеспособность после кратковременного жесткого теплового шока.

Впервые проведено сравнение базовой термотолерантности видов дрожжей, различающихся по типу энергетического метаболизма. Показано, что способность дрожжей переживать повреждающее действие теплового шока зависит от того, какой энергетический метаболизм, бродильный или окислительный, используют дрожжи. Активация окислительного энергетического метаболизма в клетках S. cerevisiae сопровождается увеличением конститутивного синтеза Hspl04p и повышением термотолерантности.

Впервые показано, что повышение температуры вызывает гиперполяризацию внутренней митохондриальной мембраны в клетках S. cerevisiae и A. thaliana, что сопровождается усилением продукции АФК и снижением жизнеспособности. В клетках S. cerevisiae гиперполяризация и значительное усиление продукции АФК наблюдаются при умеренном (45°С), но не при жестком тепловом шоке (50°С).

Анализ изучения параметров снижения жизнеспособности при умеренном тепловом шоке позволил впервые показать, что гибель дрожжей S. cerevisiae и A. thaliana имеет признаки программированной гибели клеток (ПГК): повышение продукции АФК, развитие процесса во времени и выход цитохрома с и Hsp60p из митохондрий. Гибель клеток S. cerevisiae при умеренном тепловом шоке подавляется в результате мутации petite и обработки циклогексимидом.

Изучение распределения цитоплазматического белка HsplOlp A. thaliana между митохондриальной и цитоплазматической фракциями белков впервые продемонстрировало, что Hsp 101 р при тепловом стрессе локализуется преимущественно в митохондриальной фракции белков. Гомологичный ему белок Hspl04p S. cerevisiae также обнаруживается в митохондриальной фракции клеток дрожжей.

Впервые показано, что повышение количества БТШ в результате предварительного теплового стресса ингибирует все наблюдаемые признаки развития ПГК в клетках растений и дрожжей, включая выход митохондриальных белков из митохондрий, повышение продукции АФК, а также снижает длительность гиперполяризации внутренней митохондриальной мембраны.

Продемонстрировано, что гиперполяризация внутренней митохондриальной мембраны является необходимым условием для активации экспрессии БТШ при тепловом стрессе. Агенты, которые при данных экспериментальных условиях подавляют гиперполяризацию, одновременно подавляют тепловую активацию экспрессии генов БТШ.

Изучен механизм повышения уровня внутриклеточного кальция в клетках S. cerevisiae при тепловом стрессе. Повышение уровня Са в цитозоле S. cerevisiae при тепловом стрессе определяется функционированием низкоаффинной и высокоаффинной (Midlp и Cchlp) систем транспорта Са2+ через плазматическую мембрану.

Впервые показано, что отсутствие белков Midlp и Cchlp, определяющих транспорт кальция через плазматическую мембрану, приводит к снижению потенциала на внутренней митохондриальной мембраны и к подавлению дыхательной активности в клетках S. cerevisiae.

Научно-практическая значимость. Результаты, полученные в данной работе, позволили впервые доказать, что митохондрии растительной и дрожжевой клетки, модулируя продукцию активных форм кислорода (АФК) и участвуя в гомеостазе внутриклеточного кальция, определяют судьбу растительной и дрожжевой клетки при тепловом воздействии. В ответ на повышение температуры в клетке повышается потенциал на внутренней митохондриальной мембране, что в зависимости от интенсивности воздействия, либо запускает механизм гибели, либо активирует экспрессию белков теплового шока (БТШ), защищающих клетку от гибели. Способность митохондриальных ингибиторов и разобщителей, модулируя потенциал на внутренней митохондриальной мембраны, ингибировать активацию экспрессии генов БТШ растений и дрожжей при тепловом стрессе, указывает, что обнаружен новый, ранее неизвестный, механизм, с помощью которого митохондрии регулируют экспрессию ядерных генов БТШ в ответ на стрессовое воздействие.

Изучение роли митохондрий в продукции АФК имеет огромное значение в связи с участием АФК в развитии многих заболеваний человека, таких как инсульт, инфаркт, нейродегенеративные заболевания. Понимание механизма образования АФК митохондриями может помочь в разработке новых терапевтических подходов и лекарственных средств для борьбы с этими заболеваниями. Описанный в работе феномен синергического действия теплового шока и митохондриальных ингибиторов на снижение жизнеспособности дрожжей может быть использован для разработки новых способов борьбы с патогенными микроорганизмами и лечения раковых опухолей, устойчивых к химиотерапии. В условиях глобального потепления и загрязнения окружающей среды антропогенными соединениями, влияющими на активность митохондрий, результаты диссертации могут быть использованы для оценки экологических и экономических рисков.

Материалы диссертации могут быть включены в курсы лекций по генетике, экологии и физиологии растений, использоваться в профильных научно-исследовательских институтах РАН, РАМН и РАСХН.

Полученные результаты позволяют предполагать, что БТШ растений и дрожжей, так же как БТШ животных, могут модулировать активность митохондрий и ингибировать развитие программированной гибели.

7. Тепловой стресс, активирующий экспрессию генов БТШ и развитие индуцированной термотолерантности в клетках S. cerevisiae и A. thaliana, сопровождается кратковременным повышением уровня Са в цитозоле и гиперполяризацией внутренней митохондриальной мембраны. Гиперполяризация внутренней митохондриальной мембраны является необходимым условием для активации экспрессии БТШ при тепловом стрессе. j I

8. Повышение уровня Са в цитозоле S. cerevisiae при тепловом стрессе определяется функционированием низкоаффинной и высокоаффинной (Midlp |и Cchlp)

2+ 1 ' системами транспорта Са через плазматическую мембрану. Нарушение гомеостаза внутриклеточного кальция в клетке S. cerevisiae в результате ^ одновременной утраты белков Midlp и Cchlp приводит jk ингибированию дыхательной активности и деполяризации внутренней митохондриальной мембраны, что указывает на существование связи между гомеостазом внутриклеточного кальция и функциями митохондрий в клетках дрожжей.

6. ЗАКЛЮЧЕНИЕ

В научной литературе понятия термоустойчивость или термотолерантность используются как для оценки роста при повышении температуры, так !и оценки жизнеспособности после повреждающего теплового шока. В результате сравнения способности различных видов дрожжей, а также мутантов S.1 cerevisiae переносить повреждающее действие теплового шока и расти при температуре выше оптимальной, впервые было показано, что между этими двумя явлениями отсутствует зависимость. Клетки дрожжей, неспособные поддерживать рост при повышенной температуре, могут лучше сохранять жизнеспособность после кратковременного жесткого теплового шока. Поэтому предлагается под термином «термотолерантность» понимать - способность организма выдерживать повреждающее воздействие теплового шока, а под термином «теплолюбивость» - способность к росту при температурах выше оптимальной. Поскольку эффективность производства АТФ, как правило, возрастает при повышении температуры, которое не оказывает мгновенного негативного эффекта на жизнеспособность растений и дрожжей, предполагается, что теплолюбивость сопровождается активацией окислительного фосфорилирования.

Сравнение скорости роста дрожжей при обычной температуре инкубации подтвердило литературные данные об обратной зависимости между скоростью роста и термотолерантностью. Чем медленнее скорость роста дрожжей при обычной температуре инкубации, тем лучше они переживают повреждающее действие теплового шока. Поскольку замедление скорости роста, как правило, сопровождается, повышением конститутивного синтеза БТШ, очевидно, что активация экспрессии генов БТШ в клетках дрожжей и, вероятно, в клетках растений может осуществляться не только в результате I повышения температуры, но и замедления или полной остановки роста.

Впервые проведено сравнение базовой термотолерантности i клеток ! дрожжей, различающихся по типу энергетического метаболизма. Результаты показали, что

I k 1 | способность дрожжей переживать повреждающее действие теплового шока определяется функциональной активностью митохондрий. Клетки облигатного аэроба D, vanrijiae

I I утилизируют как глюкозу, так и галактозу в ходе окислительного фосфоридирования. I

Соответственно, не обнаружено никаких различий в термотолерантности дрожжей D. vanrijiae, растущих на галактозе или глюкозе. Клетки факультативного анаэроба S. cerevisiae при росте на среде с глюкозой получают энергию преимуществен яо за счет сбраживания глюкозы до этанола, а на среде с галактозой используют окислительное фосфорилирование. При выращивании на среде с галактозой клетки S. cerevisiae лучше переживали тепловой шок, чем клетки этого же вида, выращенные на среде с глюкозой. Следовательно, клетки дрожжей, использующие окислительное фосфорилирование, оказываются гораздо более термотолерантными, чем такие же клетки, но использующие брожение. Повышенная термотолерантность клеток, использующих окислительный энергетический метаболизм, сопровождается повышением конститутивного синтеза Hspl04p и увеличением активности антиоксидантных ферментов. Таким образом, повышение уровня Hspl04p является одним из факторов повышения базовой термотолерантности клеток дрожжей, использующих окислительный энергетический метаболизм.

Впервые показано, что нарушение митохондриальных функций в клетках S. cerevisiae, использующих бродильный энергетический метаболизм, в результате утраты мтДНК (мутации petite) и в результате обработки митохондриальным ингибитором азидом натрия приводило к одному и тому же эффекту - повышению термотолерантности и подавлению генерации АФК при умеренном тепловом шоке. Наоборот, обработка азидом натрия усиливала продукцию АФК и снижала термотолерантность клеток S. cerevisiae, использующих окислительный энергетический метаболизм. Полученные данные указывают, что наблюдается прямая зависимость между усилением генерации АФК и гибелью клеток при умеренном тепловом шоке. Повышение генерации АФК происходит за счет функционирования митохондрий дрожжей, поскольку нарушение их функций либо усиливает, либо ингибирует продукцию АФК. Очевидно, что митохондрии, генерируя токсичные АФК, играют активную роль в запуске механизма гибели дрожжевой клетки при умеренном тепловом шоке.

Механизм генерации АФК при тепловом шоке в клетках дрожжей и растений не г ! известен. Однако с использованием изолированных митохондрии показано, что' генерация

АФК происходит тогда, когда на внутренней митохондриальной мембране повышается потенциал (мтДу). Полученные данные впервые показали, что повышение генерации АФК при тепловом шоке в целых клетках S. cerevisiae и A. thaliana зависит от гиперполяризации внутренней митохондриальной мембраны. Это дало ¡основание

I 1 и I предполагать причинно-следственную связь между мтДу и генерацией АФК. Такая

I j зависимость подтверждается следующими фактами. Во-первых, наблюдается прямая

I I зависимость при тепловом шоке между повышением мтДуу и усилением продукции АФК. Во-вторых, умеренный тепловой шок при 45°С вызывал повышение мтДу ' в клетках дрожжей, и это событие сопровождалось значительным усилением продукции АФК. Напротив, не было зарегистрировано повышения мтДу в клетках S. cerevisiae при жестком тепловом шоке 50°С, одновременно, продукция АФК в этих условиях лишь незначительно превышала контрольный уровень. В-третьих, подавление повышения мтДу при тепловом шоке в результате обработки азидом натрия и ДНФ ингибировало усиление продукции АФК в клетках S. cerevisiae, выращенных на среде с глюкозой. Тем не менее, зависимость между мтД\|/ и усилением генерации АФК не всегда выполняется. Обработка азидом натрия и ДНФ клеток дрожжей, выращенных на среде с этанолом, подавляла повышение мтД\|/ при тепловом шоке, но не ингибировала, а, наоборот, усиливала продукцию АФК. Предполагается, что образование АФК митохондриями в клетках растений и дрожжей происходит за счет функционирования внутренних и внешних НАДН дегидрогеназ, которые продуцируют АФК, как в результате перевосстановления компонентов дыхательной цепи (то есть, при повышении мтД\|/), так и в результате нарушения функционирования одного из ее компонентов (то есть, при снижении мтДу). Будет или нет наблюдаться образование АФК при тепловом шоке в результате функционирования внешних НАДН дегидрогеназ зависит от соотношения НАДН/НАД1" в цитозоле. В свою очередь, соотношение НАДН/НАД+ в цитозоле снижается при активации бродильного метаболизма, что вызывает снижение митохондриальной продукции АФК. Можно предположить, что одним из механизмов снижения продукции АФК при умеренном тепловом шоке в клетках дрожжей и растений и, соответственно, повышения термотолерантности является перенаправление метаболических путей, что приводит к снижению эффективности окислительного фосфорилирования и активации процессов гликолиза и брожения. Таким образом, если теплолюбивость сопровождается повышением эффективности производства АТФ митохондриями, то термотолерантность, вероятно, сопровождается снижением митохондриальной продукции АТФ.

Вероятно, гиперполяризация внутренней митохондриальной мембраны при I повышении температуры в клетках дрожжей и растений происходит за счет двух механизмов, которые не исключают друг друга. Первый, за счет повышения уровня »i 1 внутриклеточного Са2+ и последующей активации митохондриальных ферментов. Второй, за счет нарушения функционирования АДФ/АТФ транслокатора, в результате чего подавляется поступление АДФ в митохондрии, ингибируется синтез АТФ и повышается мтД\|/. |

Механизм гибели клеток животных различается в зависимости от интенсивности теплового воздействия. При умеренном тепловом шоке гибель клеток - происходит активным образом, в результате ПГК, а при более жестком тепловом воздействии клетки гибнут пассивно, в результате некроза. В данной работе впервые показано, что точно такая же ситуация наблюдается и в клетках дрожжей при тепловом шоке. Если при умеренном тепловом шоке 45°С гибель клеток дрожжей зависит от повышения мтДц/ и усиления генерации АФК, то при более жестком тепловом шоке 50°С этого не происходит. Мутация petite повышает жизнеспособность клеток S. cerevisiae после действия умеренного теплового шока и, наоборот, снижает жизнеспособность после жесткого теплового шока. Следовательно, механизмы гибели клеток дрожжей различаются в зависимости от интенсивности теплового воздействия.

Поскольку повышение генерации АФК митохондриями непременно предшествует развитию ПГК, а мутация petite ингибирует ПГК в клетках S. cerevisiae во всех описанных в литературе случаях, то результаты, приведенные выше, дали основания предполагать, что умеренный тепловой шок может активировать развитие ПГК. Последующие исследования подтвердили это предположение. На активный характер гибели клеток дрожжей при умеренном тепловом воздействии указывает способность ЦГ повышать термотолерантность. Такого эффекта не наблюдается при жестком тепловом шоке. Таким образом, гибель при умеренном тепловом шоке требует синтеза белков de novo, необходимых для развития ПГК. Динамика снижения жизнеспособности клеток дрожжей и A. thaliana после действия умеренного теплового шока также свидетельствует об активном характере гибели. Непосредственно после теплового шока клетки остаются живыми. Но во время восстановительного периода при обычной температуре клетки погибают, что сопровождается деградацией ДНК и усилением продукции АФК. Цитохром с, освобождаемый из митохондрий, является важным фактором развития ПГК. Соответственно, умеренный тепловой шок приводил к выходу1 цитохрома с из митохондрий в цитозоль в клетках S cerevisiae и A thaliana. Таким образом, вся совокупность полученных результатов указывает, что гибель клеток дрожжей и растений i -'I при умеренном тепловом шоке имеет признаки ПГК. Ранее было известно, что тепловой шок может вызывать развитие ПГК в культуре клеток растений и животных, но то, что такое же событие происходит в клетках S. cerevisiae показано впервые. >

Как правило, развитие ПГК у млекопитающих, дрожжей и ! растений сопровождается деполяризацией внутренней митохондриальной мембраны. ¡Считается, что при развитии ПГК выход белков, вызывающих развитие ПГК, из митохондрий i j происходит при открытии митохондриальной поры, что является результатом снижения мтДу. Как следует из результатов данной работы, развитие программы самоубийства в ! I клетках дрожжей и растении при тепловом шоке сопровождаемся ( значительным повышением мтДц/. На основании полученных результатов и ряда литературных данных предполагается, что гиперполяризация внутренней митохондриальной мембраны является индикатором ранней стадии ПГК, а на более поздней стадии наблюдается ее деполяризация.

Известно, что БТШ защищают клетки от гибели при тепловом шоке, восстанавливая денатурированные и агрегированные при нагревании белки. В то же время показано, что БТШ млекопитающих, модулируя выход про-апоптозных белков из митохондрий и активацию каспазного каскада могут как ингибировать, так и стимулировать развитие ПГК. В ходе данной работы впервые показано,' что БТШ могут выполнять аналогичную функцию в клетках растений и дрожжей. Нарушение функционирования Hspl04p снижало жизнеспособность клеток S. cerevisiae после умеренного и жесткого теплового шока, несмотря на различные механизмы гибели. Повышение количества БТШ в результате предварительного теплового стресса ингибировало все наблюдаемые признаки развития ПГК в клетках дрожжей и арабидопсиса, включая выход митохондриальных белков из митохондрий в цитозоль, повышение продукции АФК, а также снижало длительность гиперполяризации внутренней митохондриальной мембраны.

Изучение зависимости между индукцией синтеза БТШ и развитием ПГК при тепловом шоке позволило впервые показать, что когда клетки растений и дрожжей начинают погибать, синтез БТШ ингибируется. Следовательно, клетки дрожжей и растений не пытаются противостоять повреждению, синтезируя необходимые для своей защиты белки, когда тепловой шок переходит определенный порог. Таким образом, развитие ПГК и синтез БТШ - взаимоисключающие явления в растительной и дрожжевой клетке. 1

Известно, что выход Hsp60p из митохондрий может активировать развитие ПГК в клетках человека. Впервые показано, что аналогичное явление наблюдается в клетках i - i растений и дрожжей. Развитие ПГК при тепловом шоке в клетках A. thaliana и S. cerevisiae сопровождалось выходом митохондриального белка HspôOp из митохондрий в цитозоль. I

Поскольку подавление выхода Hsp60p из митохондрий приводило, к повышению

1 ! термотолерантности и ингибировало появление признаков ПГК, предполагается, что Hsp60p может стимулировать развитие ПГК в клетках дрожжей и растений. >

Впервые показано, что цитоплазматический белок HsplOlp A thaliana локализуется преимущественно в митохондриальной фракции белков после теплового стресса. Гомологичный ему белок Hspl04p также обнаруживается в митохондриальной фракции клеток S. cerevisiae. Такой результат позволяет предполагать, что Л/HsplOlp и IscHsp 104р ■ ! могут связываться с митохондриями при тепловом стрессе и модулировать их активность. Это предположение подтверждается тем, что инактивация гена HSP104 приводила к повышению дыхательной активности дрожжей S. cerevisiae. Предполагается, что ^¿Hsp 101 pÂScHsp 104р связывается с компонентами актинового цитоскелета, что влияет на процесс слияния и деления митохондрий, формирование митохондриальной сети и продукцию АФК митохондриями. i

Полученные результаты указывают, что не только Л/Hsp 101 pAScHsp 104р может модулировать активность митохондрий, но и митохондрии регулируют экспрессию генов БТШ. Установлена положительная связь между повышением уровня конститутивной экспрессии Hspl04p и активацией окислительного фосфорилирования. Нарушение дыхательной цепи митохондрий S. cerevisiae в результате мутации petite подавляет экспрессию Hspl04p при тепловом стрессе. Впервые показано, что нарушение митохондриальных функций в результате обработки митохондриальными ингибиторами и разобщителями подавляет активацию экспрессии генов БТШ при тепловом стрессе в клетках дрожжей и растений. Способность митохондрий регулировать экспрессию генов Hspl04p в клетках S. cerevisiae при тепловом стрессе, вероятно, зависит от функционирования транскрипционных факторов Msn2p и Msn4p. В клетках с пониженной активностью цАМФ-ПКА (то есть тогда, когда факторы Msn2/Msn4p находятся постоянно в активированном состоянии), обработка азидом натрия не приводила к ингибированию тепловой индукции синтеза Hspl04p. Показано, что способность митохондриальных ингибиторов и разобщителей подавлять активацию генов БТШ при тепловом стрессе зависит от их способности подавлять повышение мтА\|/. Антимицин А и цианид не подавляли повышение мтДу при тепловом стрессе в клетках S. cerevisiae, использующих бродильный энергетический метаболизм, одновременно отсутствовал подавляющий эффект этих агентов на тепловую индукцию синтеза Hspl04p и развитие индуцированной термотолерантности. Напротив, в клетках S. cerevisiae, использующих окислительный энергетический метаболизм, способность антимицина А и цианида подавлять повышение мтДц/ при тепловом стрессе сопровождалась ингибированием индукции синтеза! Hspl04p и i индуцированной термотолерантности после теплового стресса, i Таким! образом, наблюдается прямая зависимость между гиперполяризацией внутренней митохондриальной мембраны и активацией экспрессии генов БТШ npii тепловом стрессе. Агенты, которые при данных экспериментальных условиях подавляют повышение мтДу, одновременно подавляют тепловую активацию экспрессии генов БТШ. ^ Подобная зависимость предполагает наличие причинно-следственной связи между этими явлениями

- [ 4 и указывает, что повышение мтДц/ в клетках дрожжей и растении при тепловом стрессе является необходимым этапом в активации экспрессии генов БТШ. Следует подчеркнуть, что данная закономерность выполняется только в условиях теплового 'стресса, !поскольку деполяризация митохондриальной мембраны при обычной температуре инкубации также может приводить к активации экспрессии генов БТШ.

Результаты, показывающие, что обработка экзогенной салициловой кислотой подавляет повышение мтД\|/ при тепловом стрессе в культуре клеток A. thaliana и одновременно ингибирует тепловую экспрессию генов БТШ, указывают, что регулятором уровня БТШ у растений in situ может быть изменение в уровне эндогенной салициловой кислоты.

Фторид натрия, являющийся одним из компонентов атмосферных выбросов, подавляет повышение мтДу при тепловом стрессе в культуре клеток A. thaliana и ингибирует тепловую экспрессию генов БТШ. Следовательно, этот загрязнитель, в условиях, когда он сам не оказывает отрицательного действия, может подавлять адаптивную реакцию растений на экстремальные факторы внешней среды, ингибируя экспрессию генов БТШ.

В клетках S cerevisiae при самых различных стрессовых воздействиях повышается уровень кальция в цитозоле. В этом процессе участвуют низкоаффинная и высокоаффинная системы транспорта Са2+ через плазматическую мембрану. Известно, что тепловой стресс вызывает временное повышение уровня Са2+ в цитозоле растений и животных. Существуют косвенные данные, указывающие, что повышение [Са ]cyt наблюдается при тепловом стрессе и в клетках S cerevisiae. Чтобы установить зависимость между повышением уровня кальция в цитозоле и гиперполяризацией

2+ внутренней митохондриальной мембраны, был исследован механизм повышения [Са ]cyt в клетках S. cerevisiae при тепловом стрессе. Впервые показано, что в условиях

2+ пониженной концентрации питательных веществ и Са в среде инкубации, повышение [Ca2+]cyt при тепловом стрессе определятся функционированием высокоаффинной системы транспорта Ca2+ через плазматическую мембрану. Нарушение функционирования известных компонентов этой системы, белков Midlp и Cchlp, подавляло этот процесс.

Когда в среде инкубации концентрация питательных веществ и Са2+ повышена, поступление Са2+ в цитозоль при тепловом стрессе не зависит от присутствия Midlp и Cchlp, и, вероятно, определяется функционированием низкоаффинной| системы транспорта Са2+. Анализ максимумов повышения [Ca2+]cyt позволил предположить, что существует и третий механизм поступления кальция в цитозоль дрожжевой клетки. Этот

2+ 1 I механизм, вероятно, определяется поступлением Са в цитозоль из клеточных ! компартментов.

Повышение [Ca2+]mit в митохондриях млекопитающих стимулирует активность дыхательных ферментов, приводит к повышению сопряжения между дыханием и окислительным фосфорилированием и, соответственно, повышает мтД\|/. Система транспорта Са2+ в митохондрии функционирует и у растений. Но неизвестно, какой эффект оказывает повышение [Са ]mit на митохондрии растений. В клетках дрожжей отсутствует система активного транспорта кальция в митохондрии. Несмотря на это обстоятельство, полученные данные однозначно свидетельствуют, что нарушение гомеостаза внутриклеточного кальция оказывает значительное влияние на функционирование митохондрий S cerevisiae. Утрата белков Midlp и Cchlp, определяющих транспорт кальция через плазматическую мембрану, приводит к снижению потенциала на внутренней митохондриальной мембраны и к подавлению дыхательной активности в клетках S. cerevisiae. Представленные данные позволяют предполагать, что

2+ повышение [Са ]cyt в дрожжевой клетке может косвенно, модулируя функционирование сигнальных систем TOR и цАМФ-ПКА, влиять на активность митохондрий.

В целом, вся совокупность полученных результатов позволяет предложить следующую упрощенную гипотетическую схему участия дрожжевых и растительных митохондрий в ответе клетки на тепловое воздействие. Повышение температуры вызывает открытие кальциевых каналов на плазматической мембране. Что это за каналы и как они функционируют неизвестно. Полученные в данной работе результаты показывают, что при тепловом стрессе, в условиях низкой концентрации питательных веществ, транспорт кальция через плазматическую мембрану S cerevisiae зависит от белков Midlp и Cchlp.

Кратковременное повышение [Ca2+]cyt как непосредственно, так и косвенно активирует экспрессию генов БТШ. Длительное повышение уровня кальция в цитозоле может иметь неблагоприятные последствия для клетки. Поэтому из цитозоля кальций транспортируется либо в митохондрии, либо в другие клеточные компартменты. Поступление кальция в митохондрии и, вероятно, в другие клеточные компартменты

2+ I митохондриальными ингибиторами, то транспорт Са в митохондрии подавляется, он аккумулируется в цитозоле до критического уровня, при котором активация экспрессии БТШ не происходит. В свою очередь, обработка митохондриальными ингибиторами при обычной температуре инкубации, повышает уровень кальция в цитозоле за счет его выхода из клеточных компартментов, что в отсутствии теплового стресса также может приводить к активации экспрессии генов БТШ. !

Далее, кальций, либо косвенным образом, модулируя активность сигнальных систем, либо непосредственно, оказавшись в митохондриях, вызывает .повышение уровня I

НАДН. Повышенный уровень НАДН стимулирует транспорт электронов по дыхательной цепи, усиливает продукцию АТФ, что в результате приводит к гиперполяризации внутренней митохондриальной мембраны и усилению продукции АФК. Повышение генерации АФК до определенного уровня также играет важную роль в активации экспрессии генов БТШ. Митохондрии, таким образом, выступают как передатчик и реле кальциевого сигнала. Если повышение температуры достигает определенного порога, то нарушается функционирование АДФ/АТФ транслокатора. В результате поступление АДФ в митохондрии прекращается, синтез АТФ ингибируется, что приводит к чрезмерному повышению мтДу. В этом состоянии генерация АФК митохондриями достигает критического уровня, экспрессия БТШ ингибируется и активируются процессы гибели. Если тепловой шок не прекращается, повышенный уровень кальция в митохондриях и непрекращающаяся генерация АФК вызывают открытие митохондриальных пор, деполяризацию внутренней митохондриальной мембраны, выход цитохрома с и ШрбОр из митохондрий в цитозоль, и, в конечном счете, активацию программы самоубийства.

Следовательно, программа адаптации, включающая в себя активацию экспрессии генов БТШ, и программа самоубийства имеют ряд общих знаменателей при повышении температуры: повышение уровня кальция в цитозоле, гиперполяризация внутренней митохондриальной мембраны и усиление продукции АФК. Митохондрии, таким образом, модулируя уровень кальция в цитозоле и продукцию АФК, определяют жизнь и смерть клетки при тепловом шоке.
