Актуальность проблемы. Популяционный уровень является наименее разработанным в области сохранения биоразнообразия (Миркин, Наумова, 1998; Fallon, 2007) - проблемы, отнесенной в последние десятилетия в разряд глобальных (Розенберг, 1998). По этой причине актуальным является исследование факторов, определяющих формирование и поддержание популяционных структур во времени и в пространстве. Древесно-кустарниковые растения в этом отношении выделяются своей спецификой (Политов и др., 2006), что необходимо учитывать при разработке мер сохранения природных популяций методами ex situ и in situ (Мамаев и др., 1994). У каждого вида в течение многих поколений под влиянием комплекса биоэкологических и средовых факторов складывается определенная структура внутривидовой изменчивости, игнорирование чего может привести к множеству отрицательных последствий (Алтухов, 1995).

Обитающие на Южном Урале виды можжевельника, ранее практически не изученные в популяционно-экологическом отношении, являются важными компонентами чрезвычайно уязвимых природных экосистем региона и имеют огромное значение для сохранения их целостности (Мулдашев, Кучеров, 2005). Среди них особо выделяется можжевельник казацкий - реликт Южного Урала, образующий, как и другие виды можжевельников (Коропачинский, 1983), множество морфологических и экологических форм. Ранее он был включен во 2-е издание Красной книги Республики Башкортостан.

Цель и задачи исследований. Цель данной работы - исследование популяционной структуры можжевельника казацкого Juniperns sabina и разработка принципов его охраны на Южном Урале. Для ее выполнения решались следующие задачи: 1) выявить полиморфные аллозимные локусы, информативные при описании структуры популяций; 2) исследовать популяционную структуру и факторы ее формирования, поддержания и устойчивости на популяционном уровне; 3) изучить клоновый состав в местообитаниях древовидной формы; 4) разработать принципы сохранения ресурсов можжевельника казацкого на Южном Урале на популяционной основе.

Научная новизна работы. Выявлены полиморфные изоферментные системы Juniperus sabina L., J. sibirica Burgsd. и J. communis L., являющиеся информативными маркерами популяционного разнообразия данных видов. Установлено, что можжевельник казацкий в пределах Южного Урала обладает сравнительно высоким уровнем популяционного разнообразия. Его местообитания' обладают относительно невысокой межпопуляционной дифференциацией по частотам аллелей и сравнительной близостью аллельных частот в пределах исследованного региона. Популяционные структуры сформированы под влиянием состава и величины клонов, имеющихся в местообитаниях.

Практическая значимость работы. Полученные результаты могут быть использованы в качестве научного обоснования практических мер по сохранению видов можжевельника на Южном Урале, в том числе в качестве базы для разработки соответствующих нормативных документов в области охраны природы ведомствами Республики Башкортостан. Материалы диссертации используются при ведении образовательной деятельности в Сибайском институте (филиале) Башкирского государственного университета и Зауральском филиале Башкирского государственного аграрного университета по дисциплинам «Экология», «Ботаника», «Популяционная генетика», «Экологическая генетика».

Основные положения, выносимые на защиту: 1. Аллозимные маркеры являются эффективным средством при решении проблем изучения, сохранения и рационального использования растительных ресурсов видов можжевельника на Южном Урале - выявления популяционных структур, клонового разнообразия, идентификации генотипов.

2. Можжевельник казацкий на Южном Урале характеризуется относительно высоким сходством популяционных структур.

3. Уровни популяционного разнообразия и дифференциации популяций определяются в первую очередь составом клонов и долей их участия в местообитаниях.

4. Местообитания с наибольшим клоновым разнообразием представляют первоочередной интерес при выборке объектов сохранения генофонда in situ и при отборе образцов для сохранения ресурсов вида ex situ. Актуальна охрана и рациональное использование отдельных уникальных объектов, а для повышения эффективности этих работ - изучение популяционной структуры и факторов, ее формирующих.

Обоснованность выводов и достоверность результатов работы обеспечены достаточно большими объемами полевого материала и проведенных лабораторных экспериментов, использованием современных методов исследований, адекватных поставленным задачам и применением современных методов статистического анализа.

Личное участие автора. Работа выполнялась с 2005 года по 2009 г.г. в рамках плановой научно-исследовательской деятельности Сибайского филиала Академии наук Республики Башкортостан, по гранту АН РБ «Изучение генофонда можжевельника казацкого в Башкирском Зауралье и разработка мер по его сохранению» (2006-2007 г.г.). Автором лично проведены полевые и лабораторные исследования, осуществлены анализ и обобщение результатов.

Апробация работы. Основные результаты диссертации были представлены на международной научно-практической конференции «Современные проблемы природопользования, охотоведения и звероводства» (Киров, 2007), всероссийской научно-практической конференции «Проблемы и перспективы развития инновационной деятельности в агропромышленном комплексе» (Киров, 2007), международной конференции «Биоразнообразие и биоресурсы Урала и сопредельных территорий» (Оренбург, 2008), региональных конференциях «Неделя науки» (Сибай, 2005, 2006, 2007, 2008, 2009), международной научно-практической конференции «Актуальные проблемы дендроэкологии и адаптации растений», посвященной 80-летию со дня рождения профессора Ю.З. Кулагина (Уфа, 2009).

Публикации. Основные результаты диссертации приведены в 10 печатных работах, в т.ч. 2 - в изданиях, рекомендованных ВАК РФ для публикации результатов диссертационных работ.

Структура и объем работы. Диссертация состоит из введения, 4 глав, выводов, списка литературы, включающего 219 наименований публикаций, в том числе 165 - зарубежных авторов. Основной текст изложен на 125 страницах, включает 10 таблиц и 22 рисунка.

1. Для описания структуры популяций Juniperus sabina L. информативными маркерами являются аллозимы полиморфных локусов Gdh-1, Dia-1, Dia-2, Est-1, Fdh-1, Lap-1, Lap-2, Skdh-1 и 6Pgdh-l.

2. Популяции видов можжевельника на Южном Урале обладают высоким уровнем разнообразия аллозимных локусов. Наибольший полиморфизм (в среднем на полиморфный локус выявлены 3.4±0.4 аллелей) выявлен у Juniperus sabina L., по составу и встречаемости аллозимов он существенно отличается от Juniperus communis L. и Juniperus sibirica Burgsd.

3. Для Juniperus sabina L. характерна относительно невысокая межпопуляционная дифференциация - коэффициент межвыборочной подразделенности /^.=0.113, значения расстояния М. Нея D между парами популяций изменяются от 0.002 до 0.058. По частотам аллозимов вид существенно отличается от Juniperus communis L. (F?r=0.555, D=0.878), различия которого с Juniperus sibirica Burgsd. не выходят за пределы межпопуляционного уровня.

4. Популяционное разнообразие Juniperus sabina L. в отдельных местообитаниях испытывает большое влияние состава, встречаемости и размеров клонов. Существуют популяции с высоким генотипическим разнообразием, перспективные для охраны в природных условиях и рационального использования ресурсов растения ex situ.

5. Генофонд популяций Juniperus sabina L. на Южном Урале находится в относительно благополучном состоянии и не требуется принятия срочных мер по его охране на видовом уровне - полученные данные свидетельствуют о правомерности решения об исключении вида из последнего издания Красной книги Республики Башкортостан. Достаточным является проведение работ по выделению и охране в природных местообитаниях эталонных участков и популяций с наиболее высоким полиморфизмом в форме малых оопт.

6. Сохранение ресурсов Juniperus sabina L. необходимо в первую очередь осуществлять на популяционном уровне - в популяциях с нетипичным генофондом, а также в местообитаниях с уникальным формовым разнообразием, что отчетливо выявлено в случае с древовидным можжевельником казацким.
