8. Выводы

1. Получены и охарактеризованы стабильные линии гибридных клеток, продуцирующие моноклональные антитела к антигенам вируса чумы плотоядных (1 клон), вируса парвовирусного энтерита плотоядных (15 клонов) и вируса инфекционного гепатита собак (3 клона).

2. МАт к вирусу чумы плотоядных, секретируемые клоном 1В6, к вирусу парвовирусного энтерита - клон 1Д8 - и к вирусу инфекционного гепатита собак - клон 5С12 - проявляют наибольшую активность в ТФ ИФА, строго специфичны по отношению к гомологичному вирусу и не вступают в реакцию с гетерологичными антигенами.

3. Разработанная с использованием МАт клона 1В6 тест-система на основе ТФ ИФА для идентификации антигена вируса чумы плотоядных является специфичной и позволяет выявлять антиген при титре вируса не менее 1,21ёТЦД50/мл.

4. Тест-система на основе МАт 1Д8 и ТФ ИФА для идентификации антигена вируса парвовирусного энтерита плотоядных обеспечивает его выявление при титре вируса в РГА не менее 4 ГАЕ.

5. Тест-система на основе ТФ ИФА и МАт клона 5С12 для выявления антигена вируса инфекционного гепатита собак позволяет выявлять его в исследуемом материале при титре не менее 3,5 ^ ТИД50/мл.

6. Разработанные тест-системы ТФ ИФА на основе МАт к В 411, ВГС и возбудителю парвовирусного энтерита плотоядных позволяют выявлять специфический антиген в органах больных и павших животных различных видов, в вируссодержащих культуральных жидкостях, вакцинных препаратах и могут быть использованы как для прижизненной и посмертной диагностики указанных болезней, так и для технологического контроля сырья при производстве вакцинных препаратов.

7. Созданные на основе МАт клонов 1В6ДД8 и 5С12 диагностические тест-системы для обнаружения антител к вирусам чумы, парвовирусного энтерита плотоядных и инфекционного гепатита собак методом конкурентного ТФ ИФА позволяют выявлять специфические антитела в сыворотках больных, переболевших животных, в биологических препаратах (глобулины, ле

123 чебные сыворотки) вне зависимости от видовой принадлежности сывороток и могут быть использованы для ретроспективной диагностики этих болезней, для определения напряженности колострального, поствакцинального и постинфекционного иммунитета и контроля биологических препаратов.

На основе полученных в ходе настоящей работы гибридом, продуцирующих моноклональные антитела к антигенам вирусов чумы плотоядных, инфекционного гепатита плотоядных и парвовирусного энтерита плотоядных, разработан сэндвич-вариант ТФ-ИФА для выявления антигена вирусов чумы, энтерита и гепатита плотоядных.

Данные тест-системы могут применяться для выявления вирусных антигенов в:

-патологических материалах,

-культурах клеток после вирусовыделения и контроля биологических препаратов,

-биологиических препаратах, содержащих антигены данного вируса (определение наличия и содержания антигена в культурах клеток, гомогена-тах органов, нативных, лиофильно-высушенных, жидких инактивированных биопрепаратах - вакцинах),

- биологических препаратах, которые не должны быть контаминирова-ны данными вирусами (сырье для производства биопрепаратов, культуры клеток).

На основе полученных в ходе настоящей работы гибридом, продуцирующих моноклональные антитела к антигенам вирусов чумы плотоядных, инфекционного гепатита плотоядных и парвовирусного энтерита плотоядных разработан конкурентный сэндвич-вариант ТФ-ИФА для выявления поликло-нальных антител, специфичных к антигенам вирусов чумы, энтерита и гепатита плотоядных.

Данные тест-системы могут применяться для:

- выявления антител, специфичных к вирусным антигенам в сыворотках больных, переболевших, подозреваемых в заражении животных,

- определения напряженности колострального, поствакцинального и постинфекционного иммунитета

- контроля биологических препаратов, содержащих антитела к антигенам вирусов плотоядных (сыворотки и глобулины лечебные и диагностические),

- контроля биопрепаратов и сырья, которые не должны содержать такие антитела (сыворотки для культур клеток, фетальные сыворотки и т.д.).

Разработана лабораторная технология производства и схемы применения тест-систем для указанных целей, отраженные в 18 нормативно-технических документах, утвержденных в установленном порядке и представленных в приложении к диссертационной работе.

403. Для проведения испытаний метода ИФА на основе моноклональных антител для диагностики чумы плотоядных будут представлены: ,- моноклональные иммуноглобулины мыши, специфичные к вирусу чумы плотоядных;- моноклональные антитела меченые перрксидазой хрена.

404. Методы испытаний и оценка результатов 4.1. Испытание ДАС-ИФА (двойной антительный "сэндвич"-вариант твердофазного иммуноферментного анализа) для выявления специфического антигена на специфичность и чувствительность.

405. В процессе испытаний учет и оценку результатов проводят визуально и фотометрически.

408. О межлабораторных комиссионных испытаниях штамма гибридной линии клеток 1В6, секретирующей моиоклональныеантитела к вирусу чумы плотоядных, по определению их пригодности для выявления антигена и противовирусных антител.

409. Целью испытаний было определение специфичности указанных моноклональных антител и их пригодности для диагностических исследований для выявления антигена вируса чумы плотоядных и сывороточных антител к данному вирусу.

410. Комиссионные испытания проведены 30 января 1998 года на базе лаборатории «Гибридомной биотехнологии» в соответствии с программой, утвержденной директором 30,01.98.4. Материалы и методы.

411. Выявление антигенов проводили согласно программе испытаний

412. Выявление специфических антител согласно программе комиссионных испытаний.5. Результаты испытаний.

413. Результаты выявления антигенов ВЧП с помощью монокло-нальных антител 1В6 определяли в «сандвич»- варианте ИФА в шифрованном опыте.'Результаты исследования представлены в таблице.
