Конец двадцатого века характеризуется бурным развитием исследований в области наук о материалах. Особое внимание при этом уделяется материалам, способным к обратимой регуляции своих физико-химических свойств в ответ на тот или иной физический или химический сигнал-стимул (свет, температура, рН, ионная сила, электрические или магнитные поля и т.д.). Значительный интерес в плане конструирования подобных материалов представляют фермент-содержащие стимул-чувствительные полимерные матрицы. Активность ферментов, иммобилизованных в подобные полимерные материалы, может определяться изменениями свойств матрицы, вызываемых внешним сигналом, и, таким образом, становится зависимой от его интенсивности. В литературе появился даже термин «умные» (smart, intelligent) материалы. Своеобразным индикатором актуальности данной тематики служит тот факт, что первая международная конференция по «умным» материалам была организована в Японии в 1989 г., а в 1996 г. в мире проводилось восемнадцать конференций аналогичного содержания. Температурно-чувствительные полимеры (и гели на их основе) являются одним из интереснейших примеров стимул-чувствительных материалов, обратимо изменяющих свой объем в результате фазового перехода полимерных цепей.

Механизм фазового перехода термочувствительных гидрогелей на молекулярном уровне определяется переходом «клубок-глобула» молекул полимера. Конформация полимерной цепи определяется «качеством растворителя» % = AF/kT, где AF - свободная энергия взаимодействия полимер-растворитель, кТ - энергия собственного теплового движения полимерных цепей. В плохих растворителях (х>:>1) энергия собственного теплового движения полимерных цепей кТ много меньше, нежели чем энергия AF, связанная с взаимодействием полимер-растворитель. Для хороших растворителей (^«1) ситуация обратная. Таким образом, фазовый переход «клубок-глобула» полимерной цепи при переходе от «хорошего» растворителя к «плохому» может быть вызван путем изменения температуры и/или изменением состава растворителя (внешний стимул). В ответ на стимул происходит изменение гидрофильно-гидрофобного баланса матрицы в результате изменения энергетических характеристик взаимодействий полимера с растворителем. При этом отметим, что движущей силой фазового перехода синтетических термочувствительных полимеров (переход «клубок-глобула») является энтропийный фактор, и именно энтропийный фактор определяет процессы фолдинга природных полимеров - белков. Сходство физических причин, вызывающих эти два на первый взгляд разные процессы, уже привлекло внимание исследователей и позволило выдвинуть предположение о том, что фазовый переход в термочувствительных полимерах может рассматриваться как аналог «холодной» ренатурации небольших глобулярных белков [376, 377]. Но тогда каким образом будет реализоваться взаимодействие между полимерной матрицей, претерпевающей фазовый переход, и иммобилизованным в нее ферментом? Каковы будут кинетические проявления данного процесса? Понятно, что только понимание физических причин таких взаимодействий (в том случае, конечно, если они действительно реализуются) даст возможность практического использования фермент-содержащих стимул-чувствительных полимерных гидрогелей.

Цель и задачи исследования. Настоящая работа была направлена на создание содержащих ферменты стимул-чувствительных полимерных гидрогелей с обратимо регулируемой активностью биокатализатора, а также на выявление механизмов такой регуляции. В этой связи изучались кинетические закономерности функционирования иммобилизованных гидролитических ферментов при фазовом переходе (коллапсе) матрицы-носителя под действием различных стимулов физической и/или химической природы.

Для достижения поставленной дели в работе было необходимо:

1. Исследовать взаимосвязь относительной степени гидратации стимул-чувствительной матрицы и активности иммобилизованных в ней ферментов под действием:

- температуры;

- концентрации органического растворителя в реакционной смеси;

- совместного действия этих стимулов.

2. Провести сравнительный анализ результатов по влиянию изучаемых стимулов на кинетическое поведение нативных ферментов и их иммобилизованных в стимул-чувствительную и неколлапсирующую полимерные матрицы препаратов.

3. Выявить роль структуры субстрата и жесткости конформации белка в наблюдаемом кинетическом ответе биокатализатора на интенсивность изучаемого стимула.

Особое внимание было уделено обратимости кинетических эффектов, проявляющихся при функционировании иммобилизованных ферментов в ходе фазового перехода полимера.

выводы

1. Процессы гидролиза под действием иммобилизованных в стимул-чувствительный поли-Ы-изопропилакриламидный гель ферментов с изменением температуры и/или содержания органического растворителя демонстрируют свойства, отличающиеся от свойств процессов, протекающих в растворе или неколлапсирующей полимерной матрице. Характер наблюдаемых кинетических изменений позволил уточнить ряд существенных деталей механизма ферментативного гидролиза.

2. Показано, что в водной среде в области температур ниже Ткр поли-НИПАА геля кинетические закономерности реакций гидролиза определяются температурными свойствами иммобилизованного фермента, а в области выше Ткр геля - свойствами поли-НИПАА матрицы. Имеется четкая связь между относительной степенью гидратации полимерной матрицы и ферментативной активностью. Факторы, регулирующие относительную степень гидратации гелевой матрицы, аналогичным образом регулируют активность иммобилизованного в ней фермента.

3. Продемонстрировано, что основной особенностью ферментов, иммобилизованных в поли-НИПАА гель, является аномальная зависимость скорости процесса гидролиза с изменением температуры - во всех случаях появляются температурные интервалы, в которых увеличение температуры приводит к уменьшению активности иммобилизованных препаратов. Показано, что фазовый переход поли-НИПАА геля слабо влияет на температурное поведение равновесных параметров ферментативной реакции иммобилизованного биокатализатора, и наблюдаемое поведение иммобилизованных препаратов связано исключительно с изменением кинетических характеристик ферментативного процесса (каталитической константы иммобилизованного фермента).

4. Установлено, что специфическое взаимодействие органического растворителя с нативным а-химотрипсином происходит в S2-catoe сорбционного центра фермента, что и сказывается на кинетике гидролиза. Закрепление конформации белка путем ковалентной иммобилизации, равно как и связывание гидрофобных N-заместителей субстрата в 82-сайте, протектирует активный центр а-химотрипсина от специфического влияния органического растворителя. Кинетическая схема, описывающая гидролиз N-ацетил замещенных производных L-тирозина «конформационным изомером» а-химотрипсина, удовлетворительно согласуется с экспериментальными данныим.

5. Продемонстрировано, что в изотермических условиях в смесях вода-ДМСО кинетические закономерности для иммобилизованных в поли-НИПАА ферментов сходны с закономерностями, полученными для нативных ферментов. На этом основании выдвинуто предположение о сходных причинах влияния органического растворителя и полимерных цепей стимул-чувствительной матрицы при ее коллапсе на активный центр биокатализатора.

6. Предложен механизм регуляции активности иммобилизованного в стимул-чувствительной полимерной матрице фермента фазовым переходом носителя, заключающийся в специфических взаимодействиях полимерных цепей геля, гидрофобизующихся в ходе коллапса, с активным центром биокатализатора (82-сайтом сорбционного участка в случае а-химотрипсина).

7. Показано, что в целом система стимул-чувствительная полимерная матрица/иммобилизованный фермент приобретает возможность регуляции активности иммобилизованного биокатализатора посредством изменения степени относительного водосодержания полимерной матрицы. Продемонстрирована обратимость всех наблюдаемых кинетических эффектов. На основании полученных данных предложен и реализован ряд практических применений стимул-чувствительных систем:

- синтезирован и охарактеризован свето- и термочувствительный полимер с фотоконтролируемой температурой преципитации/растворения для иммунодиагностических целей;

- с помощью термочувствительных гелей отработаны методики концентрирования высокомолекулярных белков и отделение их от низкомолекулярных фракций в мягких условиях с быстрой регенерацией полимерного сорбента;

- получены термочувствительные фермент-содержащие мембраны, обладающие стабильным и воспроизводимым ответом, применение которых позволяет резко сократить время аналитического цикла биосенсоров;

- разработана модель ферментных сенсорных элементов триггерного типа, дающих ответ только при превышении задаваемой условиями определения концентрации субстрата.
