Взаимодействие вирусов с клеткой - одна из наиболее увлекательных проблем вирусологии. Вирусы являются доклеточными формами жизни, которые используют клеточные энергетические ресурсы для репликации собственной нуклеиновой кислоты. Кодируя ограниченный набор генов, вирусы выработали изощрённые стратегии для поражения клеток и ловко подстраивают функционирование клеточных систем для собственных нужд. Клетки-хозяева, в свою очередь, приобрели противовирусные программы, призванные уничтожать чужеродную нуклеиновую кислоту и защищать клетку от вторжения патогенов. Борьба «вирус-клетка» длится со времён обособления данных форм жизни и обеспечивает их ко-эволюцию; следить за развитием этой борьбы -интригующая и зачастую жизненно-важная для всего человечества задача учёных-вирусологов.

Настоящая работа посвящена одному из аспектов взаимодействия с клеткой цитоплазматических РНК-содержащих вирусов семейства Picornaviridae, а именно нарушению ядерно-цитоплазматического транспорта при инфекции. Система транспорта обеспечивает обмен макромолекул между ядром и цитоплазмой через белковые комплексы ядерных пор, пронизывающие ядерную оболочку. Цикл репродукции пикорнавирусов протекает в цитоплазме, и, на первый взгляд, данная группа вирусов не нуждается во взаимодействии с клеточной системой транспорта. Около 10 лет назад было сделано открытие, показавшее, что полиовирус (наиболее хорошо изученный и печально известный представитель семейства) нарушает обмен макромолекул между ядром и цитоплазмой (Belov et al., 2000). За последние годы исследователи значительно продвинулись в понимании механизмов нарушения транспорта полиовирусом: ключевую роль играет вирусная протеаза 2А, которая расщепляет белки ядерной поры и, тем самым, нарушает барьерные свойства ядерной оболочки (Gustin and Sarnow, 2001; Belov et al.

2004; Park et al., 2008). Тем не менее, на многие вопросы до сих пор нет ответа. Например, каково биологическое значение нарушения ядерного транспорта для пикорнавирусов? Является ли взаимодействие с транспортной системой абсолютно необходимым или предпочтительным условием репликации полиовируса? Уникально ли данное свойство для полиовируса или консервативно для всех представителей семейства?

В поисках ответов на некоторые из этих вопросов, в настоящей диссертационной работе нами было продолжено исследование влияния Picornaviridae на клеточный транспорт. В качестве объекта исследований мы выбрали группу Cardiovirus, у представителей которой белок 2А лишён протеолитической активности, чем данная группа и представляла для нас особый интерес. Диссертация построена по общепринятому плану и состоит из обзора литературы, экспериментальной части, а также списка литературы. В разделе 2.1 обзора литературы кратко сформулированы современные представления о механизмах функционирования и регуляции ядерно-цитоплазматического обмена. В следующем разделе (2.2) приведены примеры разносторонних взаимодействий ряда вирусов с системой ядерно-цитоплазматического транспорта. В разделе 2.3 изложена краткая характеристика семейства Picornaviridae, особое внимание уделено известным данным о нарушении клеточного транспорта при инфекции некоторыми пикорнавирусами. Экспериментальная часть состоит из разделов «Материалы и методы», «Результаты» и «Обсуждение» и включает описание основных результатов, полученных в данной диссертационной работе, а также их интерпретацию.

7. ВЫВОДЫ

1. Показано, что кардиовирусы нарушают ядерно-цитоплазматический обмен клетки: инфекция сопровождается нарушением барьерных свойств ядерной оболочки и перераспределением макромолекул между ядром и цитоплазмой.

2. Установлено, что ключевую роль в нарушении барьерных свойств ядерной оболочки играет лидерный (L) белок кардиовирусов. Выявлены участки L-белка, важные для выполнения данной функции (мотив Zn-пальца и сайт фосфорилирования). ♦

3. Подавление активности клеточных росковитин- и оломоуцин-чувствительных протеинкиназ нарушает способность кардиовирусов изменять проницаемость ядерной оболочки.

4. Установлена взаимосвязь между нарушением кардиовирусами ядерно-цитоплазматического транспорта и гиперфосфорилированием белка транспортного канала ядерной поры Nup62.

5. Охарактеризовано влияние кардиовирусов на ядерную пору: при инфекции не происходит существенных изменений набора FG-содержащих нуклеопоринов и * разрушения общей архитектуры комплекса поры.

6. Обнаружено сходство между нарушением барьерных свойств ядерной оболочки при кардиовирусной инфекции и в профазе митоза.

7. Предложен гипотетический сценарий нарушения ядсрпо-цитоплазматического транспорта кардиовирусами: вирусный L-белок активирует клеточные протеинкиназы, фосфорилирующие нуклеопорины; ядерные поры при этом не разрушаются, но повышается их проницаемость для пассивной диффузии макромолекул.
