До недавнего времени считалось, что механизмы инициации трансляции у прокариот и эукариот кардинально различны. В последнее десятилетие, однако, появились данные, которые похоже начали "перекидывать мостик" между механизмами инициации трансляции у эукариот и прокариот. Обнаружено, что несмотря на отсутствие гомологии в первичной структуре и на различные размеры, эукариотические факторы инициации elFl, elFIA и eIF5B являются структурными и функциональными аналогами бактериальных IF3, IF1 и IF2. Кроме того, оказалось, что для некоторых эукариотических мРНК, прежде всего вирусного происхождения, несомненно возможен механизм связывания рибосомы на их специфических внутренних участках - т.н. IRES-элементах (от англ Internal Ribosome Entry Site). Следует подчеркнуть, что если в случае прокариотических мРНК внутренняя посадка рибосомы требует только белка S1 и/или наличия в них последовательности Шайна-Дальгарно, а также участия в процессе инициации только трех факторов инициации (IF1, IF2 и IF3), то IRES-элементы это протяженные участки с развитой вторичной структурой. Классические IRES-элементы РНК пикорнавирусов содержат высоко специфические сайты связывания для инициирующих факторов, чего нет в бактериальных мРНК. Для инициации трансляции на таких элементах РНК пикорнавирусов требуются почти все эукариотические факторы инициации трансляции (за исключением кеп-связывающего фактора eIF4E), и ряд вспомогательных мРНК-связывающих белков. Гораздо большее сходство с бактериальной системой обнаруживают IRES-элемент из РНК вируса гепатита С (Hepatitis С Virus, HCV), который для образования 40S инициирующего комплекса (48S комплекс) обходится всего двумя факторами инициации, eIF2 и eIF3, и таким образом не требует ни кепа, ни процесса сканирования цепи мРНК при выборе инициирующего кодона. До недавнего времени IRES-элемент HCV, не считая совсем экзотического случая IRES-элемента РНК вируса паралича сверчка {Cricket Paralysis Virus, CrPV), где не нужны никакие факторы инициации трансляции и даже инициаторная Мет-тРНК, был по-существу единственным примером, показывающим сходство в механизмах связывания мРНК у про- и эукариот.

Автор диссертации и его коллеги исходили из предположения, что сравнение не только структур рибосом или отдельных факторов трансляции, но и самих механизмов трансляции у бактерий и эукариот может быть весьма полезным для более глубокого понимания этого фундаментального процесса. Подробный анализ этих механизмов позволяет более четко выявить, что же в них является сходным, а что кардинально различным и тем самым „ сфокусировать внимание исследователей на еще совсем неисследованных проблемах. В данной работе обсуждаются новые примеры, которые показывают очевидное сходство во взаимодействии мРНК с рибосомами бактерий и эукариот (млекопитающих).

Нами было показано, что 'внутренняя инициация трансляции для IRES-элемента РНК вируса черемуховой тли (Rhopalosiphum Padi Virus, RhPV) возможна не только в бесклеточных системах, но и in vivo - следовательно, все данные, полученные ранее in vitro методом реконструкции инициаторных комплексов-из-очищенных компонентов (совместно с И.М. Терениным) отражают "упрощенный" способ инициации трансляции, который напоминает инициацию трансляции у прокариот.

Основное внимание в диссертационной работе было уделено изучению механизмов инициации трансляции для безлидерных мРНК. Ранее в нашей лаборатории Балакиным и др. было показано, что безлидерная С1 мРНК фага X способна связываться с недиссоциированной прокариотической 70S рибосомой и инициаторной тРНКи в отстутствие факторов инициации трансляции. В ходе данной работы было показано, что модельная безлидерная С1 мРНК способна связываться с 80S рибосомой и инициировать белковый синтез в отсутствие факторов инициации трансляции. Установлено, что связывание безлидерной мРНК с 80S

1 рибосомой обусловливается расположением инициаторного триплета на самом 5' конце мРНК и одноцепочечной конформацией мРНК непосредственно за инициаторным триплетом.

Чтобы приблизиться к пониманию механизма инициации трансляции безлидерных мРНК, который может реализовываться in vivo в условиях конкуренции с обычными мРНК и в присутствии полного набора факторов инициации, нами был проанализирован возможный "стандартный" механизм инициации через образование 48S предынициаторного комплекса. Методом реконструкции 48S комплексов из очищенных компонентов было показано, что фактор инициации elFl оказывает дестабилизирующее влияние на 48S комплекс, собранный на безлидерной мРНК, что делает инициацию трансляции для безлидерной мРНК через связывание с 80S рибосомой предпочтительной.

Исследованные нами трансляционные свойства безлидерной мРНК в бесклеточной системе (JIPK) представили весомые аргументы в пользу того, что в системе, в которой присутствуют все компоненты трансляционного аппарата, для безлидерной мРНК реализуется (хотя бы частично) фактор -независимый механизм инициации через связывание с 80S рибосомой.

Таким образом, в данной работе был предложен новый механизм инициации трансляции у эукариот и показана практически полная аналогия в связывании безлидерной мРНК с 70S и 80S рибосомами.

Обзор литературы

Структура и функции рибосомы, ключевого компонента биосинтеза белка, с самого своего открытия привлекают внимание ученых. Действительно, сложно представить себе более фундаментальный процес в клетке, чем синтез полипептида, и более сложно устроенный рибонуклеопротеиновый комплекс, отвечающий вместе со вспомогательными белковыми факторами за выбор инициаторного триплета на мРНК, удлинение полипептидной цепи и ее высвобождение после окончания цикла трансляции.

В конце 1990 годов в изучении структуры рибосомы был сделан настоящий прорыв - увенчались успехом попытки закристаллизовать рибосомную субчастицу из термофильной бактерии Thermus thermophilics и получить ее рентген с атомным разрешением. В дальнейшем были решены структуры не только отдельных прокариотических рибосомных субъединиц и целой 70S рибосомы, но и комплексов рибосомы с мРНК, тРНК и белковыми факторами. В отличие от прокариотической, эукариотическую рибосому до сих пор закристаллизовать не удалось, однако в последнее время был достигнут значительный прогресс благодаря развитию метода криоэлектронной микроскопии. Сравнивая имеющуюся атомную структуру прокариотической рибосомы со структурой эукариотической рибосомы, полученную методом криоэлектронной микроскопии, можно с достаточно большой степенью надежности выдвигать предположения о структуре последней.

Все известные рибосомы выполняют очень похожие функции, и весьма схожи структурно. Однако механизмы трансляции в прокариотах и эукариотах отличаются довольно сильно, особенно на самом сложном этапе, который обычно и определяет уровень экспрессии данной мРНК -стадии инициации. Цель данного обзора литературы - попытка разобрать сходства (ввиду схожести функций) и различия (которые неизбежны из-за принципиально разных механизмов трансляции и биогенеза рибосом) прокариотической и эукариотической рибосомы. Для большей полноты такого сравнения необходим хотя бы краткий обзор основных трех стадий биосинтеза белка - инициации, элонгации и терминации.

Выводы

1. Показано, что безлидерная С1 мРНК способна связываться с 80S рибосомой в отстутствие факторов инициации трансляции.

2. Установлено, что тройной комплекс, образуемый 80S рибосомой, С1 мРНК и Мет-тРНКиМет, способен участвовать в элонгации трансляции, т.е. является полностью функциональным инициаторным комплексом.

3. Показано, что С1 мРНК способна образовывать 48S инициаторный комплекс с 40S рибосомой и факторами инициации трансляции, однако выход такого комплекса сравним с выходом тройного 80S инициаторного комплекса. Кроме того, показано, что фактор инициации elFl способен частично дестабилизировать инициаторный комплекс только в том случае, если в системе присутствует фактор инициации eIF4F, а также то, что eIF4F оказывает стимулирующий эффект на образование 48S инициаторного комплекса в присутствии АТР и активного фактора инициации eIF4A.

4. Изучены трансляционные свойства С1 мРНК в бесклеточной системе трансляции ЛРК - показано, что трансляция С1 мРНК не ингибируется при повышении ее концентрации, что трансляция С1 мРНК не зависит от присутствия кеп-структуры на 5'-конце, и что С1 мРНК способна эффективно транслироваться при инактивации eIF2.

5. Показано, что расположение инициаторного триплета на самом 5' конце мРНК и одноцепочечная конформация области непосредственно после инициаторного триплета являются определяющими факторами, позволяющими мРНК непосредственно связываться с 80S рибосомой в отсутствии факторов инициации трансляции.

6. Доказана способность к трансляции неспецифического IRES-элемента вируса насекомых RhPV in vivo и в усовершенствованной бесклеточной системе. Полученные данные подтверждают сделанный ранее вывод о кеп-независимом способе инициации трансляции на РНК RhPV, сходным с прокариотическим механизмом инициации трансляции.
