1.1. Актуальность темы

Репродуктивный и респираторный синдром свиней (РРСС) - контагиозная вирусная болезнь, характеризующаяся абортами в конце срока супоросно-сти, рождением мертвых и/или слабых поросят, погибающих в первые 2-3 недели жизни, и поражением органов дыхания у поросят в постотъёмный период (23, 45, 200). Вирус чаще циркулирует в изолированных фермах с полным циклом и промышленных свинокомплексах, наносит значительный экономический ущерб из-за репродуктивных потерь и низкой сохранности новорожденных и поросят на доращивании (15).

В настоящее время, на основании имеющихся сведений о вирусе и патогенезе болезни рутинные диагностические исследования базируются на использовании серологических методов для обнаружения специфических антител и обратно-транскриптазной полимеразной цепной реакции (ОТ-ПЦР) для обнаружения генома (200). Так, при типичных случаях (т.е. наличии эпизоотологи-ческих данных, клинических признаков) в хозяйстве с полным циклом при отсутствии вакцинации, первичного обнаружения генома вируса РРСС и специфических антител, особенно у поросят группы доращивания, достаточно для подтверждения диагноза, а обследуемая ферма (хозяйство) считается неблагополучной по РРСС (36, 61). Однако, результаты исследований, выполняемых на основе использования коммерческих диагностических наборов «РРСС-Серотест» производства НПО НАРВАК, широко используемого в РФ, «HerdChek PRRS 2XR» фирмы IDEXX (США) и др., не позволяют идентифицировать группы недавно зараженных животных в неблагополучной ферме, потому что реакции предназначены для обнаружения только специфических антител к вирусу РРСС класса IgG. Недостаточная информативность существующих серологических методов не позволяет использовать их для совершенствования стратегии и тактики борьбы с болезнью. Учитывая энзоотичность РРСС в свиноводческих хозяйствах, особенно с полным циклом, перспективным направлением для совершенствования мер борьбы с болезнью является разработка серологического метода для выявления животных на ранних стадиях заражения, являющихся источником вируса, а именно, обнаружение специфических вирусу РРСС ранних антител класса IgM. Несмотря на широкое использование методов обнаружения ранних антител в медицинской вирусологии, сведения о диагностическом значении специфических антител класса IgM в ветеринарии, в том числе и при РРСС, ограничены лишь несколькими работами (117, 166).

До последнего времени считалось, что в странах Европы (за исключением Дании) циркулируют изоляты вируса РРСС европейского генотипа, а в Северной Америке и странах Азии - американского. По данным литературы, в свиноводческих хозяйствах РФ циркулируют изоляты, относящиеся только к европейскому генотипу вируса РРСС (16). Однако, с учетом значительных закупок племенных свиней из стран Европы и Северной Америки, а также применения в течение нескольких лет живой вакцины из штамма «БД» американского варианта (181), ситуация может измениться, и поэтому с целью контроля заноса вируса РРСС американского варианта актуальным является серологический мониторинг свиней как неблагополучных, так и благополучных хозяйств на присутствие антител к американскому варианту вируса.

Таким образом, несмотря на существенные достижения в изучении вируса и вызываемой им болезни, применение существующих средств специфической профилактики и мер борьбы не всегда дает успех, так как эффективные вакцины отсутствуют, а длительная персистенция, различные формы проявления болезни и циркуляция генетически различных европейского и американского вариантов вируса РРСС приводят к необходимости повысить эффективность серологических исследований и улучшить схему лабораторных диагностических исследований.

6. выводы

1. Для выделения полевых изолятов вируса РРСС, циркулирующих на территории РФ, необходимо использовать первичную культуру АМС после предварительной проверки чувствительности помётов свиноматок к репродукции референс-вируса; для приготовления тест-препаратов в РНИФ - вирус европейского («Lelystad») и американского («БД») вариантов, адаптированный к культуре клеток MARC-145, при множественности заражения 0,1-1,0 ТЦД 50/клетка;

2. Усовершенствованные серологические методы (РНИФ, ГХ-ИФА) с использованием коньюгатов анти-IgM свиньи позволяют обнаруживать специфические антитела (иммуноглобулины) класса М к вирусу РРСС с 4-х по 28 сутки после заражения;

3. На основании установленной достоверной средней корреляционной связи между виремией и обнаружением специфических IgM в сыворотках крови свиней, зараженных вирусом РРСС, РНИФ и ГХ-ИФА могут быть использованы для обнаружения вирусоносителей;

4. Разработанная модификация РНИФ позволяет дифференцировать специфические антитела к европейскому и американскому вариантам в сыворотках крови инфицированных и переболевших свиней, а также, используя гомологичные или гетерогенные специфические сыворотки, определять принадлежность вируса к европейскому или американскому варианту;

5. Диагностические характеристики лабораторных методов диагностики РРСС зависят от времени отбора проб после заражения: так, метод изоляции вируса и ОТ-ПЦР имеют 75-100% чувствительность с 7 по 28 сутки после заражения и в этот же период РНИФ в 83,3-100% проб обнаруживаются специфические антитела класса IgM. Серологические методы определения специфических РРСС IgG имеют наибольшую чувствительность, начиная с 14-21 (42-100%) до 45 суток (100%>) (срок наблюдения);

6. Наиболее результативными лабораторными методами для обнаружения и скрининга инфицированных вирусом РРСС животных по диагностической

Ill чувствительности и специфичности являются РНИФ и ГХ-ИФА, обеспечивающие как обнаружение специфических антител класса IgM, IgG, так и дифференциацию европейских и американских изолятов вируса;

7. Рекомендованная схема лабораторной диагностики РРСС, включающая методы обнаружения ранних специфических IgM и IgG к вирусу РРСС, серологический метод дифференциации европейского и американского вариантов вируса, позволяет идентифицировать как стадию инфекционного процесса (острую или реконвалесценцию) у отдельных животных, так и определять эпизо-отологический статус обследуемой популяции/фермы.

7. ПРАКТИЧЕСКИЕ ПРЕДЛОЖЕНИЯ

1. Данные, полученные в результате проведения экспериментальных исследований, позволяют рекомендовать для применения в лабораторной диагностике РРСС метод выявления специфических антител IgM и серологический метод дифференциации европейского и американского вариантов вируса РРСС как чувствительные, специфичные и высоко воспроизводимые методы.

2. Подготовлена и утверждена директором ГНУ ВНИИВВиМ нормативная документация: «Временные методические указания по идентификации антител к европейскому и американскому вариантам вируса репродуктивного и респираторного синдрома свиней».

ЗАКЛЮЧЕНИЕ

Таким образом, в результате проведенных исследований раскрыты некоторые особенности клинических проявлений РРСС, проведена сравнительная оценка лабораторных методов диагностики РРСС, разработан гистохимический метод определения специфических вирусу антител IgM, позволяющий выявлять животных на ранних стадиях заражения вирусом РРСС, и серологический метод для дифференциации вариантов вируса и вирусспецифических антител. Разработанные методы позволяют полно характеризовать эпизоотологическую ситуацию по РРСС в свинокомплексах, определять группы животных, являющиеся резервуаром возбудителя болезни. Проведена сравнительная оценка характеристик основных методов, используемых для лабораторной диагностики РРСС: ОТ-ПЦР, изоляции вируса, определения специфических антител IgG с помощью РНИФ, ГХ-ИФА и ТФ-ИФА, а также разработанных методов определения специфических антител IgM и серологической дифференциации вариантов вируса и антител.

Диагностическая оценка характеристик методов при экспериментальном заражении животных и при исследовании полевого материала позволяет рекомендовать метод определения специфических вирусу антител IgM и метод серологической дифференциации европейского и американского вариантов вируса для использования при лабораторной диагностике РРСС.
