Актуальность проблемы. Рибосома является клеточной органеллой ответственной за синтез белка во всех клетках. Анализ структуры рибосомы выполненный с помощью рентгеноструктурного кристаллографии в последние годы значительно улучшил понимание структурных основ функционирования рибосомы [1]. Однако, сравнительно мало известно о биогенезе рибосом, особенно, у эукариот. В клетках млекопитающих синтез цитоплазматической рибосомы требует сборки 4 молекул РНК и 79 различных белков [7, 8]. В среднем клетка содержит примерно 4 миллиона цитоплазматических рибосом, для которых необходимо порядка 80% всей клеточной РНК и 5-10% белков. Исследование механизмов, которые контролируют экспрессию компонентов рибосомы, является важной задачей современной науки. Три различных РНК-полимеразы вовлечены в продукцию РНК и белков для рибосомы: РНК-полимераза I транскрибирует гены, кодирующие 28S, 18S и 5,8S рРНК, РНК-полимераза II - гены РБ и РНК-полимераза III -гены 5S рРНК. Нуклеотидные последовательности большинства генов РБ и рРНК уже определены, однако лишь для небольшой части генов РБ млекопитающих исследована регуляция их экспрессии. В отличие от рРНК, которые кодируются несколькими сотнями генов и кластеризуются в несколько локусов, каждый эукариотический РБ кодируется только одним геном (кроме RPS4, различные гены которого находятся на X и Y хромосомах) и эти гены распределены по всему геному [9]. Единственная функциональная копия гена генерирует большое количество процессированных псевдогенов [10], что осложняет клонирование и функциональный анализ генов РБ. Для достижения эквимолярного количества РБ в большом разнообразии условий клеточного роста используется координация различных уровней экспрессии генов. Так регуляция на транскрипционном и пост-транскрипционном уровнях экспрессии генов РБ необходима для достижения примерно равного количества всех рибосомных белков. Это демонстрируют эксперименты, показывающие равное привлечение РНК-полимеразы промоторами различных генов РБ, равное количество мРНК, сравнимую степень экспрессии репортерных генов [3, 11]. Эти данные предполагают, что гены РБ используют промоторы примерно равной силы. Исследования регуляторных элементов промоторов генов РБ выявили сайты связывания ТФ как в выше, так и ниже точки старта транскрипции [12]. Наиболее распространенными из используемых генами РБ являются ТФ GABP, Spl, YY1 [13]. Если связывание любого индивидуального ТФ с промотором гена нарушалось (сайт-специфические мутации), транскрипционная активность гена уменьшалась, но не прекращалась. Это указывает на сложную комбинированную регуляцию транскрипции многими ТФ. Однако, регуляторных факторов характерных только для генов РБ не выявлено.

Особенностью генов РБ высших эукариот является наличие ОПТ в районе старта транскрипции. ОПТ играет важную роль в определении точки старта транскрипции, а также является цис-элементом^трансляционного контроля экспрессии генов РБ [14, 15]. Трансляционный контроль ТОП мРНК (мРНК с терминальным ОПТ) выражается в селективном выходе данных мРНК из полисом (репрессия трансляции) при блокировании прохождения клетки через фазу G1 (недостаток питательных веществ, контактное ингибирование или конечная дифференцировка), G1/S (ингибирование синтеза ДНК) и G2/M (ингибирование сборки веретена деления), а также при недостатке аминокислот. Когда же покоящаяся клетка индуцируется на клеточное деление или рост или происходит восстановление запаса аминокислот, ТОП мРНК рекрутируются на полисомы (активация трансляции). Точных механизмов регуляции трансляции ТОП мРНК пока не выявлено. Интерес к биосинтезу рибосомы также стимулируется находками, что мутации в генах, кодирующих компоненты рибосомы или белки, вовлеченные в созревание, сборку или экспорт рибосомы из ядра, могут приводить к развитию некоторых наследственных заболеваний у человека. Так, например, анемия Diamond-Blackfan (DBA) - врожденная (наследственная) форма парциальной красноклеточной аплазии, причиной которой являются мутации в одном или нескольких генах РБ. Мутации, приводящие к развитию DBA, были найдены в генах RPS19 (около 25% случаев) [16], RPS24 (2%)[17], RPL11 и RPL5 (по 10%) [18] и др. Во всех случаях наследование является аутосомно-доминантным. Считается, что в основе заболевания лежит гаплонедостаточность РБ, которая приводит к недостаточному созреванию рибосом[19], что в свою очередь вызывает синтез малого количества гемоглобина и гибели предшественников эритроцитов.

Особый интерес представляют результаты, указывающие на участие многих рибосомных белков в регуляции клеточного цикла. Важность рибосом в регуляции роста и гибели клеток неудивительна, но рибосомные белки вовлекаются в эти процессы не только как структурные элементы рибосомы, но и как эффекторы или модуляторы изменений клеточного цикла. Так, например, инактивация гена рибосомного белка S6 Drosophila melanogaster приводит к образованию опухолей в некоторых тканях, что указывает на RPS6 как на ген-супрессор опухолевого роста [20]. С другой стороны, в опухолевых клетках повышен уровень некоторых рибосомных белков, например, S15 крысы [21], L18 и L37 человека [22], а увеличение экспрессии генов RPL7 [23] и RPS3a [24] человека приводит к угнетению роста клеток и вступлению их в апоптоз. Аминокислотные последовательности многих рибосомных белков сходны с функциональными мотивами

ТФ, например, "лещиновая застежка" у RPL7 человека [25] и RPL13a крысы [26], "цинковые пальцы" у RPL37 человека [27] и RPL37a D.melanogaster [28], что указывает на возможное участие этих белков в регуляции транскрипции. Поэтому интерес к регуляции экспрессии генов рибосомных белков обусловлен и наличием у некоторых из них внерибосомных функций.

Рибосомный белок L11 является компонентом 60S субъединицы рибосомы [29]. Данный белок относится к L5p семейству рибосомных белков и взаимодействует с 5S рРНК. Рибосомные белоки LI 1 и L5 необходимы для присоединения 5S рРНК к 90S прерибосомальному комплексу, играя важную роль в созревании рибосомы[30]. Кроме выполнения структурной функции в составе рибосомы, L11 играет важную роль в индукции р53-зависимого ареста клеточного цикла, обеспечивая, таким образом, связь между уменьшением сборки рибосом в ответ на какие-либо изменения условий жизни клетки и ингибированием роста. Данный эффект достигается связыванием избытка LI 1 с HDM2, что предотвращает ее взаимодействие с р53, таким образом активируя р53-зависимый арест клеточного цикла [31, 32]. Таким образом, интерес к строению и функционированию гена рибосомного белка L11 связан как с вхождением его в состав рибосомы, так и с его регуляторной ролью в клеточном цикле.

Цели и задачи исследования. Целью настоящей работы является исследование структурной организации гена рибосомного белка LI 1 человека, а также идентификация ТФ, участвующих в регуляции его экспрессии.

Для достижения поставленной цели следовало решить следующие задачи:

1. Клонирование и секвенирование гена HRPLJJ и его промоторной области.

2. Определение структурных особенностей гена HRPLJJ (точки старта транскрипции, наличие мяоРНК кодируемых интронами гена) и сравнение их со структурными особенностями генов других РБ.

3. Выявление ТФ, взаимодействующих с промоторными элементами гена HRPLJJ.

4. Сравнение ТФ, взаимодействующих с промоторными районами генов рибосомных белков RPLJ J, RPL32 и RPS26 человека

5. Выявление функционального значения ТФ, взаимодействующих с промоторными элементами гена HRPLJJ.

Основные положения, выносимые на защиту.

1. Структура гена HRPLJJ соответствует основным характеристикам генов РБ млекопитающих.

2. Промотор гена HRPLll содержит несколько участков связывания ТФ, таких как YY1 (Yin Yang 1), GABP (GA-связывающий белок) и АР2.

3. Существуют общие белковые факторы, характерные для промоторных районов генов HRPLll, HRPL32 и HRPS26, а также существуют ТФ общие для HRPLll и HRPL32 и не встречающиеся у HRPS26.

4. ТФ YY1 входит в сложный ДНК-белковый комплекс, формирующийся в районе +6.+26 гена HRPLll и является белком-активатором транскрипции гена HRPLll.

Объем и структура диссертации. Диссертация состоит из введения, обзора литературы, материалов и методов исследования, результатов исследования, обсуждения результатов, выводов и списка цитируемой литературы. Работа изложена на 100 стр. машинописного текста, иллюстрирована 32 рисунком и 5 таблицами. Список литературы содержит 175 библиографических источников.

Выводы

1. Определена нуклеотидная последовательность гена рибосомного белка L11 человека {HRPLll) и его промоторной области, проведен анализ структурных особенностей гена HRPLll. Ген HRPLll соответствует основным характеристикам генов РБ млекопитающих:

• имеет небольшой размер - 4583 п.н., короткий первый экзон (32 п.н.), короткие 5'- и З'-нетранслируемые области (26 и 52 п.н., соответственно);

• граница первого интрона лежит в непосредственной близости к AUG-кодону, стоп-кодон располагается в последнем 6-м экзоне, на расстоянии 25 п.н. от него находится участок полиаденилирования, ААТААА;

• на расстоянии 3 п.н. от AUG-кодона находится характерный для генов РБ олигопиримидиновый тракт, состоящий из 24 п.н., в котором находится точка инициации транскрипции;

• В промоторной области гена HRPLll отсутствуют TATA- и СААТ-боксы, но в районе -24.-27 находится ТАТА-подобный элемент (ATА А) в окружении GC-богатых районов.

2. Определены функциональные сайты связывания транскрипционных факторов (ТФ), расположенные в промоторном районе гена HRPLll (-59. .+170).

• Методом "задержки в геле" с использованием олигонуклеотидных копий участков связывания ТФ показано, что ген HRPLll содержит участки связывания факторов транскрипции YY1 (Yin Yang 1), GABP (GA-связывающий белок) и АР2.

• С помощью поликлональных антител к данным ТФ показано, что транскрипционный фактор YY1 входит в сложный ДНК-белковый комплекс, формирующийся в районе +6.+26 гена HRPLll.

• При сравнении набора ядерных белков, взаимодействующих с промоторным районом гена HRPLll, полученных из быстропролиферирующих и нормальных клеточных линий, а также клеток, находящихся в репрессированном состоянии, получено свидетельство того, что транскрипционный фактор YY1 является белком-активатором транскрипции гена HRPLll.

3. Проведено сравнение ТФ, взаимодействующих с промоторными районами генов РБ RPL11 (-59.+170), RPL32 (-335.+79) и RPS26 (-217.+113) человека. С помощью метода «задержки в геле» с испольхованием в качестве конкурентов фрагментов промоторных районов генов HRPL11, HRPS26 и HRPL32, а также олигонуклеотидных копий участков связывания ТФ, показано, что существуют общие ТФ, взаимодействующие с промоторными районами генов HRPL11, HRPS26 и HRPL32, а также ТФ, взаимодействующие с промоторными районами генов HRPL11 и HRPL32 и не взаимодействующие с промоторным районом гена HRPS26.
