Последние успехи в области генетического определения пола насекомых (Zhu et al., 2002; Horn, Wimmer, 2003) и интенсивная работа над созданием трансгенных комаров, не способных переносить малярию или Денге (Ito et al., 2002; Lara Capurro de et al., 2000), говорят о том, что внедрение и широкое использование эффективных программ контроля численности вредителей и контроля над трансмиссивными заболеваниями с помощью трансгенных насекомых дело ближайшего будущего. При внедрении в природную популяцию вредителей большого количества трансгенных насекомых основной задачей, стоящей перед специалистами, является соблюдение высокого уровня безопасности технологии для человека и окружающей среды. Существующая сейчас технология получения трансгенных насекомых не соответствует этому требованию.

Основным методом получения трансгенных насекомых является интеграция в геном реципиента генетического материала с помощью неавтономных транспозонов, которые представляют собой векторную молекулу, содержащую терминальные инвертированные повторы (TIR от англ. - terminal inverted repeats) транспозона, между которыми заключена последовательность трансгена (Finnegan,1989; Handler et al., 2004). При таком способе интеграции трансген оказывается окруженным TIR мобильного элемента. Оба повтора необходимы для перемещения транспозона и узнаются транспозазой, которая вырезает фланкированную ими последовательность из одного места в геноме и вставляет ее в другое ("cut and paste" механизм).

При использовании трансгенных насекомых в программах контроля численности насекомых-вредителей или при замещении дикой популяции переносчиков малярии или лихорадки Денге резистентными трансгенными насекомыми одним из главных рисков является возможность ремобилизации трансгена (Handler et al., 2004). При внешнем источнике транспозазы трансген ремобилизуется и начнет перемещаться по геному. Как поведет себя тот или иной трансген, предсказать невозможно (Fraser et al., 2000; Perera et al., 2002; Sethuraman et al., 2007). Риск повышает и то, что горизонтальный перенос транспозона и последующее его распространение в популяции могут происходить с большой скоростью (Engels et al., 1992; Kidwell et al., 1992). Кроме того, многие родственные семейства транспозонов способны к перекрестной мобилизации, т. е. транспозаза одного типа мобильных элементов может вызывать транспозиции другого транспозона (Robertson, Lampe, 1995; Handler, McCombs, 2000).

Актуальной задачей становится разработка метода стабилизации трансгенов после их интеграции в геном. В связи с этим, целью настоящей работы было разработать метод стабилизации трансгенов после их интеграции в геном на базе репарации индуцированных двуцепочечных разрывов ДНК.

Для достижения указанной цели были поставлены следующие экспериментальные задачи:

1. Разработать новый метод стабилизации трансгенов после их сайт-неспецифической интеграции в геном на основе репарации индуцированных двуцепочечных разрывов ДНК в модельной системе Dro-sophila melanogaster

2. Выявить эффективный механизм репарации двуцепочечных разрывов ДНК для реализации разработанного метода стабилизации трансгенов

3. Проанализировать характер делеций генетического материала, возникающих в результате репарации индуцированных двуцепочечных разрывов

4. Разработать новый метод стабилизации трансгенов после их сайт-специфической интеграции в геном за счет SSA репарации индуцированных двуцепочечных разрывов ДНК в модельной системе D. melanogaster

5. Показать возможность эффективного одновременного удаления всех плечей неавтономного транспозона и селективных маркеров В настоящей работе предложен принципиально новый эффективный метод стабилизации трансгена после его интеграции в геном реципиента, основанный на сайт-направленной делеции последовательности плечей транспозона с помощью репарации индуцированных двуцепочечных разрывов ДНК.

Предложенные схемы строения векторных молекул могут быть использованы при создании удобных векторов для трансгенеза насекомых в интегрированных программах контроля численности вредителей (IPM program — от англ. Integrated pest management program). Кроме того, универсальность используемых принципов стабилизации трансгенов, позволит создать серию универсальных векторов для трансгенеза животных, растений и клеточных линий со стабилизированными трансгенами.

7. Выводы

1. Создан новый метод стабилизации трансгенов после их сайт-неспецифической интеграции в геном, основанный на удалении функциональной части плеч транспозона за счет 1МНЕ1 репарации индуцированных двуцепочечных разрывов ДНК

2. Впервые показано, что размер делеций, возникающих в результате репарации двуцепочечных разрывов ДНК по ЫНЕ1 механизму, составляет в среднем 1000-2000 п.н. Выявлено, что делеции могут носить двунаправленный характер от точки двуцепочечного разрыва ДНК

3. Создан новый метод стабилизации трансгенов после их сайт-специфической интеграции в геном, основанный на удалении всех последовательностей транспозона за счет ЗБА репарации индуцированных двуцепочечных разрывов ДНК. Метод позволяет эффективно одновременно удалять все плечи неавтономного транспозона и маркеры
