В настоящее время в молекулярной биологии быстрыми темпами идет накопление экспериментального материала по первичной структуре геномной и митохондриальной ДНК различных организмов. Получаемая информация позволяет переводить на молекулярный уровень такие дисциплины как этногеномика, палеогеномика, медицинская геномика, систематика и т.д. Одним из важнейших современных направлений в молекулярной биологии и генетике является изучение изменчивости генетического материала в процессе эволюции.

Первые позвоночные на Земле появились в результате эволюции хордовых, для которых были характерны частые геномные дупликации [McLysagt et al., 2004]. Избыточные гены, образовавшиеся вследствие полиплоидизации, со временем изменялись, приобретая иногда новые функции, подвергались элиминации, гомогенизации нуклеотидных последовательностей, внедрению повторов и эпигенетическим изменениям [Blanc and Wolfe, 2004]. В течение долгого времени считалось, что амфибии произошли от рыб, рептилии - от амфибий, а млекопитающие -от рептилий. Однако за последние полвека накопилось много данных, полученных при изучении останков ископаемых организмов, и эти данные поставили под сомнение традиционную схему.

Одной из основных не решенных проблем является загадка возникновения и эволюции тетрапод. Под тетраподами имеются в виду древние (стволовые) организмы с четырьмя конечностями, не обладающие характерными чертами своих потомков, современных амфибий, рептилий и млекопитающих. Сегодня древних тетрапод представляют как рыбообразных животных с конечностями, снабженными пальцами. Ближайшими родственниками тетрапод из ныне живущих организмов являются двоякодышащие рыбы, например, Tetraodon nigriviridis — пресноводная рыба с самым маленьким из всех известных геномов позвоночных. Сравнительный анализ геномов

Tetraodon и человека привел к идентификации ключевых генов, которые ранее считались отсутствующими у рыб. Эти и другие результаты позволяют предположить, что уже в глубокой древности образовались две основные ветви позвоночных, эволюционное развитие одной из которых привело к формированию класса рыб, а другой - к формированию классов амфибий, рептилий и млекопитающих [Smith and Vos, 2007]. Мало изученным оставался в течение долгого времени процесс формирования ветви рептилий на эволюционном древе. Последние исследования позволяют думать, что класс рептилий занимает заметно обособленную позицию и по многим важным признакам отличается от других таксонов позвоночных. Наиболее известное эволюционное древо рептилий, построенное на основании морфологических признаков, требует пересмотра в свете новых данных, полученных на молекулярном уровне [Hedges and Poling, 1999]. Анализ, проведенный на основании нуклеотидных последовательностей ДНК девяти кодирующих генов, C-mos, RAG1, RAG2, R3S, НОХА13, Jun, альфа-энолазы, амелогенина и MAFB [Vidal and Hedges, 2005], для альфа-, и бета- глобиновых последовательностей [Gorr et al., 1998], и т.д. для таксонов, представляющих главные ветви рептилий, дал результаты, сильно отличающиеся от ранее существующей морфологической классификации.

Гены, кодирующие рибосомные РНК (рРНК), являются необходимым компонентом эукариотической клетки. Информация по размеру повторяющихся единиц рДНК и их представленности в геноме была в основном получена в 70-е - 80-е годы XX века [Dawid, 1980], и с тех пор редко пополнялась. Информация данного типа для геномов рептилий до сегодняшнего дня отсутствует. Нуклеотидные последовательности рДНК являются одним из перспективных объектов для изучения путей эволюции генома. Для рДНК характерно чередование консервативных и вариабельных участков, которые удобно использовать для эволюционных исследований на различных таксономических уровнях.

Для изучения эволюционных связей часто используется участок рибосомного оперона, включающий внутренние транскрибируемые спейсеры предшественников рибосомной РНК, ВТС1 и ВТС2, в особенности ВТС2, содержащий много сайтов процессинга, важных для точного формирования зрелых 18S, 5.8S и 28S рРНК. Более, чем 100 ООО последовательностей ВТС2 депонировано в настоящее время в GenBank. Подавляющее большинство этих видоспецифических последовательностей принадлежит беспозвоночным. Их интенсивно и успешно использовали для филогении и эволюционных сравнений растений, протистов и некоторых беспозвоночных. Не смотря на первое впечатление, что ВТС2 слишком вариабелен, с его помощью можно получать ценную информацию и для таксонов более высокого уровня, благодаря наличию высоко консервативных (консенсусных) последовательностей, обеспечивающих консервативность вторичной структуры соответствующего участка пре-рРНК [Joseph, 1999; Coleman, 2007]. Использование ВТС2 для изучения эволюционных связей позвоночных является сложной задачей в силу их значительной дивергенции по нуклеотидной последовательности и по длине. Однако существование консенсусных сегментов и усовершенствование программ получения потенциальных вторичных структур для протяженных нуклеотидных последовательностей делает решение этой задачи возможным. Для получения более надежных и обоснованных результатов необходимо наращивание информационной базы. На основании вышеизложенного, представляется весьма актуальным получение новых данных по молекулярной организации рДНК рептилий и эволюционной изменчивости ее различных компонентов.

1. Впервые определены размеры мономеров, образующих кластеры рибосомной ДНК в геномах 10 видов чешуйчатых рептилий, которые варьировали от 10 до 15 т.п.н.

2. Установлена степень повторности мономеров рДНК у ряда представителей рептилий, показано что у ряда ящериц она значительно меньше, чем у всех ранее исследованных позвоночных.

3. Показано, что у Uta stansburiana рДНК (Iguania) локализована на одной хромосоме.

4. Сравнение нуклеотидных последовательностей ВТС2 15 представителей отряда Squamata между собой выявило различия в размерах и характерные особенности организации этого элемента рДНК у представителей семейства Iguania и Lacertidae

5. Показано, что увеличение размеров ВТС2 у исследованных рептилий является следствием коротких инсерций и локальных дупликаций.
