Актуальность темы. Микроорганизмы (грибы, водоросли, простейшие, бактерии, актиномицеты) являются важнейшим компонентом почвы и вносят значительный вклад в формирование структуры наземных экосистем (Звягинцев, 1987; Бахнов, 2002; Звягинцев, 2005). Масса почвенной микро-биоты составляет большую часть общей массы всех микроорганизмов нашей планеты. Однако, сведения о биологии почвенных микроорганизмов, о видовом разнообразии, об их относительном обилии в различных типах почв, весьма ограничены (Алехина и др., 2002; Полянский и др., 2002; Добровольский, Никитин, 2006). Получение новых экспериментальных данных ограничивается тем, что до недавнего времени отсутствовали адекватные высокоинформативные методы исследования микроорганизмов почвы в естественной среде их обитания.

Как известно, большинство почвенных микроорганизмов не способны к культивированию в искусственных условиях вне их естественного места обитания (Muyzer et al., 1993). Соответственно, истинное количество видов микроорганизмов и их относительное изобилие в почве до сих пор не известны. Предполагается, что в почве могут обитать десятки тысяч видов бактерий, но лишь небольшая доля этих бактерий выделена и охарактеризована (Muyzer, Smalla, 1998; Whitman, Coleman, 1998; Ward, 2006). По оценкам некоторых авторов, разнообразие почвенных грибов может доходить до 1,5 млн. видов (Hawksworth, Rossman, 1997; Mueller, Schmit, 2007). Мало изученными остаются и вопросы, касающиеся количественного и качественного состава, структуры различных микробных сообществ в зависимости от физических и химических свойств почвы, при действии различных экологических факторов, таких как влажность, рельеф и т.д. Следует отметить, что в последние годы были разработаны молекулярно-генетические методы, позволяющие идентифицировать и определить разнообразие микроорганизмов в различных средах, включая и организмы, которые не поддаются изучению при помощи традиционных микробиологических методов (Dunbar et al., 1999; Roesch et al., 2007). К таким методам относится технология параллельного высокопроизводительного пиросеквенирования, которая позволяет анализировать последовательности ДНК генов 18S и 16S рРНК (Rothberg, Leamon, 2008). Другой молекулярный метод денатурирующего градиентного гель -электрофореза продуктов ПЦР позволяет сравнивать состав различных микробных сообществ в целом, без таксономической идентификации (Muyzer, 1999).

В нашей работе были разработаны методические подходы, позволяющие использовать эти технологии для анализа генетического разнообразия сообществ почвенных микроорганизмов и оценки влияния факторов окружающей среды на состав и структуру этих сообществ.

Цели и задачи работы. Целью настоящей работы является определение генетического разнообразия и изобилия бактерий, грибов в почвах различных типов ландшафта и оценка степени влияния экологических факторов на качественный и количественный состав почвенной микробиоты.

Были поставлены следующие задачи:

• Разработка методических подходов применения денатурирующего градиентного гель - электрофореза и пиросеквенирования для измерения генетического разнообразия, идентификации (классификации) эукариотиче-ских и прокариотических организмов в почвенных образцах.

• Идентификация и таксономическая классификация почвенных микроорганизмов, в т.ч. и из ризосферы растений, по первичной последовательности ДНК (генов 18S и 16S рРНК).

• Определение относительного количества таксонов прокариот и выявление доминирующих групп организмов в бактериальных сообществах почв различных типов ландшафта в зависимости от экологических факторов.

• Определение относительного количества таксономических групп почвенных грибов и выявление доминирующих таксонов в ризосфере в зависимости от вида растений и условий их произрастания. Оценка степени влияния экологических факторов на уровень генетического многообразия и изобилия различных таксонов почвенных микроорганизмов.

• Измерение и анализ параметров качественного и количественного состава арбускулярно - микоризных грибов в зависимости от вида растения - хозяина и экологических факторов.

Научная новизна. Впервые, с использованием метода пиросеквениро-вания, проведен анализ генетического разнообразия и изобилия микробиоты в ризосфере растений естественных сообществ и определена степень зависимости этих параметров от экологических факторов. Впервые выявлены доминирующие таксоны микроорганизмов в составе естественных микробоце-нозов почвы различных типов ландшафтов.

Практическая значимость. Разработаны методические подходы позволяющие применять технологии пиросеквенирования и денатурирующего градиентного гель - электрофореза ПЦР - продуктов для анализа генетического разнообразия сообществ и идентификации почвенных микроорганизмов. Предложенные методики можно использовать для мониторинга состояния естественных экосистем, агроценозов, урбанизированных территорий. Результаты исследований могут быть использованы в качестве учебного материала для освоения теоретических курсов и проведения лабораторных занятий на биологических, экологических, агрономических специальностях ВУЗов.

Положения, выносимые на защиту.

• Технология идентификации почвенных бактерий и грибов, определения степени генетической гетерогенности и изобилия почвенной микробиоты по анализу первичной структуры ДНК генов 18S - и 16S рРНК.

• Методики использования статистических методов многомерного анализа для характеристики сообществ почвенных микроорганизмов.

• Степень влияния экологических факторов на генетическое разнообразие бактерий и грибов, на структуру сообществ этих микроорганизмов.

• Зависимость структуры почвенного микробоценоза в ризосфере от видовых особенностей растения-хозяина.

Апробация работы. Основные положения диссертационной работы были доложены: на Международной конференции «Современное состояние и перспективы развития микробиологии и биотехнологии» (Минск, 2010); на 4-х международных симпозиумах Международной ассоциации наук о растительности (1АУ8): 52-ой (г. Публикация результатов работы. По материалам диссертации опубликовано 16 научных работ в т.ч. 5 статей в изданиях, включенных в перечень ВАК РФ.

Структура и объём работы. Диссертация состоит из введения, обзора литературы, методов исследования, результатов и их обсуждений, выводов, списка литературы включающего 174 наименований отечественных и зарубежных авторов. Диссертация изложена на 144 страницах, иллюстрирована 28 рисунком, 12 таблицами и приложением.

ВЫВОДЫ

• Разработаны новые технологии идентификации и классификации почвенных микроорганизмов с использованием методов ДГГЭ и пиросеквениро-вания ДНК генов 18S и 16S рРНК. Предложен новый оригинальный алгоритм компьютерного анализа таксономических групп популяций почвенных микроорганизмов и измерения уровня генетического разнообразия почвенной микробиоты при действии различных экологических факторов.

• В изученных образцах почв идентифицировано 766 таксонов бактерий (в т.ч. - 28 родов архебактерий), принадлежащих к 611 родам, 178 семьям, 86 порядкам, 40 классам, 32 типам. Показано, что наибольшее влияние на формирование структуры бактериальных сообществ почв оказывают тип ландшафта, кислотность почвы и содержание в ней ионов фосфора.

• Количество азотфиксирующих и сульфат восстанавливающих бактерий в почве коррелирует с содержанием в ней минеральных питательных элементов. В пойменных почвах, богатых сульфат - ионами и бедных азотом, количество сульфат восстанавливающих бактерий и Acidobacteria в 2,5 выше, а количество азотфиксирующих бактерии в 4 раза ниже, чем в сухих почвах со склона реки.

• В изученных образцах почв выявлено 871 вид грибов, принадлежащих к 18 классам, 61 порядку, 199 семействам, 574 родам. Структура сообществ ризосферных грибов зависит, в первую очередь, от вида растения-хозяина и типа ландшафта.

• Доминирующими в почвенных сообществах, среди идентифицированных видов грибов, оказались представители отдела Ascomycota (90-96 %). По обилию зигомицетов, грибные сообщества в ризосфере трех видов исследованных растений, существенно не различались.

• В почвенных образцах было выявлено 12 видов грибов из отдела С1отеготусо1а, принадлежащих к 3 порядкам. Показано, что структура и изобилие арбускулярно - микоризных сообществ определяется, в основном, типом ландшафта и видом растения-хозяина.
