Рецепторы активаторов пролиферации пероксисом (PPAR) принадлежат к семейству ядерных рецепторов. Эти транскрипционные факторы участвуют в регуляции экспрессии генов, ответственных за липидный и глюкозный гомеостаз, а также генов, участвующих в ответе на провоспалительные стимулы. В настоящее время описано три изотипа PPAR: PPARa, PPAR(3 и PPARy. Нарушение в функционировании PPAR-опосредованных путей может приводить к развитию целого ряда заболеваний, таких как: ожирение, диабета 2 типа, сердечно-сосудистые заболевания, рак, гипертония, нейродегенеративные заболевания и хроническое воспаление. В связи с большой медицинской значимостью PPAR и регулируемых ими процессов эти транскрипционные факторы активно изучаются.

Некоторые из синтетических агонистов PPAR уже являются действующими веществами ряда лекарств (Авандия, Актос, Трайкор, Безалип). В частности, фибраты — синтетические агонисты PPARa — используются в качестве гиполипидемических лекарств, а PPARy является мишенью такого класса гипогликемических лекарственных средств, как тиазолидиндионы, которые используются в терапии диабета 2 типа. В последнее время показано, что синтетические PPARa и PPARy агонисты обладают противовоспалительной и антиатеросклеротической активностями. Синтетические агонисты PPARJ3 предлагается использовать для лечения дислипидемии, ожирения и нарушений механизмов восстановления и регенерации тканей. Эффективность применения синтетических агонистов PPAR для терапии таких дисфункций мозга как инсульт, травма, болезни Альцгеймера, Паркинсона, Хантингтона и др. была показана на животных. Считается, что нейропротективные свойства агонистов PPAR связаны с их противовоспалительными эффектами, поэтому в настоящее время актуальны исследования механизмов действия синтетических агонистов PPAR при воспалительных ответах мозга, которые моделируются как на уровне целого организма, так и на уровне отдельных типов клеток.

В развитии воспалительных процессов в мозге ключевая роль принадлежит клеткам глии. Среди глиальных клеток следует выделить астроциты, так как они являются важными участниками воспалительных процессов, протекающих в мозге, и нарушение их функционирования приводит к развитию нейродегенеративных заболеваний. Более того, ранее было показано, что агонисты PPARy и PPARa могут модулировать воспалительные ответы в астроцитах. Также было показано, что в первичных астроцитах экспрессируются все три изотипа PPAR. Эти свойства астроцитов делают их важным объектом для изучения роли PPAR в воспалительных заболеваниях мозга.

Важно также отметить, что астроциты являются основным источником полиненасыщенных жирных кислот и простагландинов — участников воспалительных процессов и эндогенных лигандов PPAR. Ключевыми ферментами продукции полиненасыщенных жирных кислот и простагландинов являются фосфолипазы А2 и циклооксигеназы, соответственно. Эти ферменты относятся к так называемым PPAR лиганд-синтезирующим ферментам. Имеются сведения, что применение синтетических агонистов PPAR может приводить к изменениям уровней экспрессии фосфолипаз и циклооксигеназ, однако влияние лигандов PPAR на экспрессию этих ферментов в астроцитах не изучено.

При рассмотрении изотипов PPAR в качестве мишеней лекарственных средств можно заметить значительное перекрывание спектров их терапевтических эффектов. Это наблюдение привело к возникновению интереса к синтезу и изучению так называемых двойных агонистов PPAR, которые способны активировать сразу несколько изотипов PPAR. Вещества, обладающие одновременной специфичностью к PPARa и PPARy в данный момент проходят клинические испытания. Однако, при проверке терапевтической ценности двойных агонистов PPAR было обнаружено, что они обладают рядом побочных эффектов, увеличивая риск сердечнососудистых заболеваний и возникновения опухолей. Поэтому для разработки новых лекарств и терапевтических стратегий на базе двойных агонистов PPAR необходимо тщательно изучить механизм и результаты одновременной активации нескольких изоформ PPAR. Необходимость изучения эффектов совместной активации нескольких изоформ PPAR вытекает также из биологии этих ядерных рецепторов, так как их эндогенные лиганды не имеют изотип-специфичности. То есть, натуральные лиганды PPAR сами по себе обладают активностью двойных (тройных) синтетических агонистов и способны активировать сразу несколько изоформ PPAR. Таким образом, изучение эффектов одновременной активации нескольких изоформ PPAR является важной задачей при изучении механизма воспалительного ответа.

В последние годы было охарактеризовано влияние агонистов PPARa и PPARy на различные клеточные ответы астроцитов (пролиферацию, выброс цитокинов, и др.), однако экспрессия самих PPAR при этом не изучалась, хотя известно, что изменение экспрессии PPAR является одним из важных механизмов регуляции этих транскрипционных факторов. Недавно было предположено наличие функциональных взаимодействий между изотипами PPAR. Так было показано, что за счет активации PPARP может возникать репрессия PPARy- и PPARa-опосредованной активации транскрипции, а также была описана PPARP-зависимая активации PPARy. Не смотря на то, что ряд данных позволяет предположить наличие взаимодействий между изотипами PPAR на уровне взаимной регуляции их экспрессии, в настоящий момент практически не существует работ, посвященных исследованию взаимодействий всех трёх изотипов PPAR.

Способность PPAR регулировать экспрессию своих лиганд-синтезирующих ферментов с одной стороны, и участие лигандов в регуляции экспрессии самих PPAR с другой стороны, указывают на наличие сложной системы взаимодействий между фосфолипазами А2, циклооксигеназами и PPAR. Тем не менее, до сих пор не было попыток изучить эти белки и взаимосвязи между ними как единую систему функционально взаимодействующих белков. Таким образом, целью данной работы было исследовать взаимосвязь между изоформами PPAR а, р, у и их лиганд-синтезирующими ферментами циклооксигеназами и фосфолипазами А2; установить роль этой взаимосвязи при ответе астроцитов на провоспалительную стимуляцию.

5. Выводы

По результатам проделанной работы можно сделать следующие выводы:

1. Исследовано влияние синтетических агонистов ядерных рецепторов PPAR на экспрессию генов самих ядерных рецепторов PPAR а, (3, у и ферментов классов фосфолипаз А2 и циклооксигеназ. Показано, что синтетические агонисты изоформ PPAR влияют на экспрессию этих генов.

2. Изучена взаимосвязь между изоформами PPAR а, (3 и у в астроцитах, стимулированных LPS. Показано, что PPARy увеличивает, a PPARa снижает уровень экспрессии и активность PPAR(3; PPARp увеличивает уровень экспрессии и активность PPARa.

3. Показано положительное влияние агонистов PPARy и отрицательное влияние агонистов PPARa на экспрессию СОХ-2 в астроцитах, стимулированных LPS. Установлено, что механизм действия этих агонистов заключается в модуляции активности PPARP, который играет ключевую роль в регуляции экспрессии СОХ-2.

4. В результате исследования влияния ядерных рецепторов PPAR a, Р и у на экспрессию цитозольной и секреторной фосфолипаз А2 в астроцитах, стимулированных LPS, установлено, что экспрессия цитозольной фосфолипазы А2 находится под положительным контролем PPARa, Р и у, а экспрессия секреторной фосфолипазы А2 находится под положительным контролем PPARy.

5. Показано, что активность цитозольной фосфолипазы А2 и СОХ-2 необходима для PPAR-опосредованной регуляции экспрессии СОХ-2 в астроцитах, стимулированных LPS. Установлено, что в данной системе эти ферменты продуцируют эндогенные лиганды PPARp.
