VI. выводы
1. Устойчивость к додецилсульфату натрия, детектируемая по изменению электрофо-ретической подвижности в полиакриламидном геле с ДСН, может быть использована в качестве характеристического свойства правильно собранных фибритинов.
2. Показано свойство фибритина и его мутантов, содержащих интактный С-концевой домен, восстанавливать тримерность в присутствии 2% додецилсульфата натрия при температуре -20°С.
3. Фибритин содержит 3 домена, устойчивых к додецилсульфату натрия - один соответствует N-концевой части coiled coil; второй - С-концевой части coiled coil; и третий - С-концевому ß-структурному домену. Тримеризация в присутствии ДСН происходит за счет С-концевого домена (фолдона).
4. Петля L11 (ближайшая к С-концевому домену) несущественна для фолдинга, но стабилизирует структуру собранного белка.
5. Сконструирован мутант фибритин В1, для которого получены кристаллы и решается пространственная структура методом рентгеноструктурного анализа.
6. Получены мутанты фибритина, способные к правильной сборке, будучи секрети-рованными в периплазму Е. coli. Этот результат является еще одним аргументом в пользу гипотезы о том, что сборка фибритина в тример протекает пост-трансляционно.
7. Сконструирован пермутантный фибритин, имеющий фолдон на N-конце. Такой белок демонстрирует свойства tsf (temperature-sensitive folding)-мутанта и при пермиссивных условиях (27°С) собирается в стабильную компактную структуру, представляющую собой тримерный coiled coil.
8. По крайней мере, при ренатурации in vitro первым этапом является взаимодействие С-концевых участков, затем происходит формирование coiled coil структуры. С-концевая последовательность является обязательным условием формирования тримеров.
