Протеолиз и участвующие в нем протеиназы регулируют важнейшие этапы жизнедеятельности и метаболизма клетки. Протеиназы важны для всех живых организмов, включая вирусы, бактерии и эукариотические организмы. Кроме того, протеолиз является неотъемлемой частью, а в ряде случаев и причиной многих патологических процессов. Так вирулентные протеазы играют важную роль в инфекционных заболеваниях, вызываемых бактериями и вирусами. Многие вирусы, такие как ВИЧ, гепатит С, вирусы Лихорадки Западного Нила, Желтой Лихорадки, полиомиелита и гриппа содержат гены протеиназ, необходимые для жизненного цикла вируса, преодоления механизмов клеточной защиты и контроля над метаболизмом клеток. Секреторные протеиназы патогенных бактерий зачастую являются их важнейшими вирулентными факторами. Протеиназы активируют белковые токсины бактерий. Абнормальный протеолиз характерен для роста и метастазирования злокачественных опухолей.
Цель нашей работы состояла в исследовании роли фуриноподобных протеиназ и металлопротеиназы МТ1-ММР в патологических процессах.
Первой задачей нашей работы являлось исследование взаимодействия человеческих фуриновых протеиназ с транспортными белками и ферментами, определяющими токсичность летального токсина сибирской язвы. Эта система включает: токсин сибирской язвы, рецепторы токсина, находящиеся на поверхности человеческих клеток, и человеческие фуриновые протеиназы, необходимые для процессинга компонентов токсина и его последующей интернализации человеческой клеткой.
Второй задачей было изучение свойств вирусной фуриноподобной протеиназы NS3, которая играет важную роль в инфекционном механизме вируса Лихорадки Западного Нила. Для изучения двухкомпонентной сериновой протеиназы, кодируемой геномом вируса, мы использовали рекомбинантную протеиназу, модельные белки и субстраты, что позволило нам избежать работы с высокоинфекционным и опасным вирусом.
Изучение протеолитического механизма, определяющего онкогенный эффект матриксной металлопротеиназы МТ1-ММР, стало третьей задачей нашего исследования. Нами были идентифицированы новые внутриклеточные белковые субстраты мембранной металлопротеиназы МТ1-ММР. В результате МТ1-ММР зависимого внутриклеточного гидролиза центросомальных белков возникают нарушения митотического веретена деления, что приводит к генетической нестабильности, хромосомным аберрациям и анеуплоидии. Анеуплоидия, по современным представлениям, является одним из главных факторов злокачественной трансформации клеток.
Важность протеолиза в патологических процессах несомненна, и по мере увеличения количества информации в периодической научной литературе только продолжает расти. Понимание того, как функционируют протеиназы в живых организмах, способствует разработке новых методов детекции, прогноза и лечения многих заболеваний. Идентифицированные в данной работе свойства и функциональные параметры фуриноподобных протеиназ и мембранной металлопротеиназы МТ1-ММР не были известны ранее. Полученные нами данные могут быть использованы при создании новых эффективных и безопасных ингибиторов сибирской язвы, ряда вирусных заболеваний и злокачественных новообразований.

1. Идентифицирован и охарактеризован новый механизм гетероолигомеризации протективного антигена летального токсина сибирской язвы, позволяющий преодолеть дефицит про-протеинконвертаз хозяйской клетки-мишени. Использование этого механизма позволяет возбудителю сибирской язвы успешно атаковать многие дополнительные типы клеток, дефицитных по про-протеинконвертазам.
2. Определена субстратная специфичность процессирующей протеиназы NS3 вируса Лихорадки Западного Нила. Установлено, что эта протеиназа по субстратной специфичности наиболее близка к фуриноподобным протеинконвертазам. Знание этого феномена позволило нам идентифицировать ряд эффективных антагонистов фуриноподобной протеиназы NS3.
3. Установлено, что основной белок миелина является субстратом вирусной протеиназы NS3, его гидролиз может иметь важное значение в развитии симптомов энцефалопатии, вызываемых вирусом Лихорадки Западного Нила.
4. Установлена новая внутриклеточная функция матриксной металлопротеиназы МТ1-ММР. Эта функция связана с гидролизом интегрального центросомального белка перицентрина металлопротеиназой МТ1-ММР, что ведет к хромосомной нестабильности и превращению нормальных клеток в злокачественные.
