ВВЕДЕНИЕ

АКТУАЛЬНОСТЬ ПРОБЛЕМЫ

Полиамины присутствуют практически во всех видах биологических организмов, от бактерий и вирусов до человека, и участвуют в огромном количестве разнообразных биологических и биохимических реакций, включая синтез ДНК, РНК и белков [1-3]. Исследование клеток прокариот и эукариот показало, что в различных организмах функционируют не только одинаковые соединения полиаминов (чаще всего это путресцин, спермидин, спермин и, существенно реже, кадаверин), но также универсальны и многие этапы биосинтеза этих поликатионов [4,5]. Хотя не вызывает сомнений тот факт, что полиамины выполняют важные физиологические функции и совершенно необходимы для обеспечения нормального клеточного роста, показано, что избыточное накопление полиаминов в клетке ингибирует синтез белков, вызывает замедление роста, а в некоторых случаях приводит к гибели организма [6]. Ацетилирование полиаминов по так называемому ацетилазному пути деградации, помогает предотвратить эффект токсичности полиаминов как в про-, так и в эукариотах [4,7]. Образующиеся при этом ацетилпроизводные полиаминов далее окисляются полиаминоксидазами, деацетилируются, либо экскретируются из клетки [4,8]. У некоторых микроорганизмов охарактеризован также альтернативный аминотрансферазный путь деградации полиаминов.

Наиболее подробно изучен аминотрансферазный путь деградации путресцина в клетках Escherichia coli. Показано, что трансаминирование путресцина, катализируемое путресцин аминотрансферазой (КФ 2.6.1.29), и последующее декарбоксилирование образующегося аминобутиральдегида приводят к образованию у-аминобутирата (GABA), который в дальнейшем преобразуется в сукцинат, включаясь в цикл Кребса [9]. Совокупность реакций биосинтеза путресцина из аргинина и его деградации до сукцината образуют так называемый аргинин декарбоксилазный путь деградации аргинина (ADC) E.coli. Было показано, что все ферменты ADC пути, за исключением аргинин декарбоксилазы, индуцируются в условиях азотного голодания. Однако, несмотря на то, что ADC путь деградации E.coli достаточно хорошо охарактеризован как биохимически, так и генетически, еще не все гены этого пути картированы. Так до сих пор не были локализованы гены, кодирующие ключевые ферменты деградации путресцина -пиридоксальфосфат (PLP) - зависимую путресцин:а-кетоглутарат (а-КГ) аминотрансферазу (ПАТазу) и аминобутиральдегид дегидрогеназу (ABADDHa3y). Вследствие того, что уровень активности ПАТазы даже в индуцирующих условиях азотного голодания чрезвычайно низкий, изучение этого фермента до сих пор проводилось только в мутантных по утилизации полиаминов и GABA штаммах E.coli. Возможно, по этой причине в независимо выполненных работах Kim [10] и Prieto-Santos [11] приведены противоречивые биохимические характеристики частично очищенных препаратов ПАТаз.

В этой связи актуальной является задача идентификации гена E.coli, кодирующего ПАТазу, с использованием методов биоинформатики и генетической инженерии, выделение нативного фермента и изучение его свойств современными методами молекулярной биологии

ЦЕЛЬ И ЗАДА ЧИ РАБОТЫ

Целью представленной работы являлась идентификация гена E.coli, кодирующего ПАТазу, а также конструирование плазмид, обеспечивающих достаточно высокий уровень продукции этого фермента и определение основных физических и кинетических параметров очищенного препарата ПАТазы. В процессе работы были решены следующие задачи:

Идентификация гена, кодирующего ПАТазу, в хромосоме штамма E.coli К12 (ген ygjG)-,

Изучение Ntr-зависимой регуляции экспрессии генаygjG;

Конструирование экспрессионных плазмид, обеспечивающих эффективную продукцию ПАТазы как в виде слитого белка с his6-tag лидерным пептидом (Ht-YgjG), так и нативного белка (YgjG);

Разработка лабораторного метода выделение белка YgjG из растворимой фракции клеточных лизатов рекомбинантного штамма E.coli;

Исследование и сравнительная характеристика каталитических активностей и некоторых основных физических и кинетических параметров очищенных препаратов YgjG и Ht-YgjG.

НАУЧНАЯ НОВИЗНА И ПРАКТИЧЕСКАЯ ЗНАЧИМОСТЬ РАБОТЫ

Впервые идентифицирован и клонирован ген (ygjG), кодирующий путресцин аминотрансферазу в клетках Escherichia coli К12. Изучена его структурная организация и выявлены регуляторные последовательности, обеспечивающие о54 -зависимую индукцию экспрессии ygjG в условиях азотного голодания.

Разработана методика получения высокоочшценного препарата каталитически активного белка YgjG из биомассы рекомбинантного штамма E.coli.

Показано, что фермент эффективно катализирует путресцин :а-КГ аминотрансферазную реакцию, а также может использовать кадаверин и, с меньшей эффективностью, спермидин в качестве доноров аминогрупп. Фермент термостабилен, активен в щелочном диапазоне значений рН и проявляет максимум активности при рН 9.0 и температуре 70 °С. Интересной особенностью фермента является его способность катализировать глутамат:пируват (GPT, КФ 2.6.1.2) аминотрансферазную реакцию. Ни одна из известных к настоящему времени полиамин аминотрансфераз не проявляет GPT активности. Субстратная специфичность и рН зависимость активности YgjG совпадают с параметрами ПАТ азы, выделенной Kim [10] из мутантного по утилизации путресцина штамма E.coli В6.

Сравнение свойств очищенных препаратов YgjG и Ht-YgjG, несущего дополнительную his6-tag лидерную последовательность на N-конце белка, показало, что данная модификация N-концевой последовательности влияет на субстратную специфичность и ферментативную активность препарата. Также, элиминация первых 30 аминокислот YgjG приводит к практически полной потере активности фермента.

Полученные в работе данные могут быть использованы для исследования полиамин аминотрансфераз других микроорганизмов и для дальнейшего изучения путей метаболизма полиаминов в E.coli.

1. Клонирован ген ygjG E.coli К12 MG1655 и показано, что аннотированная как орнитин аминотранефераза ORF ygjG кодирует ПАТазу - белок, обладающий путресцин:а-кетоглутарат аминотрансферазной активностью и эффективно катализирующий реакцию переаминирования кадаверина.

2. Показано, что а54 -зависимая экспрессия ygjG индуцируется в условиях азотного голодания. Определен старт транскрипции мРНК ygjG и стартовый кодон (UUG) трансляции YgjG.

3. Сконструированы рекомбинантные плазмиды, обеспечивающие эффективную продукцию YgjG как в виде слитого с his6-tag лидером полипептида (Ht-YgjG) так и в виде нативного белка. Разработан метод выделения и очистки нативного белка и получены высокоочищенные препараты YgjG и Ht-YgjG.

4. Исследованы субстратная специфичность обоих белков и их основные кинетические параметры в реакциях трансаминирования с использованием путресцина, кадаверина и спермидина в качестве доноров и а-кетоглутарата в качестве акцептора аминогрупп. Определены рН и температурная зависимости ферментативной активности ПАТазы YgjG.
