Термин «прион» появился при исследовании ряда нейродегенеративных заболеваний животных и человека, таких как скрэйпи овец, бычья губчатая энцефалопатия и болезнь Крейцфельда-Якоба человека. Несмотря на то, что эти болезни были известны уже довольно давно, их природа оставалась загадкой вплоть до последнего времени. В частности, долгое время было непонятно, каков механизм возникновения этих нейродегенеративных заболеваний. Этиология этих заболеваний весьма разнообразна. Эти болезни могут наследоваться, возникать инфекционным путём а также спорадически. Такое разнообразие путей возникновения а также очень длительный инкубационный период, характерный для инфекционных форм этих заболеваний, долгое время не давали возможность исследователям выяснить, что же является инициирующим фактором таких болезней. Изначально предполагали, что эти нейродегенеративные болезни вызываются вирусом, однако впоследствии оказалось, что инфекционный агент не содержит ДНК. В 1967 году была выказана «белковая» гипотеза, согласно которой инфекционный агент представлял собой изменённую форму одного из клеточных белков, поддерживающуюся за счёт аутокаталитического механизма (Griffith, 1967). Позднее этот агент был назван «прионом» (Prusiner, 1982). Его необычные свойства обусловлены способностью белка находиться в особом конформационном состоянии, которое может передаваться другим молекулам того же типа путём белок-белковых взаимодействий.

В начале девяностых годов прошлого века прионы были обнаружены и у дрожжей Saccharomyces cerevisiae. В отличие от млекопитающих у дрожжей феномен прионов не связан с какой-либо патологией и имеет, скорее всего, адаптивное значение. Переход белков дрожжей в прионную форму определяет наследование фенотипов, связанных с агрегацией и функциональной инактивацией этих белков. Таким образом, прионные свойства белков дрожжей обуславливают особый механизм эпигенетического наследования признаков, а также регуляцию экспрессии генов на посттрансляционном уровне.

В настоящее время у млекопитающих известен один прионный белок, в то время как у дрожжей S. cerevisiae найдено три таких белка. Ещё один прионный белок выявлен у гриба Podospora anserina. Однако, результаты анализа геномов разных организмов, направленного на поиск последовательностей, гомологичных таковым уже известных прионных белков, позволяют предполагать, что прионы распространены в природе и могут играть важную роль в регуляции физиологических процессов в клетке. Возможно, феномен прионов имеет гораздо большее значение, чем известно сейчас.

Одним из фундаментальных и наименее изученных свойств прионов является существование их наследуемых вариантов (штаммов). Это свойство проявляется как у прионов млекопитающих, так и у прионов дрожжей. Белок, обладающий прионными свойствами, способен не только к конформационному переходу из неприонного состояния в прионное. По всей видимости, он способен приобретать множество прионных конформаций, которые и определяют существование вариантов приона. Данная работа посвящена исследованию молекулярных основ возникновения разных вариантов прионного состояния белка Sup35 дрожжей S. cerevisiae. Более конкретно, работа представляет собой попытку изучения роли аминокислотных повторов белка Sup35 в существовании вариантов приона, образованного этим белком.

1. Встраивание белков Sup35, имеющих меньше четырёх аминокислотных повторов, в полимеры полноразмерного белка Sup35 приводит к увеличению частоты потери [PSI+]

2. Минимальная длина прионного домена Sup35, достаточная для поддержания «сильных» и «слабых» вариантов [PSI+], равна 64-м и 83-м N-концевым аминокислотным остаткам, соответственно

5. Заключение

Прионы дрожжей вызывают существенный интерес поскольку, во-первых, сами по себе представляют новое генетическое явление, а во-вторых, являются удобной моделью для изучения фундаментальных свойств, присущих феномену прионов в целом. Одним из таких свойств является феномен штаммового разнообразия прионов. В данной работе была предпринята попытка изучения этого феномена на примере детерминанта [PS/1"] дрожжей Saccharomyces cerevisiae. Целью данной работы являлось выяснение роли области NR прионного домена белка Sup35 в поддержании различных вариантов приона [PS74]. Для осуществления этой цели в работе были поставлены следующие экспериментальные задачи: 1) исследовать возможность поддержания [PST1-] белками Sup35 с уменьшенным количеством аминокислотных повторов области NR прионного домена, 2) исследовать возможность поддержания вариант-специфичных свойств [PST1"] этими белками.

В результате выполнения этих задач, нам удалось определить минимальный участок прионного домена, необходимый для поддержания «сильных» и «слабых» вариантов [PS/4-]. Мы показали, что удаление довольно значительной области прионного домена, а именно последовательности с 65 по 112 а. к. не препятствует поддержанию [PST4-]. Таким образом, мы доказали, что информация о прионной укладке может быть сохранена оставшейся частью прионного домена белка Sup35.

Кроме того, мы показали, что длина области NR прионного домена белка Sup35 может влиять на спектр прионных конформаций, приобретаемых этим белком. Наши данные подтверждают предположение о том, что вариабельность приона [PS/4] определяется возможностью различной прионной укладки области NR прионного домена белка Sup35. Мы также обнаружили, что четыре делеционных производных белка Sup35 с разным количеством аминокислотных повторов области NR вызывают вариант-специфичную элиминацию [PS74] и изучили способность этих белков встраиваться в прионные полимеры, образованные белком Sup35 дикого типа. Помимо этого, в работе был разработан новый метод анализа распределения белка Sup35 между прионной и неприонной формами. Этот метод может быть применён для исследования прионных и неприонных амилоидов как высших, так и низших эукариот.

Мы надеемся, что полученные в данной работе результаты о молекулярных основах вариабельности прионов дрожжей имеют несомненный общебиологический интерес. Новые факты, касающиеся вариантов прионов дрожжей, могут стимулировать аналогичные исследования прионов млекопитающих. Кроме того, обнаружение мутантных молекул прионного белка дрожжей, способных элиминировать прионное состояние, может дать импульс к поиску новых мутантных форм прионного белка человека, которые могут быть применены для лечения прионных заболеваний.
