Понимание преимуществ и эффективности проведения твердофазного синтеза пептидов в проточном режиме, когда реагенты непрерывно циркулируют через плотный слой пептидилполимера, пришло в конце 1970-х годов. Однако первые попытки прямой реализации этого подхода принесли скорее разочарование и поставили вопрос о принципиальной совместимости гелевых полимеров, объём набухания которых подвержен значительным колебаниям, и обычных реакторов колоночного типа, объём которых жёстко фиксирован. Как следствие, в реакторе возникают пустоты или неприемлемо высокое давление в потоке (70 атм. и выше) [1-4] из-за спрессовывания набухающего полимера, что, в совокупности, уничтожает все преимущества проточного режима. Актуальность радикального решения этой проблемы, в котором бы учитывалась природа гелевых полимеров, многократно возросла с бурным развитием твердофазной комбинаторной химии, однако все частные её решения, предлагавшиеся нашими коллегами, носят компромиссный характер и имеют одну общую черту: они основаны на маскировании или подавлении гелевой природы полимерного реагента [5,6] с целью улучшить его совместимость со стандартным проточным реактором.

Другая проблема современного твердофазного синтеза пептидов, с которой связана эта работа, - это отсутствие универсальных неинвазивных методов мониторинга, способных распознавать трудные участки сборки полипептидной цепи в реальном времени и независимо от используемой химической методологии. От доступности таких методов, дающих основу для своевременной корректировки и оптимизации процесса, серьёзным образом зависит надёжность и результат автоматического твердофазного синтеза. Важнейшие достижения в этой области демонстрируют синтезаторы MilliGen 9050+, ABI 432А, ABI 433А [7], в которых реализовано управление с обратной связью в сочетании со спектрофотометрическим [8] и/или кондуктометрическим [9] мониторингом, однако область применимости обоих методов ограничена Fmoc-химией. На этом фоне более экономичная и лучше развитая Вос-методология оказывается в невыгодном положении, что определяет высокую актуальность разработки Вос-совместимых методов мониторинга вообще и неинвазивных автоматизируемых подходов, в особенности.

I. ЛИТЕРАТУРНЫЙ ОБЗОР ПРОБЛЕМЫ МОНИТОРИНГА ТВЕРДОФАЗНОГО СИНТЕЗА ПЕПТИДОВ

Большая часть собранной информации представлена в таблицах 1 и 2. В первой таблице краткая информация об известных методах организована на двух уровнях: все работы по каждому методу расположены в подразделах таблицы в хронологическом порядке, причем такие подразделы также представлены в порядке, который отражает дату выхода пионерской публикации по данному подходу. Этим я также хотел воздать должное первопроходцам, имена которых, увы, выпадают из поля зрения современных деятелей твердофазной органической химии. Во второй таблице собрана вся доступная мне информация об особенностях функционирования серийных синтезаторов пептидов, имеющих системы мониторинга. Наконец, во вспомогательной таблице 3 собраны УФ-спектральные характеристики молекул, имеющих отношение к теме обзора.

ЗАКЛЮЧЕНИЕ

Использование реактора предложенной конструкции обеспечиваете функционирование гелевых полимеров в полноценном проточном режиме кс позволяет осуществлять непрерывную регистрацию изменений объёма, набухания полимерного реагента, положенную в основу разработкой: свеллографического мониторинга.

Свеллографический мониторинг является новым инструментальных^ методом исследования набухания гранулированных поперечно-сшитых гелевых! полимеров, который делает возможными прямое наблюдение динамики^ набухания в реальном времени и быструю регистрацию «спектра набухаемости» образца полимера в широком наборе растворителей.

В контексте твердофазного синтеза пептидов, свеллографический подходх можно охарактеризовать как совместимый с любой химической методологией^ неинвазивный метод мониторинга. Он позволяет легко документировать каждую операцию протокола и снизить расход растворителей, как MHHHMyivx вдвое, относительно существующих стандартов.

Установлено, что динамика изменения объёма набухания: пептидилполимера в ходе наращивания цепи «в норме» характеризуется линейной зависимостью от прироста массы, а отклонения от линейносттз: указывают на наступление агрегации. Предложена теоретическая интерпретация этих явлений на молекулярном уровне.

Извлечение информации о кинетике реакций конденсации и: деблокирования из свеллографического профиля не представляется возможными, равно как и прямое использование в качестве критерия; завершённости этих реакций момента стабилизации объёма набухания. Это связано с исключительно высокой чувствительностью последнего :ес сольватационным эффектам.

• Высокая чувствительность свеллографического метода к изменению сольватации пептидилполимера определяет и его главное достоинство -возможность детектирования ранних признаков агрегации на стадии конденсации независимо от используемой химической методологии. Этот процесс может быть легко автоматизирован и использован в управляющей программе синтезатора для существенной оптимизации синтеза «трудных» пептидов.
