До недавнего времени общепринятым было представление о том, что эукариотические организмы имеют линейные хромосомы, а бактериальные геномы представляют собой кольцевые молекулы. Однако, в течение последних двадцати лет это правило перестало быть универсальным, поскольку линейные плазмиды и хромосомы были обнаружены в различных прокариотических организмах. Первым обнаруженным линейным репликоном у прокариот является умеренный бактериофаг N15, который в лизогенном состоянии не интегрируется в хромосому Е. coli, а представляет собой линейную плазмиду с ковалентно замкнутыми теломерами. Позднее было установлено, что аналогичную структуру ДНК имеют линейные хромосомы и плазмиды спирохет рода Borrelia (Barbour and Garon, 1987; Fraser et al., 1997), одна из хромосом Agrobacterium tumefaciens (Allardet-Servent et al., 1993; Goodner et al., 1999), a также профаги умеренных бактериофагов PY54 (Hertwig et al., 2003) и фК02 (Casjens et al., 2003).

В то же время, информация о молекулярных механизмах репликации линейных хромосом с ковалентно замкнутыми теломерами у прокариот весьма ограничены. В частности, до проведения этой работы практически отсутствовали данные о свойствах репликационных белков линейных плазмид и механизмах регуляции их репликации и числа копий, необходимые для разработки моделей репликации. Основное внимание в настоящей работе посвящено изучению этих проблем на примере умеренного бактериофага N15. Тот факт, что N15 является не просто плазмидой, а умеренным бактериофагом, позволяет предположить, что репликация его ДНК может следовать разным механизмам в плазмидном состоянии и при литическом развитии, с участием различных репликационных белков.

Одним из практических применений фундаментальных знаний в области репликации плазмид и контроля экспрессии их генов является конструирование векторов для продукции целевых белков. Escherichia coli является одним из наиболее привлекательных объектов, используемых для продукции гетерологичных белков, вследствие того, что она способна к быстрому росту до высоких концентраций на простых средах, ее генетика достаточно хорошо охарактеризована, доступен большой набор клонирующих и экспрессионных векторов и мутантных штаммов.

Идеальный экспрессионный вектор должен обеспечивать высокий уровень синтеза продукта в случае индукции при минимально низком базовом уровне экспрессии, что необходимо, в первую очередь, при экспрессии продуктов, которые могут быть токсичны для клетки. Несмотря на это, большинство современных систем для синтеза гетерологичных белков в Е. coli обладают относительно узким диапазоном регулирования. Вследствие чего системы, обладающие низким базовым уровнем экспрессии, не позволяют достичь высокого уровня синтеза продукта при полной индукции, а системы с высоким максимальным уровнем имеют обычно и высокий базовый уровень экспрессии, что делает их мало пригодными для получения чужеродных белков, которые могут быть токсичными для бактериальной клетки. Таким образом, задача создания экспрессионных систем, сочетающих высокий максимальный уровень экспрессии продукта с большим диапазоном регуляции (т.е. низким базовым уровнем), продолжает оставаться актуальной.

Основная идея нашего подхода состоит в использовании регулируемого промотора в сочетании с плазмидным репликоном, число копий которого может регулироваться в широком диапазоне, от 3-4 до 100 на хромосому. Диапазон регуляции синтеза продукта раширяется за счет перемножения эффектов активации промотора и эффекта дозы гена. Для создания этих систем экспресии были использованы репликон и регуляторные элементы бактериофага N15.

ЦЕЛЬ И ЗАДАЧИ ИССЛЕДОВАНИЯ

Основная цель работы состояла в функциональной характеристике генов, обеспечивающих репликацию ДНК фага N15, изучении механизмов регуляции числа копий профага и конструировании на основе репликона N15 векторов для экспрессии целевых белков в Е. coli.

При этом были поставлены следующие основные задачи:

1. Исследование механизмов регуляции числа копий профага N15 и миниплазмид на его основе.

2. Изучение механизмов действия антирепрессоров N15 и их вляние на репликацию и число копий профага.

3. Функциональная характеристика репликационного гена герА и установление роли репликационных белков клетки-хозяина в репликации профага N15 и ДНК фага N15 в ходе литического развития.

4. Конструирование и характеристика экспрессионных векторов на основе репликона N15, число копий которых может регулироваться.

выводы

1. Идентифицирован промотор репликационного гена герА фага N15 и показано, что он может быть репрессирован белком СВ фага. Инактивация СВ репрессора в результате связывания с ним белков-антирепрессоров приводит к активации промотора герА и увеличению числа копий профага N15.

2. Репликационный белок RepA фага N15 участвует в инициации репликации и обладает активностями праймазы и хеликазы.

3. Инициаторный белок DnaA Escherichia coli не требуется для инициации репликации N15 и является негативным регулятором числа копий миниплазмид, основанных на репликоне№5.

4. На основе репликона фага N15 сконструированы два вектора для экспрессии целевых белков в клетках Е. coli, содержащих регулируемые промоторы. Сконструирован штамм Е. coli, в хромосому которого интегрирован ген антирепрессора N15 под контролем промотора арабинозного оперона, позволяющий активировать репликацию и увеличивать число копий векторов. Диапазон регуляции синтеза продукта в созданных системах экспрессии расширен за счет перемножения эффектов активации промотора и увеличения числа копий вектора.
