Выводы:
1. Заряженные аминокислотные остатки аспартата, глутамата, аргинина, лизина и гистидина не только стабилизируют альфа-спирали в пептидах и белках, но и обладают уникальным свойством инициировать образование элементов вторичной структуры такого типа. Следствием данного процесса является наблюдаемое пограничное расположение таких остатков — до или после спирального участка в последовательности.
2. Механизмы инициации «прямой» и «обратной» альфа-спирализации олигопептидов имеют общую природу. Ключевой вклад в этот процесс вносят боковые группы кислых и основных аминокислотных остатков, формирующие бифуркационные водородные связи с остовом пептида, приводящие к «спиральному» закручиванию его структуры.
3. В водной среде предлагаемый механизм инициирования первого витка альфа-спирали сохраняется, однако динамика спирализации растворенной молекулы оказывается замедленной, по крайней мере, на два порядка.
4. Предлагаемый механизм запуска образования альфа-спирали не противоречит экспериментальным данным и может использоваться для уточнения предсказательных алгоритмов определения содержания такого типа вторичной структуры в новых пептидах.
Заключение:
Исследование особенностей электронной структуры отдельных аминокислот, коротких пептидов, а также исследование динамического поведения таких молекул является важной фундаментальной задачей, которая может успешно решаться с помощью современных методов компьютерного молекулярного моделирования.
В данной работе впервые в едином приближении проведены детальные и последовательные исследования сразу нескольких классов молекул: аминокислот, модельных монопептидов и олигопептидов; показана возможность формирования бифуркационной водородной связи в боковых группах кислых аминокислотных остатков и обоснована ключевая роль такой связи в инициировании альфа-спиральной нуклеации на 1Ч-конце спиральных участков; методическая новизна работы заключается в совместном использовании двух дополняющих друг друга теоретических подходов — квантово-химического и молекулярно-динамического, что позволило связать особенности электронной структуры заряженных остатков с их ролью в динамическом поведении пептидов.
Подробно исследованные особенности электронного строения пептидов и аминокислотных остатков позволили обосновать ключевую роль боковых групп кислых и основных аминокислот не только для сталибизации альфа-спиралей, но и для их ускоренного сворачивания. На основе результатов удалось уточнить существующий механизм начала спирализации полипептидной цепи.
Результаты проведённых теоретических исследований имеют большое значение как для понимания фундаментальных процессов сворачивания пептидов и белков, так и для рационального дизайна конструируемых биомакромолекул.
