Aegilops L- род однолетних злаков трибы Triticeae. На основании данных цитогенетического анализа у видов эгилопса выделяют семь основных типов генома: D, S, С, N, U, М и Т. При этом род Aegilops включает в себя виды с различным уровнем плоидности генома (диплоидный, тетраплоидный и гексаплоидный). Было показано, что полиплоидные виды рода представляют собой типичные аллополиплоиды и, таким образом, для рода Aegilops характерен сетчатый характер эволюции. Родственные виды способны обмениваться генетическим материалом, что приводит к повышенной внутривидовой изменчивости и, в ряде случаев, отсутствию четких границ между видами.

Виды Aegilops являются ближайшими родичами культурных пшениц Т. durum и Т. aestivum, и составляют основную часть так называемого вторичного генофонда пшеницы (Triticum L.). Помимо этого, виды эгилопса секции Sitopsis и Aegilops tauschii считаются донорами, соответственно, В и D геномов культурной пшеницы. Этот факт обусловил повышенный интерес к исследованию рода Aegilops. Изучению рода способствовала возможность достаточно легкого получения гибридов, а также малое число и большой размер хромосом у видов рода. Благодаря этому, на настоящий момент род Aegilops является одним из наиболее исследованных в семействе злаков как морфологически, так и методами цитогенетики. Однако, несмотря на постоянный и длительный интерес к роду Aegilops, до сих пор не существует однозначного взгляда на систематику и эволюцию данной группы. Что касается исследования рода с применением новейших молекулярных методов, на настоящий момент, его нельзя считать хорошо изученным, поскольку до сих пор молекулярный анализ рода затрагивал лишь отдельные области генома или отдельные виды эгилопса.

Достаточная охарактеризованность рода с точки зрения морфологии и цитогенетики, а также специфический характер эволюции в данной группе делает род Aegilops интересным объектом в качестве модельного таксона для изучения молекулярной изменчивости при сетчатой эволюции. Помимо этого, исследование видов эгилопса имеет прикладное значение вследствие их таксономической близости к культурной пшенице.

Выводы

1. Впервые с использованием различных молекулярных методов определены уровни меж- и внутривидового полиморфизма всех 26 видов рода Aegilops. Показано существенное различие между видами рода Aegilops по уровням внутривидового полиморфизма генома. Наиболее полиморфными являются диплоидные виды Ае. tauschii, Ае. comosa и Ае. heldreichii, Ае. mutica, Ае. speltoides s.l., Ае. caudata и полиплоидные виды с U геномом - Ае. kotschyi, Ае. variabilis, Ае. columnaris, Ае. biuncialis, Ае. ovata, Ае. triaristata, Ае. recta. Диплоидные виды Ае. uniaristata, Ае. umbellulata, Ае. longissima, Ае. sharonensis, Ае. searsii, Ае. bicornis, полиплоидные виды с D геномом - Ае. crassa, Ае. juvenalis, Ае. vavilovii, Ае. ventricosa, Ае. cylindrica, а также один из полиплоидных видов U-геномной группы - Ае. triuncialis, - обладают более низким уровнем внутривидовых различий. Не наблюдалось полного соответствия между уровнем геномного полиморфизма вида и степенью внутривидовой вариабельности морфологических признаков.

2. В пределах рода Aegilops методами молекулярного маркирования выделено 6 групп, каждая из которых включает в себя либо виды с родственными диплоидными геномами, либо диплоидный вид и родственные ему полиплоиды:

• группа, включающая 5 диплоидных видов, обладающих S геномом (Ае. speltoides s.l., Ае. longissima, Ае. sharonensis, Ае. searsii, Ае. bicornis)',

• группа, включающая 5 видов различного уровня плоидности, объединенных наличием D генома (Ае. tauschii, Ае. crassa, Ае. juvenalis, Ае. vavilovii, Ае. ventricosa)',

• группа из 9 видов различного уровня плоидности, объединенных наличием U генома (Ае. umbellulata, Ае. kotschyi, Ае. variabilis, Ае. columnaris, Ае. biuncialis, Ае. ovata, Ае. triaristata, Ае. recta, Ае. triuncialis)',

• виды Ае. caudata и Ае. cylindrica, обладающие С геномом;

• диплоидные виды с геномами М (Ае. comosa и Ае. heldreichii) и N (Ае. uniaristata)',

• вид Ае. mutica, обладающий Т- типом генома

3. Группы видов Aegilops, выделенные по результатам молекулярного анализа, совпадают с большинством таксонов внутриродовой систематики, выделяемых разными авторами на основании морфологических и цитогенетических данных. Наблюдается практически полное соответствие полученных молекулярными методами данных системам рода Aegilops, которые были предложены X. Кихарой (1954) и М.В. Слагереном (1994); более всего результатам проведенного молекулярного исследования противоречит система рода, предложенная А. Лёве (1984).

4. Охарактеризованы нуклеотидные последовательности внутренних транскрибируемых спейсерных участков (ITS1, ITS2) рибосомного оперона и гена 5.8S рРНК у 22 образцов пяти диплоидных видов рода Aegilops, а также у 26 образцов двух аллополиплоидных видов Ае. kotschyi и Ае. variabilis. Установлена низкая межвидовая вариабельность ITS-районов. Впервые выявлен внутривидовой полиморфизм ITS-последовательности у диплоидных видов рода Aegilops.

5. Впервые с помощью метода домеи-направленного маркирования (DDP-profiling) охарактеризован полиморфизм семейства NBS-LRR генов резистентности у видов Aegilops', показано наличие высокого уровня изменчивости генов данного семейства, по сравнению с другими проанализированными последовательностями генома Aegilops.

6. Клонировано и секвенировано 204 последовательности одного из семейств генов запасных белков эндосперма злаков - генов гамма-глиадинов у 8 диплоидных видов группы Triticum-Aegilops с разными типами генома. Показано наличие в геноме нескольких подсемейств гамма-глиадинов; установлены межвидовые различия по соотношению последовательностей генов и псевдогенов в геноме. На основании анализа полученных данных предложена возможная схема эволюции генов семейства гамма-глиадинов в группе Triticum-Aegilops.
