Молекула бета-казеина - одного из самых распространенных представителей молочных белков - в отличие от типичных глобулярных белков обладает слабо выраженными элементами вторичнойктуры [Creamer et al, 1981; Caessens et al., 1999]. Все полярные и заряженные остатки сконцентрированы на N-конце цепи, в то время как остальная ее часть состоит преимущественно из остатков неполярных аминокислот и содержит большое количество пролиновых остатков [Swaisgood, 1992, 1993, 2003]. В первичной последовательности бета-казеина отсутствует цистеин, что исключает возможность образования внутри- и межмолекулярных дисульфидных связей. Сочетание амфифильного характера и высокой гибкости полипептидной цепи приводит к ассоциации белка в водной среде с образованием наноразмерных коллоидных агрегатов - мицелл [Schmidt, Payens, 1972; Niki et al, 1977; Evans et al, 1979; Swaisgood, 2003]. Именно коллоидные свойства различных казеинов и бета-казеина в частности определяют многие функциональные свойства молока, например, аккумуляция и перенос ионов металлов и белков.
Бета-казеин является удобной моделью для изучения различных аспектов строения коллоидной системы молока и связанных с ней функций. Свойства бета-казеина, как природного поверхностно-активного вещества, активно используются производителями пищевых продуктов. Имеется множество работ фармакологической направленности в области инженерии молочных белков, целью которых является конструирование белков, выполняющих функции целевой сорбции, концентрирования и доставки продуктов, необходимых для лечения ряда серьезных заболеваний.
Строение поверхности коллоидных агрегатов, где гидрофильные участки перемежаются с гидрофобными, их высокая связывающая способность по отношению к гидрофобным соединениям и возможность адаптации геометрии поверхности под связываемый лиганд в последние годы вызвали интерес к шапероноподобной функции бета-казеина [Bhattacharyya, 1999; Morgan et ah, 2005; Barzegar et ah, 2008; Hassanisadi et ah, 2008; Yousefi et ah, 2009]. Коллоидные свойства и шапероноподобная активность бета-казеинов имеют и коммерческий интерес, благодаря их способности стабилизировать белки, микроэмульсии и эмульсии, что определяет текстуру и вкусовые качества пищевых продуктов.
Одним из возможных подходов для получения бета-казеина с новыми физико-химическими и функциональными свойствами является изменение структуры и амфифильных свойств белковой молекулы и, как следствие, свойств образующихся мицеллярных агрегатов. Процесс самоорганизации природного бета-казеина чрезвычайно чувствителен к физико-химическим свойствам среды (полярности и вязкости растворителя, температуре, рН и т.п.), что влияет на размер, структуру и сорбционные свойства его коллоидных агрегатов [Schmidt, 1982; Leclerc, Calmettes, 1997; De Kruif, Grinberg, 2002; Mikheeva et ah, 2003; O'Connell et ah, 2003]. Кроме того, было показано, что точечное введение цистеиновых остатков в полипептидную цепь этого белка приводит к образованию димерных или олигомерных комплексов и значительно изменяет свойства мицеллярных агрегатов (размер, форму, температуру перехода мономер-мицелла, устойчивость к изменению рН и пр.) [Gaudin et ah, 2009, Yousefi et ah, 2009]. При этом вероятно, могут происходить изменения функциональных свойств бета-казеина, в частности его шапероноподобной активности. Вместе с тем, к настоящему времени работ по исследованию взаимосвязи структурных и функциональных особенностей бета-казеина немного и нет ясности в какой степени структура белка определяет эти свойства.
Таким образом, основной целью настоящей работы было выявление вклада структурных особенностей бета-казеина в его ассоциативные, коллоидные и шапероноподобные свойства. В рамках данного исследования использовалось два способа модифицирования свойств белка: введение тиольной группы, то есть дополнительной ковалентной внутри- или межмолекулярной связи, и варьирование полярности растворителя с помощью внесения различных концентраций этанола в раствор белка.
Для достижения поставленной цели необходимо было решить следующие задачи:
1. Получить очищенные препараты рекомбинантного бета-казеина дикого типа и его модифицированных форм, содержащих цистеиновые остатки в различных участках полипептидной цепи белка.
2. Охарактеризовать структуру и процессы самоассоциации рекомбинантных бета-казеинов методами динамического светорассеяния, флуоресценции, кругового дихроизма (КД) и инфракрасной спектроскопии (ИК).
3. Проанализировать влияние температуры и состава растворителя на структуру и свойства бета-казеина.
4. Охарактеризовать шапероноподобную активность природной и рекомбинантных форм бета-казеина по отношению к термо-индуцированной агрегации ряда целевых белков.
Научная новизна работы. Впервые с применением комплекса взаимодополняющих физических методов исследована взаимосвязь структурного состояния рекомбинантных бета-казеинов с цистеиновыми остатками, введенными в различные участки полипептидной цепи белка, с их ассоциативными и шапероноподобными свойствами.
Впервые выполнен всесторонний анализ структурных данных и самоассоциации бета-казеина в смешанном растворителе вода-этанол. Показана корреляция между микрофазным разделением водно-спиртовых растворов, вторичной структурой белка и строением мицелл бета-казеина.
Впервые установлено наличие верхней и нижней границ температурного интервала самоассоциации бета-казеина, при этом увеличение концентрации спирта вызывает понижение как верхней, так и нижней температур мицеллообразования.
Впервые аргументировано, что высокая стабильность вторичной структуры бета-казеина в условиях повышения температуры является кажущейся и обусловлена ограничениями методов разложения спектров КД в приложении к нативно-неупорядоченным белкам.
Впервые исследована шапероноподобная активность бета-казеина в отношении каталитического действия фермента в условиях действия повышенной температуры.
Научно-практическая значимость работы. Полученные результаты уточняют границы коллоидного состояния бета-казеина и дополняют имеющиеся данные о механизмах мицеллообразования белка. Понимание механизмов самоассоциации и взаимодействия бета-казеина с целевыми белками на молекулярном уровне, открывает перспективы для создания белковых препаратов медицинского назначения, исследования и лечения ряда заболеваний, известных как «конформационные болезни». Результаты работы могут быть использованы для прогнозирования влияния температуры и состава растворителя на структуру и функции бета-казеина и других нативно неупорядоченных белков, а также для направленного получения белков с новыми физико-химическими свойствами для пищевой промышленности и биотехнологии.
Полученные экспериментальные данные и методические приемы, изложенные в работе, могут быть использованы в учреждениях биологического, медицинского, биотехнологического, физико-химического профилей, занимающихся исследованием взаимосвязи структуры и функций биомакромолекул, динамики белков в водно-органических средах, а также в учебном процессе при чтении курсов лекций по биохимии, биофизике, физической и органической химии, и молекулярной биологии в ВУЗах.
Связь работы с научными программами и собственный вклад автора в исследования. Исследования проводились в соответствии с планом научных исследований КИББ КазНЦ РАН по теме «Межмолекулярные взаимодействия и молекулярная динамика как факторы регуляции функциональной активности белков» (номер госрегистрации №0120.0
803026) и поддержаны грантом правительства Франции для обучения в совместной аспирантуре, грантами РФФИ, а также грантом Президиума РАН (программа «Молекулярная и клеточная биология»). Научные положения и выводы диссертации базируются на результатах собственных исследований автора.

ВЫВОДЫ
1. Структура и ассоциативные свойства модифицированных форм бета-казеина существенно зависят от места локализации введенного цистеинового остатка. Мицеллярные ассоциаты мономерных форм, содержащих цистеин в начале полипептидной цепи, оказываются менее устойчивыми к температурному воздействию, тогда как мицеллы, образованные олигомерными формами модифицированных бета-казеинов сохраняют стабильность при высоких температурах.
2. Точечное введение цистеина способствует росту внутренней упорядоченности белка, при этом наибольшее влияние на вторичную структуру оказывают замены, затрагивающие гидрофобный домен пептидной цепи бета-казеина.
3. Изменения вторичной структуры и ассоциативных свойств бета-казеина в водно-этанольных растворах определяются структурой растворителя, которая зависит от его состава и температуры. Содержание элементов вторичных структур и размеры мицелл бета-казеина коррелируют с концентрационными границами существования различных микрогетерогенных структур в смешанном растворителе. Показано наличие верхней и нижней границ температурного интервала самоассоциации бета-казеина. В отличие от существующего мнения о спирализации бета-казеина при высоких температурах, достоверно установлено, что увеличение температуры приводит к росту содержания неупорядоченной структуры.
4. На модельных системах (тепловая денатурация алкогольдегидрогеназы, каталазы, иммуноглобулина О) установлено защитное действие бета-казеина, направленное на торможение процесса агрегации и сохранение частичной ферментативной активности целевого белка. Показано, что шапероноподобная активность рекомбинантных бета-казеинов существенно зависит от места локализации цистеинового остатка и структурного состояния белка.
5. Впервые показано, что в условиях температурного стресса все олигомерные формы бета-казеина в молекулярном состоянии проявляют значительный шапероноподобный эффект по отношению к каталитической активности алкогольдегидрогеназы, а в мицеллярной форме этого эффекта не наблюдается.
ЗАКЛЮЧЕНИЕ
Одной из фундаментальных задач биоинженерии и молекулярной биологии является установление связи пространственной организации биомакромолекул с их функциональными свойствами, а также структурой и физико-химическими свойствами индивидуальных растворителей для направленного получения белков с заданными характеристиками.
В настоящей работе для исследования структуры и агрегационных свойств рекомбинантных и нативного бета-казеинов в водных и водно-этанольных растворах применен комплекс физико-химических методов (динамическое светорассеяние, флуоресцентная и инфракрасная спектроскопии, спектроскопия кругового дихроизма). Исследована шапероноподобная активность бета-казеинов по отношению к термо-индуцированной агрегации ряда целевых белков.
Обнаружено, что образование внутри- и межмолекулярных дисульфидных связей, помимо влияния на вторичную структуру, оказывает воздействие на ассоциативные свойства бета-казеина, а также на его способность взаимодействовать с различными белками-субстратами. Точечное введение цистеина в полипептидную цепь бета-казеина способствует росту упорядоченности его вторичной структуры. Наибольшее влияние оказывают замены, затрагивающие гидрофобный домен. Дисульфидные связи, в особенности между гидрофобными участками полипептидной цепи, и, как следствие, димеризация и олигомеризация способствуют самоассоциации рекомбинантных белков.
Показано влияние микрогетерогенности структуры смешанного растворителя на вторичную структуру белка и его коллоидное состояние. В исследуемом интервале концентраций спирта (0 - 50% v/v) бета-казеин претерпевает переход между различными структурными состояниями. Наблюдаемые переходы коррелируют с изменениями в характере межмолекулярных взаимодействий в бинарном растворителе. В области низких концентраций спирта основным фактором, определяющим структуру
