ВВЕДЕНИЕ

Основным условием жизнеспособности организма является поддержание постоянства внутренней среды, что обеспечивается нейроэндокринной системой. Нейроэндокринная система представляет собой сложную высоко иерархическую систему, компоненты которой связаны между собой прямыми и обратными связями (Угрюмов, 1999; Jackson, 1982; Baulieu, 1990; Tanriverdi et al., 2003). Ключевым звеном нейроэндокринной системы является мозг, и, в частности, гипоталамус (Krieger, 1980; Jackson, 1982; Baulieu, 1990). Во взрослом организме эндокринное влияние мозга на периферические органы ограничивается действием вазопрессина и окситоцина, которые, поступая в общую систему циркуляции, влияют на сердечно-сосудистую, выделительную и отчасти репродуктивную системы (Hatton, 1990; Ugrumov, 2002). Гипоталамические физиологически активные вещества (ФАВ), поступая в гипоталамо-гипофизарную портальную систему циркуляции, регулируют секрецию гормонов гипофиза, которые в свою очередь контролируют секрецию гормонов периферических эндокринных желез, таким образом, участвуя в регуляции функций висцеральных органов и самого мозга (Jackson, 1982; Baulieu, 1990). После переноса с кровью из портальной системы циркуляции в гораздо большую по объему общую систему циркуляции ФАВ гипоталамуса многократно разводятся, причем до такой незначительной концентрации, в которой они уже не в состоянии оказывать эндокринное влияние на периферии (Hoffman, 1992). Несмотря на то, что ФАВ вырабатываются не только в гипоталамусе, но и в других отделах мозга, они не могут проникать в общий кровоток из-за гемато-энцефалического барьера (ГЭБ), а, следовательно, и не могут участвовать в прямой эндокринной регуляции периферических органов (Loizou, 1970; Dahlin et al., 2001).

Нейроэндокринная система в онтогенезе, выполняет специфическую функцию регуляции развития целостного организма (Ugrumov, 2010). Это возможно благодаря тому, что на определенном этапе индивидуального развития гипоталамические ФАВ и гормоны эндокринных желез контролируют не столько функциональную активность клеток- и органов-мишеней, сколько их развитие, причем в последнем случае их действие носит необратимый - морфогенетический характер (Lauder, 1993; Ugrumov, 1997; Mirochnik et al., 2005).

До последнего времени считалось, что мозг включается как центральное звено нейроэндокринной регуляции только после полного своего "созревания" (Rajerison et al., 1976; Dlouha et al., 1982). При этом развитие самого мозга контролируется рядом

ФАВ, циркулирующих в крови, и в первую очередь стероидными гормонами (Dorner, 1981; Pakarinen et al., 1994). Тем не менее, полученные нами и другими исследователями данные, согласно которым нейроны мозга начинают синтезировать и выделять ФАВ задолго до формирования межнейрональных связей и ГЭБ (Ugrumov et al., 1989а; Borisova et al., 1991; Ugrumov et al., 1994) позволяют пересмотреть традиционные представления о роли мозга в нейроэндокринной регуляции развития целостного организма (Rajerison et al., 1976; Dlouha et al., 1982; Fuse, 1996). В связи с этим нами была выдвинута гипотеза, согласно которой, мозг в онтогенезе до формирования межнейрональных синаптических связей и ГЭБ, функционирует как эндокринный орган, участвующий в регуляции развития и функционирования периферических органов и целостного организма.

На примере дофамина (ДА), нами ранее были получены косвенные доказательства в пользу выдвинутой гипотезы. Так было показано, что ДА содержится в периферической крови плодов и новорожденных крыс в концентрации, достаточной для оказания эндокринного влияния на периферические мишени и сам мозг (ауторегуляция) (Лаврентьева, 2006). После закрытия ГЭБ концентрация ДА в периферической крови резко падает, что исключает дальнейшее участие ДА мозгового происхождения в эндокринной регуляции (Лаврентьева, 2006). Однако, прямых доказательств того, что ДА, синтезируясь в нейронах мозга, поступает в общую систему циркуляции в период онтогенеза, предшествующий формированию ГЭБ, получено не было.

Для доказательства того, что развивающийся мозг играет роль эндокринного органа не достаточно показать, что секретируемые им ФАВ поступают в общую систему циркуляции, но необходимо также показать действие этих ФАВ на периферические орган-мишени. Учитывая, что рецепторы к ДА обнаруживаются в разных органах задолго до формирования ГЭБ (Felder et al., 1989; Melnikova et al., 1998) можно ожидать, что ДА, содержащийся в периферической крови, способен оказывать влияние на развитие и/или функциональную активность этих органов, что требует дальнейшей экспериментальной проверки.

Цель настоящей диссертационной работы - получить прямые доказательства эндокринной функции ДА-ергических нейронов целостного мозга в онтогенезе до формирования ГЭБ.

Для достижения данной цели были поставлены следующие задачи:

• Разработать экспериментальную модель фармакологического ингибирования синтеза ДА нейронами мозга в онтогенезе до формирования ГЭБ.

• Оценить вклад мозга в формирование физиологически активной концентрации ДА в плазме крови после ингибирования его синтеза в нейронах мозга до формирования ГЭБ.

• Получить доказательства того, что ДА мозгового происхождения, содержащийся в периферической крови до формирования ГЭБ, оказывает прямое эндокринное влияние на выделение пролактина из лактотрофов гипофиза в моделях in vivo и in vitro.

Научная новизна полученных результатов

Впервые разработана модель фармакологического ингибирования синтеза ДА избирательно в мозгу у крыс в раннем постнатальном периоде. Экспериментально подобрана доза ингибитор - а-метил-пара-тирозина (аМПТ) - ингибитора тирозингидроксилазы (ТГ) - ключевого фермента синтеза ДА, существенно снижающая синтез ДА в мозгу и не влияющая на его синтез в периферических ДА-синтезирующих источниках.

Впервые получено прямое доказательство того, что до формирования ГЭБ ДА поступает из мозга в общую систему циркуляции, обеспечивая поддержание его физиологически активной концентрации в крови.

Впервые на моделях in vivo и in vitro показано, что ДА мозгового происхождения, содержащийся в периферической крови у неонатальных крыс до формирования ГЭБ, способен оказывать специфическое влияние на функциональную активность лактотрофов, ингибируя выделение пролактина из передней доли гипофиза.

Научная и практическая значимость работы

Получены убедительные доказательства в пользу гипотезы о том, что мозг до формирования ГЭБ функционирует как эндокринный орган, выделяя ФАВ в общую систему циркуляции, которые в дальнейшем участвуют в регуляции функциональной

активности органов-мишеней, что принципиально меняет существующую концепцию формирования нейроэндокринной регуляции развития целостного организма.

Новый вариант концепции формирования нейроэндокринной регуляции в онтогенезе и ее роли в развитии целостного организма, а также основанные на этой концепции новые представления о патогенезе врожденных заболеваний, могут быть широко представлены в курсах и учебниках по физиологии и фундаментальной медицине для студентов университетов и медицинских институтов.

Работа вносит вклад не только в экспериментальную физиологию, но и в клиническую медицину, в частности перинаталогию. Изучение молекулярно-клеточных механизмов эндокринной регуляции развития периферических мишеней со стороны ФАВ мозгового происхождения позволит по-новому представить себе механизмы развития врожденных заболеваний репродуктивной и выделительной функций, нарушения функционирования сердечно-сосудистой системы и ряда других. Это, в свою очередь, приведет к изменению стратегии ранней диагностики и лечения врожденных заболеваний, существенно повысив их эффективность.

выводы

1. Впервые разработана экспериментальная модель ингибирования синтеза дофамина в мозгу у крыс в перинатальном периоде до формирования гемато-энцефалического барьера. Подобрана минимальная доза ингибитора, максимально снижающая уровень дофамина в мозгу и не влияющая на его синтез в периферических органах.

2. На разработанной модели показан существенный вклад развивающегося мозга в формирование физиологически активной концентрации дофамина в общей системе циркуляции до формирования гемато-энцефалического барьера, о чем свидетельствует значительное снижение концентрации дофамина в плазме крови после ингибирования его синтеза в мозгу.

3. На примере гипофиза как периферического органа-мишени для дофамина в экспериментах in vivo и in vitro получены доказательства того, что до формирования гемато-энцефалического барьера дофамин, поступающий из мозга в общую систему циркуляции, оказывает прямое эндокринное влияние на выделение пролактина из лактотрофов гипофиза.

4. Полученные данные подтверждают гипотезу о том, что мозг в онтогенезе до формирования гемато-энцефалического барьера играет роль эндокринного органа, секретируя в общую систему циркуляции физиологически активные вещества, и оказывает прямое эндокринное влияние на развитие и функционирование периферических органов-мишеней.
